The Presence of Borrelia valaisiana-Related Genospecies in Ticks and a Rodent in Taiwan

A field survey was conducted to investigate the presence of Borrelia burgdorferi sensu lato (s.l.) in six counties of Taiwan. Spirochetes were successfully isolated from one rodent ear sample out of 485 rodent ears and 53live, fed tick (Ixodes granulatus) samples. The spirochetes were confirmed to be B. burgdorferi s.l. by real-time PCR. In addition, 23 of 113 tick samples were tested positive for Borrelia DNA according to real-time PCR. The Borrelia isolate from the rodent and the 23 Borrelia DNA samples from the ticks were identified as B. valaisiana-related genospecies by phylogenetic analysis based on flagellin gene sequences. These findings suggest that the Borrelia valaisiana-related strains are maintained in a zoonotic cycle between tick vectors and reservoir hosts in Taiwan.
Predictors of Positive Bone Metastasis in Newly Diagnosed Prostate Cancer Patients

Background: The prevalence of prostate cancer (PCa) has been increasing in recent years. Treatment strategies are largely based on the results of bone scan screening. Therefore, our aim was to investigate predictors of positive bone metastasis in newly diagnosed PCa patients. Materials and Methods: After extensive review, 336 consecutive patients newly diagnosed with PCa between April 2010 and November 2013 at our institution were enlisted in the study. Patients were divided into two groups according to bone scan results. Univariate analyses (Chi-square test for discrete variables and independent t-test for continuous variables) were applied to determine the potentially significant risk factors associated with distant bone metastasis. Binary logistic regression analyses were used to further investigate the influence of these factors on bone metastasis. Results: The patient mean age was $71.9{\pm}8.6years$ (range: 48 to 94 years). The mean prostate specific antigen (PSA) level and biopsy Gleason score were $260.2{\pm}1107.8ng/mL$ and $7.4{\pm}1.5$, respectively. The body mass index (BMI) for the series was $24.5{\pm}3.4kg/m^2$. Sixty-four patients (19.0%) had a positive bone scan result. Patients with positive bone scan results had a significantly lower BMI ($23.3{\pm}3.5$ vs. $24.8{\pm}3.3$; p=0.003), a higher Gleason score ($8.5{\pm}1.1$ vs. $7.1{\pm}1.5$; p < 0.001), and a higher PSA level ($1071.3{\pm}2337.1$ vs. $69.4{\pm}235.5$; p < 0.001) than those without bone metastasis. Multivariate logistic regression analysis employing the above independent predictors demonstrated that a Gleason score of ${\geq}7$, clinical stage ${\geq}T3$, $BMI{\leq}22kg/m^2$, and an initial PSA level of ${\geq}20ng/mL$ were all independent predictors of bone metastasis. Conclusions: A bone scan might be necessary in newly diagnosed PCa patients with any of the following criteria: clinical stage T3 or higher, a Gleason score of 7 or higher, BMI equal to or less than 22, and a PSA level of 20 or higher.

This paper investigates the Minjiang River Bridge No. 3 in Dujiangyan based on actual bridge tests and theoretical methods ofanalysis; here, we analyze the bearing capacity and the strengthening of the bridge. The detailed reinforcement measures for the bridgepile included the following actions: taking pile-patching measures on both sides of the severely scoured pile, adding bearing platforms,and adopting concrete-filled steel tube on the part of the pile leakage that lies above the current scour line. The bearing capacityevaluation of the bridge was performed before the reinforcement under strong earthquake and current scour conditions. Afterreinforcement, a bearing capacity evaluation for the bridge was performed under strong earthquake and current scour conditions usingthe load test for bridge piles. The analysis indicates that under current scour conditions, the maximum bending moments at the tops ofpiles 15-1, 15-3 and 16-3 are -4,824.5 kN.m, 5,206.9 kN.m, and 5,229.9 kN.m, respectively. These values are greater than the yieldbending moment 4,787.7 kN.m obtained by the M-ϕ curve. Thus, the pile foundation will incur bending failure. The bearing capacity ofthe bridge after reinforcement can meet the design requirements and tends to be stable under the action of a strong earthquake. Throughstatic load tests using vehicles, the maximum strain effect coefficient was 0.62, which meets the specific requirements for calibrationcoefficients (i.e., ξ ≤ 1.0) . The maximum pier settlement was 0.01 mm, and the settlement tended to stabilize during the loading process. After unloading, the settlement nearly completely recovered, and the bridge foundation was stable; thus, the strengthening scheme isfeasible. These results provide an important reference for bridge reinforcement in earthquake-prone areas and river erosion areas.

