      • 有機鹽素系 化合物(γ-BHC)을 投與한 마우스의 脂肪代射에 관한 硏究

        正常群, γ-BHC低用量 投與群, γ-BHC中等用量 投與群, 및 γ-BHC高用量 投與群에 對하여 Liver lipid classes의 定性分析, liver total lipid量 및 liver total cholesterol量을 測定하여 γ-BHC의 肝臟脂肪代謝에 미치는 영향을 檢討하였다. 測定方法은 Thin-layer chromatography와 吸光度測定法을 使用하였다. 그結果를 綜合하면 다음과 같다. 1) 正常群과 γ-BHC投與各群들의 liver lipid chasses는 同一하였다. 2) Liyer total lipid量은 正常群에 比해 γ-BHC 投與各群들에서 增加하였다. 3) Liyer total chalesterol量은 正常群에 比해 γ-BHC 投與各群들에서 增加하였다. The qualitative analysis of mouse liver lipid chasses was carried out by TLC. The total mouse liver lipid was determined by spectrophotometry. The total mouse liver cholesterol was determined by spectrophotometry. The above experimentes were carried out at non, low, medium and high concentration γ-BHC administrated mouses. The results were as follows: 1) The lipid classes of mouse liver were the same in normal and γ-BHC administrated mouses. 2) The total lipid of mouse liver was increased in γ-BHC administrated mouses. 3) The total cholesterol of mouse liver was increased in γ-BHC administrated mouses.

        본 연구는 항공사 객실승무원의 근무환경 개선과 근로생활의 질 향상을 위해 외국항공사의 단체협약을 통하여 그 방안을 모색하고자 하였다. 첫째 근무환경개선을 위해 단체협약에 각 직종의 근무특성을 명시한 근무협정서가 명시되어야 한다. 둘째 객실승무원의 합리적인 근무할당 및 승무원운영방안이 모색되어 합리적인 근무시간과 휴식시간의 분배가 있어야 한다. 셋째 동기부여 및 책임자 권리를 위한 적절한 보상과 근무시간에 대한 규정이 필요하다. 넷째 안전에 관한 보호규정이 고려되어야 한다. 다섯째 의사결정과정에의 참여와 협력증진에 관한 제도 보장 및 개선으로 근무할당 및 근로조건의 개선, 개발에 객실승무원의 참여의 길을 열어 노사 공동의 이익을 위한 방안이 필요하다. This study is for development of working condition and circumstances of flight attendants of Airline after review their agreement of union and through it. First, to protect and avoid the consumable confliction in between of company and union, agreement of union of flight attendant must be included with specified job description and clear statement of clarify. Second, Systematically organized timetable for working & rest schedule for flight attendant to arrange and development their labor efficient. Third, to inspire responsibility and motivation for their job and for their rights of job, prepare the adequate regulation for working hour management and compensation. Fourth, to increase their mental stability, set the security protection systems, regulation and its enforcement by inaction. Fifth, Necessary of channels and system for to negotiation of working condition and circumstances reflect and its improvements and development for bath benefit of company and labors.

      • 전립선 기질세포의 증식과 COX-2 발현에 대한 프로게스테론의 영향

        전립선비대증은 노인 남성에서 흔히 유발되는 질환이며, 노화가 진행될 수록 빈도가 높아지는 특징을 가진다. 이 질환의 원인은 전립선기질세표의 과도한 증식으로 유발된다고 알려져 있지만 그 자세한 기전에 대해서는 잘 알려져 있지 않다. 전립선비대증에서 progesterone 수용체 양성 세포가 다른 전립선 종양에 비해서 많고, progesterone은 testosterone에서 DHT로 전환되는 것을 감소시키는 역할을 가진다고 알려졋다. 또한 남성 전립선 평활근의 과증식에 의한 질환이므로 평활근 세포의 증식과 관련성이 있다고 보고된 COX-2의 전립선비대증에 대한 영향에 대한 연구가 필요하다. 전립선 기질세포에 progesterone을 3일간 투여하여 배양한 경우 기질세포 증식은 차이가 없었다. Progesterone을 단독 또는 DHT와 같이 투여한 기질세포에서 남성호르몬 수용체 mRNA 발현은 비처리군과 비교하여 유의한 차이가 없었다. 또한 progesterone과 DHT 동시 투여에 의한 COX-2 mRNA 발현에도 차이가 없었다. 그러나 progesterone에 의한 남성 호르몬 수용체와 COX-2 단백 발현에서는 대조군과 비교하여 유의하게 감소시켰다. 이상의 결과는 progesterone은 남성호르몬 수용체에 대해 전사 후 반응 (post-transcriptional response)에 효과를 나타내어 남성호르몬 수용체 발현을 감소시키는 작용은 가지며, COX-2 발현 억제효과를 나타내므로 전립선비대증의 치료에 이용될 수 있을 것으로 사료된다. Background: Benign prostatic hyperplasia (BPH) is the most common benign tumor in older men; the etiology of this disease remains poorly understood. Testosterone and dihydrotestosterone (DHT) both act as androgen via a single androgen receptor. Testosterone is converted to DHT by 5α-reductase in prostatic stromal cells. Progesterone has been reported to inhibit DHT conversion; howevwe, its effect on prostatic stromal cells remains to be elucidated. Materials and Methods: In this experiment, we investigated the effect of progesterone on androgen receptor expression induced by DHT. We also tested the effect of progesterone on cyclooxygenase-2 (COX-2) expression, as well as prostate stromal cell proliferation using the cell count kit-8. Results: Progesterone did not cause an increase of prostate stromal cell proliferation. The mRNA expression of the androgen receptor and COX-2 were not changed by progesterone; the expressions of androgen receptor and COX-2 proteins were decreased by progesterone in prostate stromal cells. Conclusion: These results suggest that in prostate stromal cells, progesterone decreases androgen receptor protein expression, which results in decrement of COX-2 protein expression. This effect might be mediated by post-transcriptional regulation.

