Collagen 2-induced Arthritis 생쥐에 대한 두중(杜仲),속단(續斷) 배합약물의 관절염 억제 효과

Objectives: The object of this study was to verify the inhibitory effect of a decoction of Eucommiae ulmides OLIVER (EU) and Dipsacus asperoides C. Y. Cheng et T.M.Ai (DA) on Collagen II-induced Arthritis Mice (CIA mice). Methods: DBA/1OlaHsd mice were immunized with bovine type II collagen. Boostnig same collagen 21 days later, arthritis was induced and then administrated orally the extract of EU+DA (200 or 50 mg/kg) once a day for 4 weeks and compared with that of methotrexate (MTX, 0.3 mg/kg) as a positive control. Results: Administration of EU+DA suppressed the inflammatory progression of CIA mice and the results were 1. Arthritis index of CIA mice was decreased 2. EU+DA decreased the production of TNF-α, IL-6, IL-1β in the serum of CIA mice. 3. EU+DA decreased the level of IgM. 4. EU+DAincreasaed CD3+, CD4+, CD4+/CD25 but decreased CD19+, CD3+/CD49b+(NKT), CD3-/CD49b+(NK), B220+/CD23+ in PBMC of CIA mice. 5. EU+DA decreased CD3+, CD4+, CD3+/CD69+ of paw joint in CIA mice. 6. EU+DA decreased subsynovial inflammation. Conclusions: This results demonstrated that extract of EU+DA suppressed the inflammatory progression of CIA mice and supported further studies are required to clarify a mechanism of therapeutic role.

본 연구는 배경지역 미세먼지의 질량 및 성분 연속측정데이터의 신뢰성 확보 및 물리ㆍ화학적 특성파악을 통한 생성과정 규명, 에피소드/기상 조건에 따른 미세먼지의 농도 및 화학적 특성 파악을 통하여 배경대기지역 입자상 물질의 구성 특성을 파악하였다. 그 결과 백령도의 2009년 1∼10월 먼지 질량농도는 PM₁?은 중량법으로 52.6 ㎍/㎥, 베타레이법 53.2 ㎍/㎥, PM2.5는 중량법으로 24.0 ㎍/㎥, 베타레이법으로 19.6 ㎍/㎥으로 나타났다. 그리고 중금속 성분의 실시간 연속측정 결과와 여지(filter) 분석 결과는 XRF 측정결과가 ICP-MS 분석결과보다 약간 높게 측정되었으나, 일별 월별 평균치의 변동경향은 유사하였다. 평상시의 백령도 PM2.5 성분은 다른 지역에 비해 SO₄²? 성분 비율이 높고, NO₃? 성분 비율은 낮은 편이었다. 입경별 질량분포 측정결과 중위경은 320 ㎚ 이었으며, 평상시는 입경 1.8 ㎛ 이하 미세입자 영역이 PM₁? 질량농도의 약 70% 이상을 차지하였다. 금속 성분은 1.8 ㎛ 이상의 조대영역에서 Fe, Mg, Ca 성분의 농도가 높았다. 이온 성분은 SO₄²?, NH₄? 성분은 주로 입경 1.8 ㎛ 이하의 초미세입자 영역, NO₃? 성분은 입경 1.8 ㎛ 이상의 조대입자 영역에서 많이 존재하였다. PM2.5 구성성분 특성은 백령도가 수도권 다른 지역에 비해 양이온 성분, OC 성분이 약간 많은 부분을 차지하는 것으로 나타났다. The concentration levels of PM in Baengnyeong-do during January~October 2009 show that PM₁? was 52.6 ㎍/㎥ by gravimetric method and 53.2 ㎍/㎥ by β-ray method, while PM2.5 was 24.0 ㎍/㎥ by gravimetric method and 19.6 ㎍/㎥ by β-ray method. The result of a quasi-real time monitoring instrument and lab filter analysis show that the continuos monitoring instrument XRF’s estimates were slightly higher than the ones from ICP-MS, but the trend of daily variations were similar. Fe, Pb, and Mg had a high correlation, while the correlation between As, Cr, and Se was relatively low. In the analysis of ion elements in PM2.5 concentration, SO₄²? was higher in Baengnyeong-do than in other regions, while NO₃? was lower. In high concentration cases, heavy metal concentration among PM10 elements and anion content(%) in PM2.5 increased rapidly. For Ion components in diameter in usual times, SO₄²? and NH4+ accounted for large amounts of all. SO₄²? and NH₄? components existed mostly as ultrafine particles below 1.8 ㎛ in diameter, while NO₃? was present mostly as corse particles above 1.8 ㎛ in diameter. OC concentration in PM2.5 ranged between 3.5∼8.4 ㎍/㎥, whereas the concentration of EC was between 0.7∼2.3 ㎍/㎥.

