        Evaluation of ginsenoside bioconversion of lactic acid bacteria isolated from kimchi

        Background: Panax ginseng is a physiologically active plant widely used in traditional medicine that is characterized by the presence of ginsenosides. Rb1, a major ginsenoside, is used as the starting material for producing ginsenoside derivatives with enhanced pharmaceutical potentials through chemical, enzymatic, or microbial transformation. Methods: To investigate the bioconversion of ginsenoside Rb1, we prepared kimchi originated bacterial strains Leuconostoc mensenteroides WiKim19, Pediococcus pentosaceus WiKim20, Lactobacillus brevis WiKim47, Leuconostoc lactis WiKim48, and Lactobacillus sakei WiKim49 and analyzed bioconversion products using LC-MS/MS mass spectrometer. Results: L. mesenteroides WiKim19 and Pediococcus pentosaceus WiKim20 converted ginsenoside Rb1 into the ginsenoside Rg3 approximately five times more than Lactobacillus brevis WiKim47, Leuconostoc lactis WiKim48, and Lactobacillus sakei WiKim49. L mesenteroides WIKim19 showed positive correlation with b-glucosidase activity and higher transformation ability of ginsenoside Rb1 into Rg3 than the other strains whereas, P. pentosaceus WiKim20 showed an elevated production of Rb3 even with lack of b-glucosidase activity but have the highest acidity among the five lactic acid bacteria (LAB). Conclusion: Ginsenoside Rg5 concentration of five LABs have ranged from w2.6 mg/mL to 6.5 mg/mL and increased in accordance with the incubation periods. Our results indicate that the enzymatic activity along with acidic condition contribute to the production of minor ginsenoside from lactic acid bacteria.

        수확시기 및 원료 조건에 따른 풋귤 식초의 항산화 활성

        항산화 활성이 우수한 풋귤 식초를 개발하기 위하여 풋귤의 수확 시기(7월-9월)에 따른 총 페놀 및 플라보노이드 함량 및 항산화 활성을 측정하고, 3가지 원료 조건(풋귤 분쇄액, 10% 건조 청피 첨가 풋귤 분쇄액, 10% 로스팅 청피 첨가 풋귤 분쇄액)으로 제조된 풋귤 식초의 발효 특성, 항산화 성분 및 항산화 활성을 비교하였다. 7월에 수확된 풋귤 착즙액은 1.02±0.02 μg GAE/mL의 가장 높은 총 페놀 함량이 확인되었으며, 플라보노이드 함량은 수확시기에 따른 유의적인 차이가 존재하지 않았다. 항산화 활성은 7월-8월에 수확된 풋귤 착즙액이 9월에 수확된 풋귤 착즙액보다 유의적으로 높은 DPPH 라디칼 소거능 및 환원력을 나타내었으며, 이러한 결과를 바탕으로 7월에 수확된 풋귤 착즙액을 식초 원료로 선정하였다. 원료 조건을 달리하여 제조된 식초의 최종 산도는 풋귤 식초가 6.0%, 건조 청피 첨가 풋귤 식초가 6.3%, 로스팅 청피 첨가 풋귤 식초가 6.7%로 모두 85%의 높은 발효 효율을 보였다. 건조 또는 로스팅 청피가 첨가된 풋귤 식초는 발효 13일차에 5% 산도에 도달하였으며, 풋귤 식초보다 발효기간이 단축되고 높은 산 생성능을 나타내었다. 원료조건에 따라 색도의 변화가 확인되었지만 유의적인 차이는 없었다. 유기산 함량은 건조 청피가 첨가된 풋귤 식초가 acetic acid를 비롯한 총 유기산 함량이 30,201 mg/L으로 가장 높았으며, 특히 풋귤 식초 보다 1.4배 높았다. 총 페놀 함량은 로스팅 청피 첨가 풋귤 식초에서 0.62±0.02 μg GAE/mL으로 풋귤 식초와 약 2배 이상의 차이를 보였으나, 총 플라보노이드 함량은 풋귤 식초가 3.00±0.15 μg CE/mL로 가장 높았다. DPPH 라디칼 소거능의 EC<SUB>50</SUB> 값은 건조 및 로스팅 청피 첨가 풋귤 식초가 각각 5.83±1.66 nL/mL, 4.71±0.07 nL/mL으로 풋귤 식초(9.16±0.87 nL/mL)보다 유의적으로 높은 활성을 나타냈으며, 환원력에서는 로스팅 청피 첨가 풋귤 식초가 7.47±0.28 nL/mL으로 활성이 가장 높았다. 이상의 결과로부터 7월에 생산된 풋귤 착즙액을 원료로 로스팅 청피를 첨가한 풋귤 식초는 초산 발효가 우수하고 높은 유기산 및 페놀 함량으로 우수한 항산화 활성을 나타내어 기존 풋귤 식초보다 항산화능이 강화된 기능성 식초로 활용될 수 있을 것으로 사료된다. This study aimed at comparing the antioxidant activity of premature mandarin (PM) vinegars by varying harvest periods (July, August, and September) and raw material conditions (PM fruit, PMF; PM fruit with 10% dried Citri Unshius Pericarpium Immaturus (CUPI), PMF-D, and PM fruit with 10% roasted CUPI, PMF-R). We found that the PM harvested in July exhibited the highest phenol content. Meanwhile, the July and August harvests showed stronger DPPH radical scavenging and reducing power. The phenol content of PMF-R vinegar was 0.62±0.02 μg GAE/mL, more than two times higher than that of PMF vinegar. The DPPH radical scavenging capacity and reducing power were the highest in the PMF-R vinegar, at 4.71±0.07 and 7.47±0.28 nL/mL, respectively. Therefore, it could be expected that PM vinegar prepared by adding roasted CUPI and harvested in July would exhibit high antioxidant activity and could be used as functional vinegar.