        벼줄무늬잎마름바이러스의 대 발생과 발생 요인

        The genetic diagnosis methods by RT-PCR and Virion capture (VC)/RT-PCR against Rice stripe virus (RSV) were developed. Three diagnosis methods of seedling test, ELISA and RT-PCR were compared in virus detection sensitivity (VDS) for RSV. The VDS of ELISA for RSV viruliferous small brown plant hopper (SBPH) was higher with 40.5% than that of seedling test. The VDS of RT-PCR was higher with 21% than that of ELISA. The VDS of ELISA and VC/RT-PCR was same with 9.2% in average on the SBPH collected from fields at the areas of Gimpo, Pyungtaeg and Sihueng, Gyeonggi province in 2009. The specific primers of RSV for SBPH and rice plant were developed for the diagnosis by Real time PCR. The RQ value of Real time PCR for the viruliferous and non viruliferous SBPH was 1 for 50 heads of non viruliferous SBPH, 96.5 for 50 heads of viruliferous SBPH, 23.1 for 10 heads of viruliferous SBPH + 40 heads of non viruliferous SBPH, and 75.6 for 30 heads of viruliferous SBPH + 20 heads of non viruliferous SBPH. The RQ value was increased positively by the ratio of viruliferous SBPH. Full sequences of 4 genomes of RSV RNA1, RNA2, RNA3 and RNA4 were analysed for the 13 RSV isolates from rice plants collected from different areas. Genetic relationships among the RSV isolates of Korea, Japan and China were classified as China + Korea, and China + Korea + Japan by phylogenetic analysis for RSV RNA1 and RNA2. In case of RNA3 involved in pathogenicity, genetic relationship of RSV among the three countries was grouped into 3 as China, China + Korea, and Korea + Japan. According to the genetic relationships in RSV RNA4, RSV isolates were grouped into 4 as China, Korea, China + Korea + Japan, and Korea + Japan. Viruliferous insect rate (VIR) of RSV in average increased in each year from 2008 to 2010, and the rates were 4.3%, 6.1%, and 7.2%, respectively, at the 28 major rice production areas in 7 provinces including Gyeonggido. The highest VIR in each year was 11.3% of Gyeonggido in 2008, 20.1% of Jellanamdo in 2009 and 14.2% of Chungcheongbukdo in 2이 0. The highest VIR depending upon the investigated areas was 22.1% at Buan of Jellabukdo in 2008, 36% at Wando and Jindo of Jellanamdo in 2009, and 30.0% at Boeun of Chungcheongbukdo in 2이0. Average population density (APD) of overwintered SBPH was 13.1 heads in 2008, 13.9 heads in 2009 and 5.6 heads in 2이0. The highest APD was 39.1 and 60.4 heads at Buan of Jellabukdo in 2008 and 2009, respectively, and 14.0 heads at Pyungtaeg of Gyeonggido. The acreage of RSV occurred fields was 869 ha in the western and southern parts, mainly at Jindo and Wando areas, of Jellanamdo in 2008. In 2009, RSV occurred in the acreage of 21,541 ha covered whole country, especially, partial and whole plant death were occurred with infection rate of 55.2% at 3,025 plots in 53 Li, 39 Eup/Myun, 19 Si/Gun of Gyeonggido, Incheonsi, Chungcheongnamdo, Jeollabukdo and Jeollanamdo. Seasonal development of overwintered SBPH was investigated at Buan, Jeollabukdo, and Jindo, Jeollanamdo for 3 years from 2008. Most SBPH developed to the 3rd and 4th instar on the periods of May 20 to June 10,and they developed to the adult stage for the 1st generation on Mid and Late June. In 2009, all SBPH trapped by sky net trap were adult on May 31 to June 1 at Mid-western aeas of Taean, Seosan and Buan, and South-western areas of Sinan and Jindo. The population density of adult SBPH was 963 heads at Taean, 919 at Seocheon and 819 at Sinan area. The origin of these higher population of adult SBPH were verified from the population of non-overwintered SBPH but immigrant SBPH. From Mid May to Mid June in 2010, adult SBPH could not be counted as immigrant insects by sky net trap. The variation of RSV VIR was high with 2.1% to 9.5% for immigrant adult SBPH trapped by sky net trap at Hongsung of Chungcheongbukdo, Buan of Jeollabukdo and so forth in 2009. The highest VIR for the immigrant adult SBPH was 9.5% at Boryung of Chungcheongnamdo, followed by 7.9% at Hongsung

