        Ultrastructure structure of antennal sensilla of carabid beetle Carabus elysii Thomson, 1856 (Coleoptera: Carabidae)

        The sensilla type, number and distribution in male and female adults of Carabus elysii Thomson, 1856 (Coleoptera: Carabidae) were studied using scanning electron microscopy. The results showed that there are seven categories and 12 types of sensilla in C. elysii adults: three types of S.Ch, three types of S.T, two types of S.B and one each of B.B, S.Co, S.Ca and S.Cam. There is no difference between male and female in the types of sensilla. Apart from the significant difference in the number and distribution of S.B.2, S.Co and S.Ca between male dorsal and ventral surfaces, there are no significant differences between male and female antennae. In general, the number of sensilla in females is larger than that in males. The results provide a basic reference for future ultrastructure, electrophysiological, and comparative behavioral studies of Carabus species.

        Molecular functions and therapeutic applications of exosomal noncoding RNAs in cancer

        Cancer is one of the most difficult diseases in human society. Therefore, it is urgent for us to understand its pathogenesis and improve the cure rate. Exosomes are nanoscale membrane vesicles formed by a variety of cells through endocytosis. As a new means of intercellular information exchange, exosomes have attracted much attention. Noncoding RNAs exist in various cell compartments and participate in a variety of cellular reactions; in particular, they can be detected in exosomes bound to lipoproteins and free circulating molecules. Increasing evidence has suggested the potential roles of exosomal noncoding RNAs in the progression of tumors. Herein, we present a comprehensive update on the biological functions of exosomal noncoding RNAs in the development of cancer. Specifically, we mainly focus on the effects of exosomal noncoding RNAs, including microRNAs, circular RNAs, long noncoding RNAs, small nuclear RNAs, and small nucleolar RNAs, on tumor growth, metastasis, angiogenesis, and chemoresistance. Moreover, we outline the current clinical implications concerning exosomal noncoding RNAs in cancer treatment.

      • Whole Brain Radiotherapy Plus Chemotherapy in the Treatment of Brain Metastases from Lung Cancer: A Meta-analysis of 19 Randomized Controlled Trails

        Objective: To evaluate the efficacy and safety of whole brain radiotherapy (WBRT) plus chemotherapy versus WBRT alone for treating brain metastases (BM) from lung cancer by performing a meta-analysis based on randomized controlled trials (RCTs). Methods: The PubMed, Embase, CENTRAL, ASCO, ESMO, CBM, CNKI, and VIP databases were searched for relevant RCTs performed between January 2000 and March 2012. After quality assessment and data extraction, the meta-analysis was performed using the RevMan 5.1 software, with funnel plot evaluation of publication bias. Results: 19 RCTs involving 1,343 patients were included. The meta-analyses demonstrated that compared to WBRT alone, WBRT plus chemotherapy was more effective with regard to the objective response rate (OR = 2.30, 95% CI = 1.79 - 2.98; P < 0.001); however, the incidences of gastrointestinal reactions (RR = 3.82, 95% CI = 2.33 - 6.28, P <0.001), bone marrow suppression (RR = 5.49, 95% CI = 3.65 - 8.25, P < 0.001), thrombocytopenia (RR = 5.83, 95% CI = 0.39 - 86.59; P = 0.20), leukopenia (RR = 3.13, 95% CI = 1.77 - 5.51; P < 0.001), and neutropenia (RR = 2.75, 95% CI = 1.61 - 4.68; P < 0.001) in patients treated with WBRT plus chemotherapy were higher than with WBRT alone. There was no obvious publication bias detected. Conclusion: WBRT plus chemotherapy can obviously improve total efficacy rate, butalso increases the incidence of adverse reactions compared to WBRT alone. From the limitations of this study, more large-scale, high-quality RCTs are suggested for further verification.