        바이오칩을 이용하여 유전자를 분석하는데 이때 바이오 칩 분석 시스템을 이용한다. 바이오 칩은 유전자와 실험의 두 축으로 이루어져 있으며 바이오 칩 분석 시스템을 사용하여 바이오 칩에서 자료를 추출하고 필요한 정보를 얻기 위해 데이터를 분석하는 시스템이다. 데이터를 분석하는 기법 중 클러스터링을 사용하는데 유사한 유전자들을 찾아 내어 정해놓은 클러스터로 정의한다. 같은 클러스터 안에 있는 유전자들은 서로 비슷한 성질을 가지고 있기 때문에 사용자들은 이 바이오 칩 으로부터 나온 정보를 효율적이게 사용할 수 있다. 더욱 효율적으로 사용하기 위해 본 논문에서는 방대한 양의 데이터의 최적화에 효율적인 생태계 모방 알고리즘 Particle Swarm Optimization을 이용하여 데이터들을 클러스터링을 하여 분류하는 시스템을 기술하고 있다.

      • The effects of conjugated linoleic acid (CLA) on metabolic syndrome patients: A systematic review and meta-analysis

        <P><B>Abstract</B></P> <P>The aim of the systematic review and meta-analysis was to evaluate the effects of conjugated linoleic acid (CLA) in metabolic syndrome (MS) patients. Randomized controlled trials (RCTs) conducted from inception to February 2015 were collected from seven databases. Among 1111 potentially relevant articles, nine RCTs (total 334 patients, 90% male) met inclusion criteria. A significant reduction in the CLA group in body weight and BMI (−0.51 kg [95% confidence interval (CI): −0.96, −0.07; p = 0.02] and −0.18 kg/m<SUP>2</SUP> [95% CI: −0.32, −0.04; p = 0.01], respectively) was found compared to control. No effect was seen on blood pressure, insulin resistance, or triacylglycerol, but a reduction in HDL in the CLA group (−0.06 mmol/L [95% CI: −0.09, −0.03; p = 0.0002]) was found to be statistically and clinically significant. In MS patients, CLA was effective on weight loss and BMI, but adversely reduced HDL. Thus, precautions should be taken with CLA use for MS patients.</P> <P><B>Highlights</B></P> <P> <UL> <LI> We aimed to evaluate the effects of CLA in metabolic syndrome patients. </LI> <LI> A significant reduction in the CLA group in body weight and BMI was found. </LI> <LI> A reduction in HDL in the CLA group was found to be statistically and clinically significant. </LI> <LI> Precautions should be taken with CLA use for metabolic syndrome patients with a low HDL. </LI> </UL> </P>

