      • A Mononuclear Non-Heme Manganese(IV)–Oxo Complex Binding Redox-Inactive Metal Ions

        <P>Redox-inactive metal ions play pivotal roles in regulating the reactivities of high-valent metal–oxo species in a variety of enzymatic and chemical reactions. A mononuclear non-heme Mn(IV)–oxo complex bearing a pentadentate N<SUB>5</SUB> ligand has been synthesized and used in the synthesis of a Mn(IV)–oxo complex binding scandium ions. The Mn(IV)–oxo complexes were characterized with various spectroscopic methods. The reactivities of the Mn(IV)–oxo complex are markedly influenced by binding of Sc<SUP>3+</SUP> ions in oxidation reactions, such as a ∼2200-fold increase in the rate of oxidation of thioanisole (i.e., oxygen atom transfer) but a ∼180-fold decrease in the rate of C–H bond activation of 1,4-cyclohexadiene (i.e., hydrogen atom transfer). The present results provide the first example of a non-heme Mn(IV)–oxo complex binding redox-inactive metal ions that shows a contrasting effect of the redox-inactive metal ions on the reactivities of metal–oxo species in the oxygen atom transfer and hydrogen atom transfer reactions.</P><P><B>Graphic Abstract</B> <IMG SRC='http://pubs.acs.org/appl/literatum/publisher/achs/journals/content/jacsat/2013/jacsat.2013.135.issue-17/ja312113p/production/images/medium/ja-2012-12113p_0007.gif'></P><P><A href='http://pubs.acs.org/doi/suppl/10.1021/ja312113p'>ACS Electronic Supporting Info</A></P>

        Individualized Treatment of Allergic Rhinitis According to Nasal Cytology

        Purpose: Nasal cytology is important in the diagnosis and treatment of nasal inflammatory diseases. Treatment of allergic rhinitis (AR) according to nasal cytology has not been fully studied. We plan to explore the individualized treatment of AR according to nasal cytology. Methods: Nasal cytology from 468 AR patients was examined for inflammatory cell quantity (grade 0-5) and the percentage of neutrophils and eosinophils. Results were subdivided into the following categories: AR(Eos), eosinophil ≥50% of the whole inflammatory cells; AR(Neu), neutrophils ≥90%; AR(Eos/Neu), 10%≤ eosinophil <50%; AR(Low), grade 0/1 inflammatory cell quantity. Nasal cytology-guided treatment was implemented: all AR(Eos) patients (n=22) and half of the AR(Neu) patients (AR[Neu1], n=22) were treated with mometasone furoate spray and oral loratadine. Another half of the AR(Neu) patients (AR[Neu2], n=22) were treated with oral clarithromycin. Visual analog scale (VAS), symptom scores, and nasal cytology were evaluated 2 weeks before and after treatment. Results: There were 224/468 (47.86%) AR(Eos), 67/468 (14.32%) AR(Neu), 112/468 (23.93%) AR(Eos/Neu), and 65/468 (13.89%) AR(Low) of the AR patients studied. There were no significant differences in clinical characteristics among these subgroups, except that the nasal blockage score was higher in AR(Eos) patients than in AR(Neu) patients (1.99 vs 1.50, P=0.02). Comparing AR(Eos) patients with AR(Neu1) patients 2 weeks after treatment, nasal symptoms and VAS were significantly lower in AR(Eos) patients, except for nasal blockage symptoms (P<0.05 of nasal itching and sneezing; P<0.01 for nasal secretion, total scores, and VAS). Comparing AR(Neu1) with AR(Neu2) patients, nasal symptoms, and VAS were significantly lower in AR(Neu2), except for nasal blockage and nasal itching symptoms (P<0.05 for nasal secretions, sneezing, total score, and VAS). Conclusions: Nasal cytology may have important value in subtyping AR and optimizing AR treatment. Treating neutrophils is very important in AR patients with locally predominant neutrophils.

