Distributed optimization and statistical learning for large-scale penalized expectile regression

Large-scale data from various research felds are not only heterogeneous and sparse but also difcult to store on a single machine. Expectile regression is a popular alternative for modeling heterogeneous data. In this paper, we devise a distributed optimization approach to SCAD and adaptive LASSO penalized expectile regression, where the observations are randomly partitioned across multiple machines. We construct a penalized communication-efcient surrogate loss (CSL) function. Computationally, our method based on the CSL function requires only the master machine to solve a regular M-estimation problem, while other worker machines compute the gradient of the loss function on local data. Our method matches the estimation error bound of the centralized method during consecutive rounds of communication. Under some mild assumptions, we establish the oracle properties of the SCAD and adaptive LASSO penalized expectile regression. We then develop a modifed alternating direction method of multipliers (ADMM) algorithm for the implementation of the proposed estimator. A series of simulation studies are conducted to assess the fnite-sample performance of the proposed estimator. Applications to an HIV study demonstrate the practicability of the proposed method.
The Anti-detective Element in the New York Trilogy by Paul Auster

This paper mainly studies the anti-detective element in the New York Trilogy (1987) by Paul Auster, who is a famous American postmodern novelist. As Auster’s masterpiece, this work is a collection of three typically detective novels City of Glass (1985), Ghosts (1986) and The Locked Room (1986). It typically belongs to the anti-detective genre because Auster subverts the traditional detective conventions. The anti-detective element is one of the essential postmodern elements. The two anti-detective elements of language and identity are investigated in this paper. Paul Auster suggests that language is arbitrary and the signifer and the signified are separated from each other. The identity is uncertain and shifting. Auster intends to search truth and meaning by using these two tools. However, there is no absolute truth in the postmodern world. Auster himself claims that it is fiction, not reality. This work is studied a lot from the perspective of intertextuality, narration and so on. It is innovative to study the anti-detective element in Auster’s detective novels by analysing the element of language and identity, which presents the readers a new perspective to interpret Auster’s detective fiction.
Query Algebra for the Very Loosely Structured Data Model

In order to realize the idea of pay-as-you-go (PAYG) data management which dataspace emphasizes, the very loosely structured data model is usually used to describe the massive, heterogeneous and dynamic data in dataspace. However, the present study mainly concentrates on the applications of the data model, and the query theory research is less. The query algebra is a theoretical foundation for query and its optimization in a PAYG fashion, so that how to establish a complete algebra based on the characteristics of loosely structured data model is an important problem need to be solved. In this paper, a formal definition of very loosely structured data model is given, then the query model and query algebra based on the model are proposed, which support to not only the operations such as set operators, selection, projection and join, but also association query in dataspace.
Curcumin Alleviates Dystrophic Muscle Pathology in mdx Mice

Abnormal activation of nuclear factor kappa B (NF-κB) probably plays an important role in the pathogenesis of Duchenne’s muscular dystrophy (DMD). In this report, we evaluated the efficacy of curcumin, a potent NF-κB inhibitor, in mdx mice, a mouse model of DMD. We found that it improved sarcolemmic integrity and enhanced muscle strength after intraperitoneal (i.p.) injection. Histological analysis revealed that the structural defects of myofibrils were reduced, and biochemical analysis showed that creatine kinase (CK) activity was decreased. We also found that levels of tumor necrosis factor alpha (TNF-α), interleukin-1 beta (IL-1β) and inducible nitric oxide synthase (iNOS) in the mdx mice were decreased by curcumin administration. EMSA analysis showed that NF-κB activity was also inhibited. We thus conclude that curcumin is effective in the therapy of muscular dystrophy in mdx mice, and that the mechanism may involve inhibition of NF-κB activity. Since curcumin is a nontoxic compound derived from plants, we propose that it may be useful for DMD therapy.
Profiling Gene Expression During Gland Morphogenesis of a Glanded and a Glandless Upland Cotton

The pigment gland is an important character of the Gossypium plant. With the aim of identifying genes involved in pigment gland morphogenesis in cotton, gene expression during pigment gland morphogenesis in Chuan 2802, which is glanded both in seed and plant, and a glandless line N5 was profiled using Affymetrix Cotton microarray. The results showed that there were 564 differentially expressed genes greater than twofold during gland morphogenesis. About 60.2% of these genes shares similarity with known genes on GenBank and about 39.8% with no functional description in the database. These described genes may play roles in defense response, response to oxidative stress, peroxidase activity, and the other metabolic pathways. The KEGG Orthology-Based Annotation System indicated that these above twofold expressed genes involved seven biochemical pathways on KEGG. These findings suggest that a complicated regulation is associated with pigment gland morphogenesis and the associated defense response including gossypol biosynthesis in cotton.

Background: Ginsenoside Rg3 and gemcitabine have mutual enhancing antitumor effects. However, the underlying mechanisms are not clear. This study explored the influence of ginsenoside Rg3 on Zinc finger protein 91 homolog (ZFP91) expression in pancreatic adenocarcinoma (PAAD) and their regulatory mechanisms on gemcitabine sensitivity. Methods: RNA-seq and survival data from The Cancer Genome Atlas (TCGA)-PAAD and Genotype-Tissue Expression (GTEx) were used for in-silicon analysis. PANC-1, BxPC-3, and PANC-1 gemcitabine-resistant (PANC-1/GR) cells were used for in vitro analysis. PANC-1 derived tumor xenograft nude mice model was used to assess the influence of ginsenoside Rg3 and ZFP91 on tumor growth in vivo. Results: Ginsenoside Rg3 reduced ZFP91 expression in PAAD cells in a dose-dependent manner. ZFP91 upregulation was associated with significantly shorter survival of patients with PAAD. ZFP91 overexpression induced gemcitabine resistance, which was partly conquered by ginsenoside Rg3 treatment. ZFP91 depletion sensitized PANC-1/GR cells to gemcitabine treatment. ZFP91 interacted with Testis-Specific Y-Encoded-Like Protein 2 (TSPYL2), induced its poly-ubiquitination, and promoted proteasomal degradation. Ginsenoside Rg3 treatment weakened ZFP91-induced TSPYL2 poly-ubiquitination and degradation. Enforced TSPYL2 expression increased gemcitabine sensitivity of PAAD cells and partly reversed induced gemcitabine resistance in PANC-1/GR cells. Conclusion: Ginsenoside Rg3 can increase gemcitabine sensitivity of pancreatic adenocarcinoma at least via reducing ZFP91 mediated TSPYL2 destabilization.
Reading The New York Trilogy from thePerspective of Existentialism

Paul Auster (1947-) is a remarkable American postmodern novelist. The New York Trilogy is regarded as his masterpiece, which is a postmodern interpretation of detective fiction, exploring various philosophical themes. This paper targets at using the theory of existentialism to analyze this work. Existentialism focuses on humanity, individuality and freedom. It insists the idea that the existence of people is absurd and meaningless. However, based on existence, people can create their own self to attain a colorful life. Existentialist perspectives can be found in Paul Auster’s novels. The features of absurdity, identity crisis and free choice will be discussed in this paper. The absurdity shows in two angles. One is the image of the wanderer. The other one is the absurdity of the plot. The identity crisis is an important theme and all the characters suffer from the identity crisis. People have freedom to make their choices and become responsible for their free choices in their life. So it is significant to study The New York Trilogy from the perspective of existentialism. This study is valuable and can offer readers a new perspective to interpret Paul Auster’s other novels.
