        Electroanalytical Approach for Determination of Tanshinone IIA Based on Electrochemically Reduced Graphene Oxide Modified Gold Nanoparticles-Incorporated Carbon Paste Electrode

        In this work, a novel electrochemical sensor was presented for Tanshinone IIA determination. The as-prepared electrode was prepared by electrodepositing a reduced graphene oxide film onto a carbon paste electrode modified with gold nanoparticles, which combined the advantages of both gold nanoparticles and electrochemically reduced graphene oxide. It was characterized with scanning electron microscopy and cyclic voltammetry. The electrochemical behavior of Tanshinone IIA at the modified electrode was investigated for the first time. Results revealed that the modified electrode can effectively increase response toward Tanshinone IIA by significantly enhancing the redox peak currents and decreasing the peak-to-peak separation. Under the optimized conditions, the modified electrode showed a linear voltammetric response to Tanshinone IIA at concentrations ranging from 5.0 x 10-8 mol L-1 to 1.0 x 10-5 mol L-1, with a detection limit of 1.0 x 10-8 mol L-1 (S/N = 3). The proposed method was successfully applied in determination of Tanshinone IIA in pharmaceutical samples with satisfactory results.

        New dammarane-type triterpenoids from the leaves of Panax notoginseng and their protein tyrosine phosphatase 1B inhibitory activity

        Background: Panax notoginseng has been used as a general tonic agent to invigorate human body for millennia in China and continued to be used until present. Methods: Some chromatographic methods were performed to isolate pure triterpenoids, and their structures were determined by nuclear magnetic resonance (NMR) experiments. Anti-diabetes activities of isolated compounds were evaluated through their inhibitory activity of protein tyrosine phosphatase 1B (PTP1B) enzyme. Results and Conclusion: Three new dammarane-type triterpenoids, notoginsenoside-LX (1), notoginsenoside-LY (2), and notoginsenoside-FZ (3) together with eighteen known compounds were isolated from the Panax notoginseng leaves. The structure-activity relationship of the compounds with dammaranetype triterpenoids and their PTP1B inhibitory activity were also reported. Results showed that compounds 2, 15, 20, and 21 can significantly inhibit the enzyme activity of PTP1B in a dose-dependent manner, with inhibitory concentration 50 ($IC_{50}$) values of $29.08{\mu}M$, $21.27{\mu}M$, $28.12{\mu}M$, and $26.59{\mu}M$, respectively. The results suggested that Panax notoginseng leaves might have potential as a new therapeutic agent for the treatment of diabetes.

        PM2.5 in poultry houses synergizes with Pseudomonas aeruginosa to aggravate lung inflammation in mice through the NF-κB pathway

        Background: High concentrations of particulate matter less than 2.5 µm in diameter (PM2.5) in poultry houses is an important cause of respiratory disease in animals and humans. Pseudomonas aeruginosa is an opportunistic pathogen that can induce severe respiratory disease in animals under stress or with abnormal immune functions. When excessively high concentrations of PM2.5 in poultry houses damage the respiratory system and impair host immunity, secondary infections with P. aeruginosa can occur and produce a more intense inflammatory response, resulting in more severe lung injury. Objectives: In this study, we focused on the synergistic induction of inflammatory injury in the respiratory system and the related molecular mechanisms induced by PM2.5 and P. aeruginosa in poultry houses. Methods: High-throughput 16S rDNA sequence analysis was used for characterizing the bacterial diversity and relative abundance of the PM2.5 samples, and the effects of PM2.5 and P. aeruginosa stimulation on inflammation were detected by in vitro and in vivo. Results: Sequencing results indicated that the PM2.5 in poultry houses contained a high abundance of potentially pathogenic genera, such as Pseudomonas (2.94%). The lung tissues of mice had more significant pathological damage when co-stimulated by PM2.5 and P. aeruginosa, and it can increase the expression levels of interleukin (IL)-6, IL-8, and tumor necrosis factor-α through nuclear factor (NF)-κB pathway in vivo and in vitro. Conclusions: The results confirmed that poultry house PM2.5 in combination with P. aeruginosa could aggravate the inflammatory response and cause more severe respiratory system injuries through a process closely related to the activation of the NF-κB pathway.

