Conceptual Study for Tissue-Regenerative Biodegradable Magnesium Implant Integrated with Nitric Oxide-Releasing Nanofi bers

        Jin‑Kyung Jeon,Hyunseon Seo,Jimin Park,Soo Ji Son,Yeong Rim Kim,Eun Shil Kim,Jong Woong Park,Woong‑Gyo Jung,Hojeong Jeon,Yu‑Chan Kim,Hyun‑Kwang Seok,Jae Ho Shin,Myoung‑Ryul Ok 대한금속·재료학회 2019 METALS AND MATERIALS International Vol.25 No.4

The excessive initial corrosion rate of Mg is a critical limitation in the clinical application of biodegradable Mg implantsbecause the device loses its fi xation strength before the fractured bone heals. This study suggests a new approach to overcomethis hurdle by accelerating tissue regeneration instead of delaying the implant biodegradation. As angiogenesis is anessential process in early bone regeneration, a Mg implant coated with electrospun nanofi bers containing nitric oxide (NO),which physiologically promotes angiogenesis, is designed. The integrated device enables adjustable amounts of NO to bestored on the NO donor-conjugated nanofi ber coating, stably delivered, and released to the fractured bone tissue near theimplanted sites. An in vitro corrosion test reveals no adverse eff ect of the released NO on the corrosion behavior of the Mgimplant. Simultaneously, the optimal concentration level of NO released from the implant signifi cantly enhances tube networkformation of human umbilical vein endothelial cells without any cytotoxicity problem. This indicates that angiogenesis canbe accelerated by combining NO-releasing nanofi bers with a Mg implant. With its proven feasibility, the proposed approachcould be a novel solution for the initial stability problem of biodegradable Mg implants, leading to successful bone fi xation.

This study aims to draw up a basic plan to construct an agricultural theme park in a farming city as a way of establishing its regional identity represented by “Three Whites.” Sangju, Kyongsangbuk-do which is the subject area of the study has been well-known for "Three Whites (rice, silkworm cocoon and dried persimmon)." The purpose of the study is to take a closer look at the folk life and culture of Sangju and develop tourism resources to promote regional development and competitiveness of the city. This study was conducted in several phases in consideration of the characteristics of plan and design, on the basis of the theoretical review of agricultural tourism. In the first phase, planning conditions were analyzed as per development objectives. In the second phase, strategies and concepts were established for the theme park, based on which development methods were decided by phase and facility. Then, a preliminary review was conducted on functions and facilities to introduce to the theme park. In the third phase, a comprehensive plan was drawn up, which includes basic plans for respective parts. In consideration of the characteristics of the study, construction was not looked at in detail. The total size of land subject to the study is approx. 97,960㎡, which was divided into four separate zones-Sambaek Cultural Experience Zone, Natural Observation & Experience Zone, Agricultural Life Experience Zone and other facilities zone--in order to connect facilities, functions and experience with the physical characteristics of the city, and separate space planning was conducted for each zone. The outcome of the study carries significance as an example of alternative tourism which considers regional characteristics, develops local resources and makes use of obsolete rural facilities, in what is called the eco-friendly development where nature and eco-system, landscape and culture, and recreation and leisure come together as one.

C-reactive protein accelerates DRP1-mediated mitochondrial fission by modulating ERK1/2-YAP signaling in cardiomyocytes

        ( Suyeon Jin ),( Chan Joo Lee ),( Gibbeum Lim ),( Sungha Park ),( Sang-hak Lee ),( Ji Hyung Chung ),( Jaewon Oh ),( Seok-min Kang ) 생화학분자생물학회 2023 BMB Reports Vol.56 No.12

C-reactive protein (CRP) is an inflammatory marker and risk factor for atherosclerosis and cardiovascular diseases. However, the mechanism through which CRP induces myocardial damage remains unclear. This study aimed to determine how CRP damages cardiomyocytes via the change of mitochondrial dynamics and whether survivin, an anti-apoptotic protein, exerts a cardioprotective effect in this process. We treated H9c2 cardiomyocytes with CRP and found increased intracellular ROS production and shortened mitochondrial length. CRP treatment phosphorylated ERK1/2 and promoted increased expression, phosphorylation, and translocation of DRP1, a mitochondrial fission-related protein, from the cytoplasm to the mitochondria. The expression of mitophagy proteins PINK1 and PARK2 was also increased by CRP. YAP, a transcriptional regulator of PINK1 and PARK2, was also increased by CRP. Knockdown of YAP prevented CRP-induced increases in DRP1, PINK1, and PARK2. Furthermore, CRP-induced changes in the expression of DRP1 and increases in YAP, PINK1, and PARK2 were inhibited by ERK1/2 inhibition, suggesting that ERK1/2 signaling is involved in CRP-induced mitochondrial fission. We treated H9c2 cardiomyocytes with a recombinant TAT-survivin protein before CRP treatment, which reduced CRP-induced ROS accumulation and reduced mitochondrial fission. CRP-induced activation of ERK1/2 and increases in the expression and activity of YAP and its downstream mitochondrial proteins were inhibited by TAT-survivin. This study shows that mitochondrial fission occurs during CRPinduced cardiomyocyte damage and that the ERK1/2-YAP axis is involved in this process, and identifies that survivin alters these mechanisms to prevent CRP-induced mitochondrial damage.