        The soft reverse recovery characteristics of P-I-N power diodes by different lifetime killer were compared in this paper. It was concluded that the best local lifetime control at N" base was achieved through the optimization of penetrated depth into the wafer by 5 MeV proton irradiation, resulting in significant soft recovery performance in our study. The results of 5~12 MeV electron irradiation and platinum diffusion were also discussed here.<br/>

      • Mammalian Systems Biotechnology Reveals Global Cellular Adaptations in a Recombinant CHO Cell Line

        Effective development of host cells for therapeutic protein production is hampered by the poor characterization of cellular transfection. Here, we employed a multi-omics-based systems biotechnology approach to elucidate the genotypic and phenotypic differences between a wild-type and recombinant antibody-producing Chinese hamster ovary (CHO) cell line. At the genomic level, we observed extensive rearrangements in specific targeted loci linked to transgene integration sites. Transcriptional re-wiring of DNA damage repair and cellular metabolism in the antibody producer, via changes in gene copy numbers, was also detected. Subsequent integration of transcriptomic data with a genome-scale metabolic model showed a substantial increase in energy metabolism in the antibody producer. Metabolomics, lipidomics, and glycomics analyses revealed an elevation in long-chain lipid species, potentially associated with protein transport and secretion requirements, and a surprising stability of N-glycosylation profiles between both cell lines. Overall, the proposed knowledge-based systems biotechnology framework can further accelerate mammalian cell-line engineering in a targeted manner.

        Functional Characterization of Mammary Gland of Holstein Cows under Humid Tropical Summer Climates

        Physiological parameters were measured on six primiparous, non-pregnant Holstein cows prior to peak lactation over a 3-month summer season in southwestern Taiwan. The objectives were to characterize heat stress-induced change in functionality of mammary gland under natural climates of tropical summer and to establish physiological indices applicable to this environment in referring to this change. Environmental and physiological readings, milk and blood samples were taken at 15:00 h biweekly for totally five time points during the study. Climate readings showed that the afternoon humidex value reached the highest (53.5) around mid summer. Rectal temperature of cows taken simultaneously varied between $38.26^{\circ}C$ and $40.02^{\circ}C$ in parallel to humidex. Milk production declined drastically from 29.2 to 22.2 kg/d the first month entering summer but leveled up at end of the summer season suggesting effects exerted by heat stress rather than stages of lactation. Lactose content decreased linearly (p<0.05) with times in summer, from 4.69 to 4.38%. On the other hand, activity of N-acetylglucosaminidase (NAGase) in milk increased linearly to over two folds (p<0.05) during the same intervals. Elevations of fractional constituent of BSA in whey protein and serum cortisol level were also noticed in the course. Measurement of arteriovenous concentration (A-V) difference across the mammary gland demonstrated net uptake of glucose and net release of urea throughout the study period. The amount of urea released from mammary gland increased (p<0.05) progressively from 1.54 to 7.76 mg/dl during summer. It is concluded that gradual regression of mammary gland occurred along the humid tropical summer season. This regression is likely initiated through elevation of body temperature, which is irreversible above certain point. The increased release of urea from mammary gland during heat stress suggests its potential role as an early indicator of suboptimal mammary function.

      • Differential Expression of MMAC/PTEN in Glioblastoma Multiforme : Relationship to Localization and Prognosis

        MMAC/PTEN, a tumor suppressor gene located on chromosome 10q, has recently been shown to act as a phosphatidylinositol 3,4,5-triphosphate phosphatase and to modulate cell growth and apoptosis. Somatic mutations of MMAC/PTEN have been reported in a number of human cancers, especially in glioblastoma multiforme (GBM), although the number of identified mutations (10-35%) is significantly lower than the frequency of LOH affecting the MMAC/PTEN locus in the specimens (75-95%). To further investigate the possible alterations that may affect MMAC/PTEN, we examined the expression of the gene by reverse transcription-PCR in a series of gliomas. A significant difference (P<0.001) was observed between the expression of MMAC/PTEN in GBMs versus lower grades of gliomas, thus mimicking the difference in allelic deletion associated with the locus in these tumors. Furthermore, Kaplan-Meier survival plots, adjusted for age and tumor grade, showed a significantly better prognosis for patients whose tumors expressed high level of MMAC/PTEN. Additionally, immunostaining of GBMs revealed little or no MMAC/PTEN expression in about two-thirds of the tumors, whereas the other approximately one-third of tumors had significantly higher levels of expression. However, in about two-thirds of the gigh-expressing specimens, a heterogeneous pattern of expression was observed, indicating that certain cells within the tumor failed to express MMAC/PTEN. The combination of these results suggest that, in addition to molecular alterations affecting the gene, altered expression of MMAC/PTEN may play a significant role in the progression of GBM and patient outcome.         Cerebral Edema in Patients with Severe Hemispheric Syndrome: Incidence, Risk Factors, and Outcomes―Data from SITS-ISTR

