This paper deals with cytogenetical and cytochemical studies of the carp (Cyprinus carpio), the funa (Carassius carassius) and their hybrids. When kept under a confined condition, the carp and the funa mate andcan produce hybrids. Reciprocal crosses are also possible with similar results. The hybrids grow regularly with no observed abnormalities in the course of their development. They rank intermediate between the parent species in several characters. The hybrid males are completely sterile, while a hybrid female laid eggs in backcrossing. The spermatogenetic activity in hybrid testes is greatly disturbed. The chromosomes as observed in spermatogonial devision of hybrids are 100 in number, being the total sum of the haploid numbers of the parents, 50 for the carp and 50 for the funa. Meiosis in the hybrid testes is highly disturbed being arrested at early stages of the meiotic prophase. Most of the germ-cells undergo pycnotic degeneration during the period from late leptotene, and no spermatozoa are produced. In some hybrid specimens, the gonads show mosaic structures composed of testicular and ovarian elements, anevidence suggesting that sterility is associated with intersexuality caused by genetic unbalance between the parent species. The DNA amount in spermatogonial nuclei of thehybrids is approximately the same as that of liver nuclei, showing the 2n value. The DNA amount in the pachytene nuclei of the hybrids is less than the 4n value, while the parent species have the reduced amount of DNA in their pachytene nuclei. A consideration was made that the reduced amount of DNA in the hybrid cells may cause the disturbance of cellular activity leading to the subsequent degeneration of cells. Epithelial-mesenchymal transition (EMT), a biological process through which epithelial cells transdifferentiate into mesenchymal cells, is involved in several pathological events, such as cancer progression and organ fibrosis. So far, we have found that methotrexate (MTX), an anticancer drug, induced EMT in the human A549 alveolar adenocarcinoma cell line. However, the relationship between EMT and the cytotoxicity induced by MTX remains unclear. In this study, we compared the processes of MTX-induced EMT and apoptosis in A549 cells. Q-VD-Oph, a caspase inhibitor, suppressed MTX-induced apoptosis, but not the increase in mRNA expression of α-smooth muscle actin (SMA), a representative EMT marker. In addition, SB431542, an EMT inhibitor, did not inhibit MTX-induced apoptosis. By using isolated clonal cells from wild-type A549 cells, the induction of EMT and apoptosis by MTX in each clone was analyzed, and no significant correlation was observed between the MTX-induced increase in α-SMA mRNA expression and the proportion of cells undergoing apoptosis. Furthermore, the increase in the mRNA expression of α-SMA was well correlated with cyclin-dependent kinase inhibitor 1A, a cell cycle arrest marker, but not with BCL-2 binding component 3 and Fas cell surface death receptor, which are both pro-apoptotic factors, indicating that the MTX-induced EMT may be related to cell cycle arrest, but not to apoptosis. These findings suggested that different mechanisms were involved in the MTX-induced EMT and apoptosis.
Cytological and Cytochemical Studies of the Hybrids in Fresh Water Teleosts

Genetical characters and abilities will be appeared by the special gene or gene group on the chromosomes. We have two different stand-points to this problem. (I) If these genes become homozygotic, the genetic presentation will be emphasized and fixed as a pure line. (II) The genetic presentation will be emphasized by the heterozygotic genes and, on the contrary, homozygotic genes only make no efficiency to emphasis in genetical characters. The problem which is by far the better of the two standpoints, should be evaluated and determined by the purposes of human beings.

Methotrexate (MTX) is widely used to treat various diseases. However, it induces adverse reactions like serious lung injury, including pulmonary fibrosis. Increasing evidence suggests that epithelial-mesenchymal transition (EMT) in injured alveolar epithelium contributes to the development of the pathophysiological state of the lung. We demonstrated that MTX induced EMT in cultured alveolar epithelial cell lines. Integrin-mediated signaling is considered a significant factor in recognizing the EMT process. However, the relationship between MTX-induced EMT and integrin family members is poorly understood. In the present study, we aimed to clarify the role of integrin in MTX-induced EMT in A549 and NCI-H1299 (H1299) cells by focusing on the integrin alpha 2 (ITGA2) subunit, selected based on our microarray analysis. MTX treatment for 72 h significantly increased the mRNA and cell surface expression of ITGA2 in both cell lines. However, this upregulation by MTX was suppressed by co-treatment with SB431542 and folic acid, which are inhibitors of MTX-induced EMT in A549 cells. The mRNA expression levels of EMT-related genes were more affected in the MTX-treated A549 cells with high ITGA2 expression than in those with low ITGA2 expression. Finally, E7820, an ITGA2 inhibitor, suppressed MTX-induced EMTrelated phenotypic changes, such as morphology and mRNA and protein expression of α-smooth muscle actin, a representative EMT marker. These findings suggest that ITGA2 may play a key role in MTX-induced EMT in alveolar epithelial cells.

The purpose of this paper is to clarify analytically the relation between the characteristics and the pricing of the products of individual manufacturers, so we can offer beneficial information for designing and developing products. Light trucks of 1 ton to 4 ton have been taken up as examples. We carried out analyses based on the information on the evaluations of the functional characteristics by surveying the specifications obtained from light truck catalogues.
Useful Control Equations for Practitioners on Dynamic Process Control

System identification and controller formulation are essential in dynamic process control. In system identification, data for system identification are obtained, and then they are analyzed so that the system model of the process is built, identified, and diagnosed. In controller formulation, the control equation is derived based on the result of the system identification. There has been much theoretical research on system identification and controller formulation. These theories are very useful when they are appropriately applied. To our regret, however, these theories are not always effectively applied in practice because the engineers and the operators who manage the process often do not have the necessary understanding of required time series analysis methods. On the other hand, because of widespread use of statistical packages, system identification such as estimating ARMA models can be done with little understanding of time series analysis methods. Therefore, it might be said that the most theoretically difficult part in practice is the controller formulation. In this paper, lists of control equations are proposed as a useful tool for practitioners to use. The tool supports bridging the gap between theory and practice in dynamic process control. Also, for some models, the generalized control equations are obtained.

Background Appropriate position of the nipple-areolar complex (NAC) is crucial following nipple-sparing mastectomy (NSM). The prevention of NAC malposition in two-stage implant-based breast reconstruction has not been well described, and the efficacy of the techniques has not been evaluated. This study aimed to evaluate the efficacy of our technique to prevent NAC malposition in patients who underwent implant-based breast reconstruction after NSM. Methods Patients who underwent two-stage implant-based breast reconstruction with NSM between January 2012 and December 2019 were included. We used a surgical technique to fix the NAC to the rigid base, assuming a pocket-like appearance, with pectoralismajormuscle and lateral adipofascial flap at the time of tissue expander (TE) insertion. Patients were classified into two groups based on the performance of the technique for the prevention of NAC malposition. Results In 35 patients who underwent implant-based breast reconstruction after NSM, the clavicle-to-nipple distance ratio was 96.05.0% in thosewho underwent NAC fixation and 86.111.5% in those who did not undergo NAC fixation. Conclusions Using our technique, NAC malposition could be prevented in two-stage implant-based breast reconstruction. NAC fixation during TE insertion was found to be extremely effective. This procedure successfully prevented NAC malposition without the formation of extra scars.