Background/Aims: The aim of this study was to investigate whether genetic variations at positions -1082, -819, and 592 in the interleukin (IL)-10 promoter affect IL-10 production in children with irritable bowel syndrome (IBS). Methods: Ninety-four children with IBS and 102 children as healthy controls (HCs) were enrolled. Genomic DNA was extracted, and IL-10 -1082, -819, and -592 polymorphisms were detected by direct sequencing from all participants. Peripheral blood mononuclear cells (PBMCs) from 46 IBS children and 38 HCs were isolated and cultured with and without 5 ng/mL Escherichia coli lipopolysaccharide (LPS). IL-10 levels in the culture supernatants were measured by enzyme-linked immunosorbent assay. Results: There were no significant differences in the distribution of IL-10 -1082, -819, and -592 polymorphisms or in the allele and haplotype frequencies between IBS children and HCs. PBMCs from children with IBS had significantly lower IL-10 levels after LPS stimulation than PBMCs from HCs (p=0.011); however, LPS-induced IL-10 levels in PBMCs with different genotypes of -819 and 592 polymorphisms were not significantly different between IBS patients and HCs. Conclusions: Although significantly lower LPS-induced IL-10 production by PBMCs was noted, it is unlikely that IL-10 production was fully genetically determined in our IBS children. A novel organic/inorganic hybrid TVE-resin containing SiO₂ structure for phenol removal is successfully prepared by dispersion polymerization, in which the inorganic phase is composed of nano-SiO₂ modified by vinyltrimethoxy silane (VTMS) and the organic phase is constituted of ethylene glycol dimethacrylate (EGDMA). The chemical structure and physical properties of TVE-resin are characterized by FTIR, TGA, SEM and BET. The adsorption studies imply that the novel materials show optimum adsorption capacity at pH=6, dose 0.1 g, contact time 60 min, initial concentration 3000 mg/L and room temperature. The pseudo first-order model can be well fitted with the kinetic process. According to the adsorption isotherm analysis, the Freundlich model gives a better fit to the experimental data, indicating a multiple molecular adsorption for phenol removal. The calculated thermodynamic parameters indicate an exothermic and spontaneous process. A mixture desorption solvent containing methanol and deionized water (v:v, 1:1) can regenerate the TVE-resin completely and the resin displays good reusability after five regeneration recycles.

Although the current glucocorticoids (GCs) treatment for systemic lupus erythematosus (SLE) is effective to a certain extent, the difference in therapeutic effect between patients is still a widespread problem. Some patients can have repeated attacks that greatly diminish their quality of life. This study was conducted to investigate the relationship between HSP90AA2 polymorphisms and disease susceptibility, GCs efficacy and health-related quality of life (HRQoL) in Chinese SLE patients. A case–control study was performed in 470 SLE patients and 470 normal controls. Then, 444 patients in the case group were followed up for 12 weeks to observe efficacy of GCs and improvement of HRQoL. Two single nucleotide polymorphisms (SNPs) of HSP90AA2 were selected for genotyping: rs1826330 and rs6484340. HRQoL was assessed using the SF-36 questionnaire. The minor T allele of rs1826330 and the TT haplotype formed by rs1826330 and rs6484340 showed associations with decreased SLE risk (T allele: PBH = 0.022; TT haplotype: PBH = 0.033). A significant association between rs6484340 and improvement of HRQoL was revealed in the follow-up study. Five subscales of SF-36 were appeared to be influenced by rs6484340: total score of SF-36 (additive model: PBH = 0.026), physical function (additive model: PBH = 0.026), rolephysical (recessive model: PBH = 0.041), mental health (dominant model: PBH = 0.047), and physical component summary (additive model: PBH = 0.026). No statistical significance was found between HSP90AA2 gene polymorphisms and GCs efficacy. These results revealed a genetic association between HSP90AA2 and SLE. Remarkably, HSP90AA2 has an impact on the improvement of HRQoL in Chinese population with SLE.