        Exploration of the Glycosyltransferase BmmGT1 from a Marine-Derived Bacillus Strain as a Potential Enzyme Tool for Compound Glycol-Diversification

        Glycosyltransferases (GTs) from microbes are an emerging and rich source for efficient glycol-transformation of natural/unnatural compounds. Here, we probed the catalytic capability and substrate promiscuity of BmmGT1 from marine-derived Bacillus methylotrophicus B-9987. The regioselectivity of BmmGT1 on macrolactin A (1) was explored by optimization of the reaction conditions, in which a series of O-glycosylated macrolactins (1a-1e) were generated, including two new di/tri-O-glucosyl analogs (1b and 1e). Furthermore, BmmGT1 was able to catalyze the glycosylation of the thiol (S-) or amine (N-) sites of phenolic compounds (2 and 3), leading to the generation of N- (2a) or S-glycosides (3a and 3b). The present study demonstrates that BmmGT1 could serve as a potential enzyme tool for O-, N-, or S-glycosyl structural diversification of compounds for drug discovery.

      • Suppression of Ku80 Correlates with Radiosensitivity and Telomere Shortening in the U2OS Telomerase-negative Osteosarcoma Cell Line

        Ku70/80 heterodimer is a central element in the nonhomologous end joining (NHEJ) DNA repair pathway, Ku80 playing a key role in regulating the multiple functions of Ku proteins. It has been found that the Ku80 protein located at telomeres is a major contributor to radiosensitivity in some telomerase positive human cancer cells. However, in ALT human osteosarcoma cells, the precise function in radiosensitivity and telomere maintenance is still unknown. The aim of this study was to investigate the effects of Ku80 depletion in the U2OS ALT cell line cell line. Suppression of Ku80 expression was performed using a vector-based shRNA and stable Ku80 knockdown in cells was verified by Western blotting. U2OS cells treated with shRNA-Ku80 showed lower radiobiological parameters (D0, Dq and SF2) in clonogenic assays. Furthermore, shRNA-Ku80 vector transfected cells displayed shortening of the telomere length and showed less expression of TRF2 protein. These results demonstrated that down-regulation of Ku80 can sensitize ALT cells U2OS to radiation, and this radiosensitization is related to telomere length shortening.

      • Microarray Analysis of Long Non-coding RNA Expression Profile Associated with 5-Fluorouracil-Based Chemoradiation Resistance in Colorectal Cancer Cells

        Background: Preoperative 5-fluorouracil (5-FU)-based chemoradiotherapy is a standard treatment for locally advanced colorectal cancer (CRC). However, CRC cells often develop chemoradiation resistance (CRR). Recent studies have shown that long non-coding RNA (lncRNA) plays critical roles in a myriad of biological processes and human diseases, as well as chemotherapy resistance. Since the roles of lncRNAs in 5-FU-based CRR in human CRC cells remain unknown, they were investigated in this study. Materials and Methods: A 5-FU-based concurrent CRR cell model was established using human CRC cell line HCT116. Microarray expression profiling of lncRNAs and mRNAs was undertaken in parental HCT116 and 5-FU-based CRR cell lines. Results: In total, 2,662 differentially expressed lncRNAs and 2,398 mRNAs were identified in 5-FU-based CRR HCT116 cells when compared with those in parental HCT116. Moreover, 6 lncRNAs and 6 mRNAs found to be differentially expressed were validated by quantitative real time PCR (qRT-PCR). Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis for the differentially expressed mRNAs indicated involvement of many, such as Jak-STAT, PI3K-Akt and NF-kappa B signaling pathways. To better understand the molecular basis of 5-FU-based CRR in CRC cells, correlated expression networks were constructed based on 8 intergenic lncRNAs and their nearby coding genes. Conclusions: Changes in lncRNA expression are involved in 5-FU-based CRR in CRC cells. These findings may provide novel insight for the prognosis and prediction of response to therapy in CRC patients.

