        Isoindolin-1-ones from the stems of Nicotiana tabacum and their antiviral activities

        In previous studies, several isoindolin-1-oneanalogs that exhibited signifi cant anti-tobacco mosaic virus(anti-TMV) activities were isolated from Nicotiana tabacum . Since gene-editing mutants provide a new sample for thediscovery of active metabolites, we focused on the stems ofYN-18–23 (a mutant N. tabacum for gene editing with thealkaloid metabolic pathway cultivated by Yunnan TobaccoCompany), which led to the isolation of four new ( 1–4 )and four known ( 5–8 ) isoindolin-1-ones. To the best of ourknowledge, nicindole C ( 3 ) is the fi rst subclass of isoindolin-1-one bearing a pentacyclic ketone, while nicindole D ( 4 )is the fi rst example of isoindolin-1-one bearing a methylpyridin-2-(1 H )-one moiety. Compounds 1–4 were testedfor their anti-TMV activities, and the results revealed thatcompounds 1 , 3 , and 4 exhibited high anti-TMV activities atconcentrations of 20 μM with inhibition rates of 48.6, 42.8,and 71.5%, respectively. These rates are higher than the inhibitionrate of the positive control (33.2%). The mechanisticstudy of compound 4 , which had the highest anti-TMV activityrevealed that increased potentiation of defense-related enzyme activities and downregulation of expression of theNtHsp70 protein may induce resistance in tobacco againstthe viral pathogen TMV. Molecular docking studies alsorevealed that the isoindolin-1-one substructure is fundamentalfor anti-TMV activity. The methyl-pyridin-2-(1 H )-onemoiety in compound 4 and the 2-oxopropyl groups in compounds1 and 3 at the N -2 position may increase inhibitoryactivities. This study of the structure–activity relationshipis helpful for fi nding new anti-TMV activity inhibitors. Tostudy whether the isoindolin-1-ones have broader antiviralactivities, compounds 1–4 were also tested for their antirotavirusactivities. Compound 4 exhibited high anti-rotavirusactivity with a therapeutic index (TI) value of 20.7. This TI value is close to that of the positive control (20.2).

      • Research and Implementation of a Vertical Search Engine in the Financial Domain

        The exponential growth of the data on the web makes it difficult for web users to locate and obtain the interesting information. The general web search engines, such as Google, Baidu, reduce the degree of difficulty to some extent. The rise of vertical search engine provides a promising approach to address the big challenge. A Chinese vertical search engine is urgently needed to provide professional financial information retrieval service for related financial institutions and government agencies. Therefore, in this paper we investigate, design and implement a vertical search engine in the financial domain. Based onNutch plug-in mechanism, we provide and implement the details of a vertical search engine in the financial domain, especially in the finance domain focused crawler. Experimental results show that, the finance domain focused crawler based on Nutch plug-in mechanism has good performance and can satisfy practical requirements of medium search application, and the system runs well.

        A two-sided estimate in the Hsu-Robbins-Erds law of large numbers for i.i.d. random variables sequence

        Protein Kinase C Mediates the Corticosterone-induced Sensitization of Dorsal Root Ganglion Neurons Innervating the Rat Stomach

        Background/Aims Gastric hypersensitivity contributes to abdominal pain in patients with functional dyspepsia. Recent studies showed that hormones induced by stress are correlated with visceral hypersensitivity. However, the precise mechanisms underlying gastric hypersensitivity remain largely unknown. The aim of the present study was designed to investigate the roles of corticosterone (CORT) on excitability of dorsal root ganglion (DRG) neurons innervating the stomach. Methods DRG neurons innervating the stomach were labeled by DiI injection into the stomach wall. Patch clamp recordings were employed to examine neural excitability and voltage-gated sodium channel currents. Electromyograph technique was used to determine the responses of neck muscles to gastric distension. Results Incubation of acutely isolated DRG neurons with CORT significantly depolarized action potential threshold and enhanced the number of action potentials induced by current stimulation of the neuron. Under voltage-clamp mode, incubation of CORT enhanced voltage-gated sodium current density of the recorded neurons. Pre-incubation of GF109203X, an inhibitor of protein kinase C, blocked the CORT-induced hyperexcitability and potentiation of sodium currents. However, pre-incubation of H-89, an inhibitor of protein kinase A, did not alter the sodium current density. More importantly, intraperitoneal injection of CORT produced gastric hypersensitivity of healthy rats, which was blocked by pre-administration of GF109203X but not H-89. Conclusions Our data strongly suggest that CORT rapidly enhanced neuronal excitability and sodium channel functions, which is most likely mediated by protein kinase C but not protein kinase A signaling pathway in DRG neurons innervating the stomach, thus underlying the gastric hypersensitivity induced by CORT injection.         Zero Steady-state Error Tracking Control for Ball and Plate System Based on Principle of Internal Model

        In order to improve trajectory tracking accuracy and disturbance-rejection performance of the ball and plate system, a zero steady-state error tracking control strategy based on the principle of internal mode was proposed, and the backstepping controller with a low-pass filter was designed to suppress the system’s internal disturbance. Firstly, a linear model of the ball and plate system is established and the common instability model for reference input and disturbance signals is derived. Secondly, the zero steady-state error tracking control strategy composed of three controllers which have respective function is proposed. Thirdly, the solving procedure of each controller is given and the stability of the closed-loop system is guaranteed by Lyapunov stability theory. Finally, simulation results show the presented control scheme has better dynamic performance and disturbance rejection ability, compared with other methods including H∞control, sliding mode control, and LQR control.