Objectives : The objective of the paper was to study cutting method of Gardeniae frutus, that tends to show symptom of separations in a packing, in accordance with the comparison experiment of quantification and antioxidant effect proceed with the intact Gardeniae fructus(Gf), seed of Gardeniae fructus (Gs) and the pericarp of Gardeniae fructus (Gp) separately. Methods : The Gf, Gs, and Gp were extracted using 80% MeOH, followed by quantizing geniposide contained in each group. A MTT assay was conducted and ROS generation and NO production were measured for comparing its antioxidant effect. Results : As a result of quantizing geniposide contained in the Gf, Gp, and Gs, respectively, the geniposide content was shown to be the highest in the Gs. MTT assay showed that no cytotoxicity was observed in the groups treated with Gf, Gp, and Gs, respectively, at a dose of 500 μg/ml. The ROS generation was shown to have more significantly decreased in the group pretreated with Gf 500 μg/ml than in the group treated with LPS. The NO level was shown to have more significantly decreased in the group pretreated with Gp 500 μg/ml than in the group treated with LPS. Conclusion : As the geniposide content and antioxidant effect of Gf varies according to its each part, it is recommended that Gf should be distributed as an intact form other than segregation in packing.
拱辰丹의 "麝香 代入 沈香 或 木香"에 관한 古典文獻 硏究

Objectives : The object of this paper is to seek and provide evidences for the possibilities of substitutional use of Aquilariae Resinatum Lignum and Aucklandiae Radix for Moschus as an ingredient of Gongjin-dan by clarifying the similarities and differences among these herbs in the classic literatures. Methods : We have found out the features of Moschus, Aquilariae Resinatum Lignum and Aucklandiae Radix by searching Qi, Flavor and Effect of each herb in 『Shennongbencaojing』, 『Zhengleibencao』, 『Bencaogangmu』, 『Dongeuibogam』, 『Bencaobeiyao』, 『Euijongsonik』 and 『Bangyakhappyeon』. And we have compared and analyzed the fomula of Gongjin-dan and other prescriptions in 『Beijiqianjinyaofang』, 『Shengjizonglu』, 『Shiyidexiaofang』, 『Dongeuibogam』, 『Euijongsonik』, 『Euibanghwaltu』 and 『Bangyakhappyeon』. Results : We could find out the similarities and differences in Qi, Flavor and Effect among Moschus, Aquilariae Resinatum Lignum and Aucklandiae Radix. And we could also find the examples of substitutional use of Aquilariae Resinatum Lignum as an ingredient of Gongjin-dan by comparing the formula of Gongjin-dan in the classic literatures as above. And in the other prescription, Aucklandiae Radix was also used instead of Moschus. Conclusions : All of three herbs have pungent in Flavor and warm in Qi. And these herbs also have similar effects in dispersing invading pathogenic factors and smoothening the flow of Qi. And according to 『Bencaobeiyao』, 『Euijongsonik』 and 『Bangyakhappyeon』, Aquilariae Resinatum Lignum and Aucklandiae Radix direct and regulate the Qi in common. The substitutional use of Aquilariae Resinatum Lignum and Aucklandiae Radix instead of Moschus in Gonjin-dan seems to be related to 'the ascent of Water and descent of Fire' effect of Gongjin-dan.

Solanum Iyratum Thunb (Solanaceae) has multiple applications in korean traditional medicine because of its cytotoxic, immunological and anti-inflammatory activities. However, no study on the anti-arthritic activity of Solanum Iyratum Thunb has been reported in vivo. Rheumatoid arthritis (RA) is a systemic autoimmune disease with chronic inflammation characterized by hyperplasia of synovial cells in affected joints, which ultimately leads to the destruction of cartilage and bone. Cytokine production were assessed during CIA(collagen-induced arthritis) model mice in lymph node (LN), in knee joint and spleen, using ELISA. DBA/1j mice were immunized with bovine type II collagen. After a second collagen immunization, mice were treated with Solanum Iyratum Thunb (SLT) orally at 400, 200 mg/kg once a day for 4 weeks. The severity of arthritis within the knee joints was evaluated by histological assessment of cartilage destruction and pannus formation. SLT significantly suppressed the progression of CIA and inhibited the production of TNF-alpha and IFN-gamma in serum and spleen cell culture supernatant. The erosion of cartilage was dramatically reduced in mouse knees after treatment with SLT. In conclusion, our results demonstrates that SLT significantly suppressed the progression of CIA. This action was characterized by suppression of arthritis index, cartilage erosion and synovial cell infiltration and the decreased production of $TNF-{\alpha}$, $IFN-{\gamma}$, CD4+, CD19+, CD3+/CD69+ cells (in lymph node), CD11b+/Gr-1 + (in knee joint).

Since the amendment of the law on the private sector investment in social infrastructure in January of 2005, the government has been actively promoting Build-Transfer-Lease (BTL) projects. Notably, most new educational facilities have been built as BTL projects. For these facilities, the unit cost per unit area has been applied to predict construction costs. However, since construction costs are mostly managed after the detailed design phase, the costs can be estimated incorrectly. For this reason, cost management is needed in the planning phase, with a sound approximate estimate to prevent the wasteful use of funds. To address this shortcoming, this study aims to develop a quantity prediction model for education facilities using regression analysis in the planning phase. The developed model is focused on the required quantities of reinforced concrete and bricks. In order to achieve the objective, the data of 44 educational facility projects collected from Gyeonggi-do was used in the regression model. This study can be utilized by major stakeholders to accurately predict construction costs by estimating the appropriate quantities of reinforced concrete and bricks in the planning design phase.
