      • The Typical Application of Building Energy Consumption Monitoring and Energy Management : A Case Study

        Building energy consumption monitoring and management system plays a vital role in the advance of building energy conservation, because it offers abundant online real time data, which are mainly used to further analyze energy saving potential and put forward a feasibility methodology for implementation. In addition to its extreme importance in decreasing energy consumption and saving costs, the platform can also help to find the anomalies of the main equipment. In this paper, we first introduce the structure of the monitoring platform, and then put forward an error analysis principle to improve the precision of heat meters. A case study is given for a building in “hot summer and cold winter” area, including the installation of various measuring meters and data verification. Finally, the energy consumption data of the building from January to December in 2013 were analyzed.

        Effect of Electrostatic Spraying Conditions on Droplet Deposition in Different Depth Areas on the Back of Target

        In order to explore the optimal spraying effect of diff erent electrostatic spraying parameters on droplet deposition on the back of target, the application height, angle, charging voltage, and nozzle water pressure had been applied as spraying variables for the test designed to investigate on droplet density, deposition and coverage in diff erent depth areas of the target. The results of the study showed that the height and angle of the spatial parameters of the application had a significant effect on the application effect. The droplet deposition effect decreased by 94.71%, 89.09% and 94.17% at diff erent depths on the back of the target as the height increased from 20 to 50 cm. The deposition effects of droplets at diff erent depths on the back of the target at 60° tilt angle are 5.91%, 9.81% and 51.97% of them at 0° tilt angle respectively. The results of orthogonal experiment showed that the optimal parameters of diff erent depth areas on the back of different targets were inconsistent. The height and angle of application are the main influencing factors, when the target is warped up. The depth range of the target (5–7.5 cm) is greatly affected by the nozzle water pressure. In the actual spraying pesticides application, the appropriate operating parameters can be selected according to the size and attitude of the leaf surface to improve the spraying effect.

        Responses of GS-NS0 Myeloma Cells to Osmolality: Cell Growth, Intracellular Mass Metabolism, Energy Metabolism, and Antibody Production

        The influence of osmolality on growth, metabolism, and antibody production of mammalian cells has been widely reported in the past. However, more information about the responses of GS-NS0 Myeloma cells to osmolality, especially regarding the intracellular mass and energy metabolism, has not been available in detail. Fed-batch cultures started at different osmolalities in the range of 280~370 mOsm/kg were designed to investigate the effects. As the osmolality and cell status changed during the process, cell performance was evaluated in the comparable periods with similar growth rates, nutrition concentrations, and relatively consistent environments. Metabolic flux analysis indicated most of extra consumed glucose at higher osmolalities flowed into lactate formation pathway. The proportion of glucose flux flowed into glycolysis pathway remained approximately 90% and the need of glucose for biomass synthesis was constantly. Also, more than 88% of the glutamine was used in biomass synthesis and the absolute flux remained constant. The specific consumption rate of glutamine declined significantly when cells were cultured in hypo-osmolality (276 mOsm/kg) and a portion of glutamine was synthesized from glutamate. Furthermore, cells were in the state of high energy production at osmolality of 276 mOsm/kg. More glucose flowed into TCA circle with the high efficiency of energy production to meet the demand. Thus, the IVC, the specific antibody production rate, and maximal antibody concentration in fed-batch culture started at 280 mOsm/kg decreased by 35, 36, and 48% compared to those in the culture started at 330 mOsm/kg The influence of osmolality on growth, metabolism, and antibody production of mammalian cells has been widely reported in the past.         A One-Step System for Convenient and Flexible Assembly of Transcription Activator-Like Effector Nucleases (TALENs)

        Transcription activator-like effector nucleases (TALENs) are powerful tools for targeted genome editing in diverse cell types and organisms. However, the highly identical TALE repeat sequences make it challenging to assemble TALEs using conventional cloning approaches, and multiple repeats in one plasmid are easily catalyzed for homologous recombination in bacteria. Although the methods for TALE assembly are constantly improving, these methods are not convenient because of laborious assembly steps or large module libraries, limiting their broad utility. To overcome the barrier of multiple assembly steps, we report a one-step system for the con-venient and flexible assembly of a 180 TALE module library. This study is the first demonstration to ligate 9 mono-/dimer modules and one circular TALEN backbone vector in a one step process, generating 9.5 to 18.5 repeat sequences with an overall assembly rate higher than 50%. This system makes TALEN assembly much simpler than the conventional cloning of two DNA fragments because this strategy combines digestion and ligation into one step using circular vectors and different modules to avoid gel extraction. Therefore, this system provides a convenient tool for the application of TALEN-mediated genome editing in scientific studies and clinical trials.