        System identification of the suspension tower of Runyang Bridge based on ambient vibration tests

        A series of field vibration tests are conducted on the Runyang Suspension Bridge during both the construction and operational stages. The purpose of this study is devoted to the analysis of the dynamic characteristics of the suspension tower. After the tower was erected, an array of accelerometers was deployed to study the evolution of its modal parameters during the construction process. Dynamic tests were first performed under the freestanding tower condition and then under the tower-cable condition after the superstructure was installed. Based on the identified modal parameters, the effect of the pile-soil-structure interaction on dynamic characteristics of the suspension tower is investigated. Moreover, the stiffness of the pile foundation is successfully identified using a probabilistic finite model updating method. Furthermore, challenges of identifying the dynamic properties of the tower from the coupled responses of the tower-cable system are discussed in detail. It\'s found that compared with the identified results from the freestanding tower, the longitudinal and torsional natural frequencies of the tower in the tower-cable system have changed significantly, while the lateral mode frequencies change slightly. The identified modal results from measurements by the structural health monitoring system further confirmed that the vibrations of the bridge subsystems (i.e., the tower, the suspended deck and the main cable) are strongly coupled with one another.

        Numerical Simulation of Site Deformation Induced by Shield Tunneling in Typical Upper-Soft-Lower-Hard Soil-Rock Composite Stratum Site of Changchun

        In China, with the rapid development of subway construction, the site deformation induced by subway construction is an interesting and hot issue in civil engineering. Focusing on the mechanism and characteristics of site deformation induced by parallel shield tunnel construction in typical soil-rock composite strata of Changchun region, and relying on the subway tunnel between Northeast Normal University Station and Gongnong Square Station, which belongs to the Changchun Metro Line 1, the tunnel construction process is simulated using FLAC3D software. In this paper, the numerical modeling and parameters for shield tunneling are described in detail, and the mechanism and characteristics of site deformation induced by shield tunneling are discussed systematically. Furthermore, the influence of construction parameters on site deformation, including construction sequence, tunnel spacing and earth chamber pressure, are studied. The results can provide reference for similar projects in the future.

        Experimental study of beam-column connections with web opening in a low-rise steel frame

        Steel frame structures have been widely used in multi-storey and high-rise buildings and the connections in these structures are critical. In the Northridge and Kobe Earthquake, beam-column connections suffered damage due to brittle fracture. According to seismic design codes, ductility of the beam to column connection is also necessary. A study on the behavior of a beam to column connection with the aim of improving ductility as well as preventing brittle failure was carried out. In order to control the position of a plastic hinge on the beam, a connection with a hole in the beam web was developed. Five specimens with different parameters under cyclic load were assessed. The results are presented in terms of the stress distribution of the beam, hysteretic behavior, and ultimate capacity. Furthermore, the finite element method was also used to analyze the model, and the results were compared with those obtained from the experiment. It is shown from the analysis and experimental results that this type of connection is effective in terms of improving ductility for a beam to column connection in low-rise buildings.

        Preparation and Investigation of Ulex Europaeus Agglutinin IConjugated Liposomes as Potential Oral Vaccine Carriers

        We prepared and optimized Ulex europaeus agglutinin I (UEAI)-modified Bovine serum albumin (BSA)-encapsulating liposomes (UEAI-LIP) as oral vaccine carriers and examined the feasibility of inducing systemic and mucosal immune responses by oral administration of UEAILIP. The prepared systems were characterized in vitro for their average size, zeta potential,encapsulation efficiency (EE%) and conjugation efficiency (CE%). In vitro release studies indicated that the presence of UEAI around the optimized liposomes was able to prevent a burst release of loaded BSA and provide sustained release of the encapsulated protein. In vivo immune-stimulating results in KM mice showed that BSA given intramuscularly generated systemic response only but both systemic and mucosal immune responses could be induced simultaneously in the groups in which BSA-loaded liposomes (LIP) and UEAI-LIP were administered intragastrically. Furthermore, the modification of UEAI on the surface of liposomes could further enhance the IgA and IgG levels obviously. In conclusion, this study demonstrated the high potential of lectin-modified liposomes containing the antigen as carriers for oral vaccine.