        Comparison of TNF-Mediated Glucose Catabolism between the TNF-Sensitive and -Resistant Cell Lines

        When murine fibrosarcoma L929 cells, a TNF-sensitive cell line, were treated with recombinant human tumor necrosis factor-a (rhTNF-α), the activities of glycolytic regulatory enzymes and lactate dehydrogenase increased up to 100∼150% compared to the control L929 cells after TNF treatment. By using various metabolic inhibitors and activators, it was found that cAMP-dependent protein kinase is responsible for the increase of activities of the glycolytic enzymes. The activities of glycolytic regulatory enzymes and lactate dehydrogenase of TNF-resistant A549 cells, a human lung carcinoma cell line, did not increase significantly compared to TNF-sensitive L929 cells upon TNF treatment. In contrast, the pyruvate carboxylase activities of A549 cells, but not L929 cells, increased up to 30∼40% after TNF treatment. The data suggest that pyruvate carboxylase activity may contribute to the compensation of energy loss mediated by TNF treatment in TNF-resistant A549 cells.

        First Report of Cucumber mosaic virus Infecting Pinewood Coneflower (Rudbeckia bicolor) in Korea

        A virus isolate causing symptoms of yellow mosaic, fern leaves, malformation and plant necrosis on Rudbeckia bicolor was prevalent around Pyeongchang area in Korea. The causal virus was identified as Cucumber mosaic virus (CMV) using characteristics from biological, serological and molecular analyses and named as CMV-Rb. CMV-Rb caused mosaic on Nicotiana benthamiana, N. tabacum, Capsicum annuum, and Lycopersicon esculentum. However, typical local lesions did not develop on inoculated Pisum sativum, Cucurbita moschata, Datura stramonium and Tetragonia expansa plants. Full-length genome sequences of CMV-Rb RNAs 1, 2 and 3 were obtained using 12 primer pairs by RT-PCR analysis. The genome of CMV-Rb RNA segments 1, 2, and 3 consists of 3363nt, 3049nt, and 2214nt in length, respectively. In order to ascertain their taxonomic identity, nucleotide and the deduced amino acid sequence analyses RNAs 1, 2 and 3 of CMV-Rb isolates were conducted with previously reported sequences of CMV strains and/or isolates. CMV-Rb RNAs showed about 90 to 99% sequence identity to those of subgroup I strains suggesting that CMV-Rb is more closely related to CMV isolates belong to subgroup I. To our knowledge, this is the first report of CMV on Rudbeckia bicolor in Korea.

        Analysis of Leak and Water Absorption Test Results for Water-Cooled Generator Stator Windings

        Cases of insulation breakdown damage of water-cooled generator stator windings occur frequently due to coolant leakage and water absorption worldwide. Such serious accidents may cause not only enormous economic loss but also very serious grid accidents in terms of stable supply of electric power. More than 50 % of domestic generators have been operated for more than 15 years, and leak and water absorption problem of windings are often found during the planned preventive maintenance period. Since 2005, leak and water absorption tests have been performed for total watercooled stator windings after fully drying the inside of the windings. The results are then comprehensively analyzed. The result of the test performed by GE, a foreign manufacturer, for 141 generators showed failures in 80 of them (failure rate: 57 %), whereas in the tests carried out in Korean domestic power plants, only 14 out of 50 generators showed failures (failure rate: 28 %).