        Background and Purpose Cerebral edema (CED) in ischemic stroke can worsen prognosis and about 70% of patients who develop severe CED die if treated conservatively. We aimed to describe incidence, risk factors and outcomes of CED in patients with extensive ischemia. Methods Oservational study based on Safe Implementation of Treatments in Stroke-International Stroke Treatment Registry (2003–2019). Severe hemispheric syndrome (SHS) at baseline and persistent SHS (pSHS) at 24 hours were defined as National Institutes of Health Stroke Score (NIHSS) >15. Outcomes were moderate/severe CED detected by neuroimaging, functional independence (modified Rankin Scale 0–2) and death at 90 days. Results Patients (n=8,560) presented with SHS and developed pSHS at 24 hours; 82.2% received intravenous thrombolysis (IVT), 10.5% IVT+thrombectomy, and 7.3% thrombectomy alone. Median age was 77 and NIHSS 21. Of 7,949 patients with CED data, 3,780 (47.6%) had any CED and 2,297 (28.9%) moderate/severe CED. In the multivariable analysis, age <50 years (relative risk [RR], 1.56), signs of acute infarct (RR, 1.29), hyperdense artery sign (RR, 1.39), blood glucose >128.5 mg/dL (RR, 1.21), and decreased level of consciousness (RR, 1.14) were associated with moderate/severe CED (for all P<0.05). Patients with moderate/severe CED had lower odds to achieve functional Independence (adjusted odds ratio [aOR], 0.35; 95% confidence interval [CI], 0.23 to 0.55) and higher odds of death at 90 days (aOR, 2.54; 95% CI, 2.14 to 3.02). Conclusion In patients with extensive ischemia, the most important predictors for moderate/ severe CED were age <50, high blood glucose, signs of acute infarct, hyperdense artery on baseline scans, and decreased level of consciousness. CED was associated with worse functional outcome and a higher risk of death at 3 months.

        Postoperative Low-Dose Tranexamic Acid After Major Spine Surgery: A Matched Cohort Analysis

        Objective: This was a retrospective, cohort study investigating the efficacy and safety of continuous low-dose postoperative tranexamic acid (PTXA) on drain output and transfusion requirements following adult spinal deformity surgery. Methods: One hundred forty-seven patients undergoing posterior instrumented thoracolumbar fusion of ≥3 vertebral levels at a single institution who received low-dose PTXA infusion (0.5–1 mg/kg/hr) for 24 hours were compared to 292 control patients who did not receive PTXA. The cohorts were propensity matched based on age, sex, American Society of Anesthesiologist physical status classification, body mass index, number of surgical levels, revision surgery, operative duration, and total intraoperative TXA dose (n=106 in each group). Primary outcome was 72-hour postoperative drain output. Secondary outcomes were number of allogeneic blood transfusions. Results: There was no significant difference in postoperative drain output in the PTXA group compared to control (660±420 mL vs. 710±490 mL, p=0.46). The PTXA group received significantly more crystalloid (6,100±3,100 mL vs. 4,600±2,400 mL, p<0.001) and red blood cell transfusions postoperatively (median [interquartile range]: 1 [0–2] units vs. 0 [0–1] units; incidence rate ratio [95% confidence interval], 1.6 [1.2–2.2]; p=0.001). Rates of adverse events were comparable between groups. Conclusion: Continuous low-dose PTXA infusion was not associated with reduced drain output after spinal deformity surgery. No difference in thromboembolic incidence was observed. A prospective dose escalation study is warranted to investigate the efficacy of higher dose PTXA.

        Porous Al2O3 catalyst carrier by 3D additive manufacturing for syngas reforming

        A disk-shaped ceramic catalyst made by 3D printing (3DP) is used to reform syngas. We select two Al2O3 powders (α- andhigh-surface-area θ-Al2O3) and polymeric binders for preparing the 3DP feedstock. Catalyst stacked Al2O3 disks are made bykneading the polymers with ceramic powders and extruding to produce filaments, which are melted and used for 3D additivemanufacturing (AM). The processing parameters (sequence of addition, viscosity of the feedstock, etc.), the phasetransformation of the θ-powder, and catalytic properties of the made carrier are examined. The made alumina disks weredensified to a relative density (RD) of 40% −65%. Two porous disks were coated with catalyst NiO and CeO2, and used toreform syngas made from waste paper. The best case reforms the CH4 content in the syngas from 25.7% down to 0.15% orlower. The reformed gaseous fuel is suitable for solid oxide fuel cells (SOFCs).