        Complete mesocolic excision in right hemicolectomy

        Purpose: To demonstrate the feasibility, safety, and technical strategies of hand-assisted laparoscopic complete mesocolic excision (HAL-CME) and to compare oncological outcomes between HAL-CME and the open approach (O-CME) for right colon cancers. Methods: Patients who were scheduled to undergo a right hemicolectomy were divided into HAL-CME and O-CME groups. Measured outcomes included demographic variables, perioperative parameters, and follow-up data. Demographic variables included age, sex distribution, body mass index (BMI), American Society of Anesthesiologists (ASA) physical status classification, previous abdominal surgery, tumor localization, and potential comorbidities. Perioperative parameters included incision length, operative time, blood loss, conversion rate, postoperative pain score, postoperative first passage of flatus, duration of hospital stay, total cost, number of lymph nodes retrieved, TNM classification, and postoperative complications. Follow-up data included follow-up time, use of chemotherapy, local recurrence rate, distant metastasis rate, and short-term survival rate. Results: In total, 150 patients (HAL-CME, 78; O-CME, 72) were included. The groups were similar in age, sex distribution, BMI, ASA classification, history of previous abdominal surgeries, tumor localization, and potential comorbidities. Patients in the HAL-CME group had shorter incision lengths, longer operative times, less operative blood loss, lower pain scores, earlier first passage of flatus, shorter hospital stay, higher total costs, similar numbers of lymph nodes retrieved, similar TNM classifications, and a comparable incidence of postoperative complications. The 2 groups were also similar in local recurrence rate, distant metastasis rate, and short-term survival rate. Conclusion: The results demonstrate that the HAL-CME procedure is a safe, valid, and feasible surgical method for right hemicolon cancers.

      • Tea seed saponin(TSS) improve lipid metabolism and oxidative stress in high fat diet(HFD)-induced obese mice

        To investigate the effect of TSS on lipid metabolism and oxidative stress in HFD mice. The body weight and food intake were observed. The serum SOD, MDA, GSH, TC, TG, HDL-C, LDL-C were determined by kits. Western Blotting was used to detect the levels of PPAR-γ, AMPK, SIRT1 and PGC-1α. TSS treatment reduced the body weight, Lee"s index and fat organ indices of mice in the HFD group. Compared with the control(CON) group, the serum TC, TG and LDL-C in the HFD group were increased, Administrated with TSS can improve abnormal blood lipid levels. Compared with the CON group, the serum SOD level in HFD group was significantly reduced(P<0.05), and MDA level was increased; while the levels of serum MDA in the TSS group decreased and SOD level increased. The pathological sections showed that TSS could improve the degree of hepatic steatosis. TSS also increased the levels of PPAR-γ, AMPK, SIRT1 and PGC-1α, and the effect of the high-dose group was the most significant. TSS can reduce body weight and fat accumulation, improve lipid metabolism disorder and oxidative stress caused by HFD.

      • The Datum Design Study of High Precision GPS Height Monitoring Network

        There are still some key problems having to be solved in theory and technique applications when GPS is used to monitor the vertical deformation of ground with high precision. Utilizing the GPS technology to monitor the deformation of the land subsidence and ground fissure in Xi’an, this paper puts forward advice that the datum frame of GPS network has significant influence on the precision and accuracy of the vertical deformation results by some research. The co-authors make some theoretical study of the datum error and practice by establishing the datum error models, especially the influence of scale and azimuth datum errors on GPS monitoring network. Then the datum frame design methods and arithmetic of GPS monitoring network are presented and have taken a good effect.

        Enhanced photoactivity of N-doped TiO2 for Cr(VI) removal: Influencing factors and mechanism

        To further enhance the photoactivity of virgin TiO2, we prepared N-doped TiO2 (N-TiO2) by sol-gel method and used it to remove Cr(VI) ions from water under visible light. The catalysts were characterized by TEM, XRD, BET and UV-vis techniques. Better crystalline structure, larger specific surface area and decreased band-gap width were obtained after the insertion of N atoms into TiO2 lattice. Several influencing factors were also investigated. The removal efficiency of Cr(VI) increases with the decrease of initial Cr(VI) concentration or the increase of catalyst dosage. Addition of glucose or some inorganic ions (Mg2+, Al3+) can promote the photoreduction process by improving the quantum efficiency. The kinetics and mechanism were discussed as well.