        Importance-Performance Analysis와 Borich 요구도 분석 방법을 활용한 임상간호실무 교육 요구도: 일개 상급종합병원 일반병동 간호사 대상

        Purpose: This study’s aim was to confirm the importance and performance of the clinical practice of nurses working in the general wards of a tertiary hospital and to analyze their educational needs. Methods: The study employed a descriptive research design, and a self-reported questionnaire was developed and used for nurses in a tertiary hospital. Data were collected from July 22 to July 29, 2022, analyzed by an independent t-test, paired t-test, and one-way ANOVA for the importance and performance of each clinical nursing practice according to general characteristics, and then a post hoc verification was performed by Scheffé’s test. An Importance-Performance Analysis and Borich needs assessment model were used to analyze clinical nursing education needs. Results: Clinical nursing practice performance showed a significant difference according to length of total clinical career, time working in current department, and preceptor experience. According to the results of the Importance-Performance Analysis, neurological evaluation, and nursing intervention, artificial respirator and high-flow oxygen inhalation nursing, chemotherapy, emergency nursing, and cardiopulmonary resuscitation were identified as belonging to the “concentrate here” quadrant. All these items ranked in the top 10 in the Borich needs assessment model. Conclusion: Based on these results, the current education system should be reviewed, and short and long term education strategies based on educational needs should be established to strengthen the competence of nurses.

      • Applicability of a colorimetric method for evaluation of lactic acid bacteria with probiotic properties

        <P><B>Abstract</B></P> <P>Rapid colorimetric methods using various indicator reagents have been developed to monitor bacterial viability. Here, we examined the applicability of a method based on the reduction of resazurin or water-soluble tetrazolium salt-8 (WST-8) to screen lactic acid bacteria (LAB) for growth, tolerance against bile acid and low pH. The resazurin reduction test proved unsuitable for screening LAB such as <I>Lactobacillus plantarum</I> and <I>Leuconostoc mesenteroides</I> since it reacted with acid present in the cultures. LAB growth could be indirectly quantified by measuring WST-8 reduction. This method proved more sensitive and quickly results than counting bacterial colony forming units and turbidity at 600 nm in the presence of bile and acid. Our results suggested that the WST-8-based method could be useful for the characterization of growth and tolerance of the lactic acid producing bacteria.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Resazurin cannot be used to measure lactic acid bacterial growth because of low pH. </LI> <LI> The WST-8 assay is a rapid, accurate, and sensitive approach for screening LAB. </LI> <LI> The WST-8 assay is useful for screening for LAB resistant to acid and bile. </LI> </UL> </P>

        DNA Chip을 이용한 실험은 그 결과에 대하여 대용량의 정보를 쏟아내고 있다. 이러한 데이터를 분석하는 다양한 기법 중, 미리 정해진 클래스에 데이터를 해당하는 클래스로 분류하는 기법인 분류화를 수행하여 의도한 목표를 위한 규칙을 찾아내고자 한다. 본 논문에서는 이를 위해 DNA Chip과 같은 방대한 양의 정보 분석에 대하여 적합한 생태계 모방 알고리즘인 PSO Algorithm을 사용하여 분류 규칙을 발견하여 이를 데이터에 적용, 분류하는 연구를 기술하고 있다.