        Quantified Analyses of Musk Deer Farming in China: A Tool for Sustainable Musk Production and Ex situ Conservation

        Adult male musk deer (Moschus spp.) secrete musk, a widely used ingredient in traditional Asian medicine and the international perfume industry. Musk deer are endangered due to historic over-utilization of musk and habitat loss. Musk deer farming, provides an important way of conserving musk deer and ensuring a sustainable musk supply. For over 50 years musk deer farming has been conducted in China with the endangered Alpine musk deer (Moschus sifanicus) the predominant farmed musk deer species. To date, few studies have examined the musk production of captive musk deer. This study analyzed musk-extraction data collected from 1997 to 2009 at Xinglongshan Musk Deer Farm, Gansu, China. The musk-extraction ratio (MER) of captive male musk deer was 90.30% (n = 732), while the annual average musk extraction (AME) per animal was $7.90{\pm}0.17\;g$ with the range from 0.00 g to 34.20 g (n = 732). The origin of the deer had an influence on AME and MER production, with male wild-captured (WC) individuals recording higher values (AME, $8.76{\pm}0.27\;g$, n = 272; MER, 93.75%, n = 272) than those of captive breeding (CB) males (AME: $7.39{\pm}0.22\;g$, n = 460; MER: 88.26%, n = 460). The origin of an individual's parents, however, did not influence AME and MER. Age also influenced musk production with the MER of 1.5-year-old males being 87.5% with an average musk production of $8.27{\pm}0.47\;g$ (n = 96). The peak period for musk production was from 1.5 to 8.5 years of age. The results of our study demonstrate musk deer farming could work as an effective measure to protect musk deer and provide sustainable musk resources, however, the musk production including MER and AME could be improved through optimizing the managing and breeding system in endangered musk deer farming.

      • Heat-Shock Protein 70 as a Tumor Antigen for in vitro Dendritic Cell Pulsing in Renal Cell Carcinoma Cases

        Immunological functions of heat shock proteins (HSPs) have long been recognized. In this study we aimed to efficiently purify HSP70 from renal cell carcinoma and test it as a tumor antigen for pulsing dendritic cells in vitro. HSP70 was purified from renal cell carcinoma specimens by serial column chromatography on Con A-sepharose, PD-10, ADP-agarose and DEAE-cellulose, and finally subjected to fast protein liquid chromatography (FPLC). Dendritic cells derived from the adherent fraction of peripheral blood mononuclear cells were cultured in the presence of IL-4 and GM-CSF and exposed to tumor HSP70. After 24 hours, dendritic cells were phenotypically characterized by flow cytometry. T cells obtained from the non-adherent fraction of peripheral blood mononuclear cells were then co-cultured with HSP70-pulsed dendritic cells and after 3 days T cell cytotoxicity towards primary cultured renal cell carcinoma cells was examined by Cell Counting Kit-8 assay. Dendritic cells pulsed in vitro with tumor-derived HSP70 expressed higher levels of CD83, CD80, CD86 and HLA-DR maturation markers than those pulsed with tumor cell lysate and comparable to that of dendritic cells pulsed with tumor cell lysate plus TNF-${\alpha}$. Concomitantly, cytotoxic T-lymphocytes induced by HSP70-pulsed dendritic cells presented the highest cytotoxic activity. There were no significant differences when using homologous or autologous HSP70 as the tumor antigen. HSP70 can be efficiently purified by chromatography and induces in vitro dendritic cell maturation in the absence of TNF-${\alpha}$. Conspecific HSP70 may effectively be used as a tumor antigen to pulse dendritic cells in vitro.

        Current Status of Etiology, Epidemiology, Clinical Manifestations and Imagings for COVID-19

        Coronavirus disease 2019 (COVID-19) is a transmissible respiratory disease that was initially reported in Wuhan, China in December 2019. With the alarming levels of COVID-19 spread worldwide, the World Health Organization characterized COVID-19 as a pandemic. Over the past several months, chest CT has played a vital role in early identification, disease severity assessment, and dynamic disease course monitoring of COVID-19. The published data has enriched our knowledge on the etiology, epidemiology, clinical manifestations, and pathologic findings of COVID-19. Additionally, as the imaging spectrum of the disease continues to be defined, extrapulmonary infections or other complications will require further attention. This review aims to provide an updated framework and essential knowledge with which radiologists can better understand COVID-19.