        HARNACK INEQUALITY FOR A NONLINEAR PARABOLIC EQUATION UNDER GEOMETRIC FLOW

        In this paper, we obtain some gradient estimates for positive solutions to the following nonlinear parabolic equation $$\frac{{\partial}u}{{\partial}t}={\triangle}u-b(x,t)u^{\sigma}$$ under general geometric flow on complete noncompact manifolds, where 0 < ${\sigma}$ < 1 is a real constant and $b(x,t)$ is a function which is $C^2$ in the $x$-variable and $C^1$ in the$t$-variable. As an application, we get an interesting Harnack inequality.

        Ram semen preserved at 0℃ with soybean lecithin Tris-based extender substituted for egg yolk

        Objective: The present study evaluated the preservation of ram semen at 0℃ using soybean lecithin with a Tris-fructose extender. Methods: Semen was collected by artificial vagina ejaculation from six rams with proven fertility. High quality ejaculates were diluted by soybean lecithin (0.25%, 0.5%, 0.75%, 1.0%, 1.25%) using Tris-fructose extender and control (Tris-fructose egg yolk extender), respectively. The ejaculates were diluted to a concentration of 5×108 sperm/mL, followed by cooling to 0℃ in 90 min and maintaining the temperature for 12 days. The diluted semen samples were examined and recorded for sperm progressive motility, acrosome integrity at 0, 24, 72, 144, 216, 288 h, respectively. Two hundred and twenty-three ewes were inseminated for 216 h with optimal soybean lecithin concentrated semen or control via trans-cervical insemination. Results: The results showed that there were no differences in sperm progressive motility at 0, 24, 72, and 144 h (p>0.05). After 216 h, the sperm progressive motility in the control group and 0.5% concentration groups was significantly higher when compared to 0.25% concentration (p<0.05). The 0.5% concentration group demonstrated the highest survival rate and had no difference with the control group (p>0.05). At 216 h, the sperm progressive motility of all groups was still above 50%. The acrosome integrity of all groups was decreased with prolongation of storage time, but there was no difference at each time point (p>0.05). There was no significant difference in the lambing rate and pregnancy rate between the 0.5% concentration group and the control group (p>0.05). Conclusion: These results suggest that ram sperm is capable of fertilization after preservation at 0℃ with 0.5% of soybean lecithin in Tris-based extender substituted for egg yolk and produce normal offspring after insemination.

      • The Research on the Multi-Sensor Information Fusion Identifying of Alcohol based on Modified PCA and ANN

        Electronic nose (EN) is a equipment with ability of identification of simple or complex odors. Because of its low cost and accurate identification rate, the researches of it attract more attention and it develops quickly. Firstly, through Gas Sensor Array, we get large amount sample data and all data which is preprocessed. Secondly, modified Bp algorithm and RBF algorithm combining nearest neighbor - clustering algorithm and K-means clustering algorithm is proposed to realize the identification. Principal component analysis (PCA) method wipes off redundant sensor from the sensor array. Principle components and brief sensor signals are tested by above two algorithms. The test result indicated that the rapid and exact identification measure of ANN combining PCA is provided to the pattern identification with sensor information fusion.

      • The Application of Building Energy Consumption Index in Campus Energy Efficiency Management Platform

        Categorized, itemized energy consumption model of buildings is established using energy consumption data of campus energy efficiency management platform; existing categorized, itemized energy consumption evaluation indices of buildings are summarized; and a mathematical model of building energy consumption indices is established, providing the basis for the quantitative evaluation of building energy consumption. The scope of application of each index is summed up; and the methods and steps for evaluation of building energy consumption using categorized, itemized energy consumption indices of buildings are elaborated through case study. The results show that the categorized energy consumption indices of buildings can be used to analyze the whereabouts and proportion of various energy consumptions of buildings, grasp the energy structure of buildings, and help analyze the power consumption trends of buildings and the proportion of various sub items, providing the basis for the formulation of energy efficiency management plan.
