Human Embryonic Stem Cells Experience a Typical Apoptotic Process upon Oxidative Stress

Embryonic stem (ES) cells, derived from preimplantation embryos, are able to differentiate into various types of cells consisting the whole body, or pluripotency. In addition to the plasticity, ES cells are expected to be different from terminally differentiated cells in very many ways, such as patterns of gene expressions, ability and response of the cells in confronting environmental stimulations, metabolism, and growth rate. As a model system to differentiate these two types of cells, human ES (hES, MB03) cells and terminally differentiated cells (HeLa), we examined the ability of these two types of cells in confronting a severe oxidative insult, that is . Ratio of dying cells as determined by the relative amount of dye neutral red entrapped within the cells after the exposures. Cell death rates were not significantly different when either MB03 or HeLa were exposed up to 0.4 mM . However, relative amount of dye entrapped within the cells sharply decreased down to 0.12% in HeLa cells when the cells were exposed to 0.8 mM , while it was approximately 54% in MB03. Pretreatment of cells with BSO (GSH chelator) and measurement of GSH content results suggest that cellular GSH is the major defensive mechanism of hES cells. Induction of apoptosis in hES cell was confirmed by DNA laddering, induction of Bax, and chromatin condensation. In summary, hES cells 1) are extremely resistant to oxidative stress, 2) utilize GSH as a major defensive mechanism. and 3) experience apoptosis upon exposure to oxidative stress.
축력과 반복수평력을 받는 TR-CFT기둥에 관한 실험적 연구

CFT (Concrete filled steel tube) column has become popular for building construction due to not only its composite effect but also economic effect. However, the conventional CFT column also has its own disadvantages having plastic buckling at the end of column followed by the reduction of strength by yielding of steel tube. An experiment on TR-CFT (Transversely reinforced CFT) column are conducted for making up for conventional CFT column's disadvantages. The experiment parameters are strength of concrete, the layer numbers of carbon fiber sheet. In this study, hysteretic curve, initial stiffness, strength, plastic deformation capacity, and dissipated energy are compared and analyzed between CFT and TR-CFT columns. CFT(Concrete filled steel tube)기둥은 콘크리트와 강관의 이질적인 성질을 상호보완 할 수 있는 구조체 일뿐만 아니라 시공적인 측면에서도 공기단축으로 인한 공사원가 절감으로 인해 최근 널리 사용되고 있다. 그러나 기존CFT기둥은 강관의 항복이후 일정지점의 국부좌굴을 생기는 단점을 지니고 있다. 이를 개선하고자 본 연구에서는 예상국부좌굴위치에 탄소섬유쉬트로 보강을 한 TR(Tranversely reinforced) -CFT기둥에 대해 실험을 수행하였다. 실험변수는 콘크리트강도와 탄소섬유쉬트보강량이며 CFT실험체와 TR-CFT실험체의 이력곡선, 초기강성, 내력, 소성변형능력, 흡수에너지를 각각 비교분석하였다.

This paper presents an experimental results for the compression behavior of TR-CFT (Transversely Reinforced - Concrete Filled Steel Tubes) reinforced by carbon fiber sheets(CFSs). Nine specimens were made and axial compression tests were conducted. The various parameters are width-thickness ratio and the number of CFS layers. From compression tests, load-displacement curves of the specimens can be obtained. Finally, the failure behavior, maximum load capacity, and load-displacement curve are compared and analyzed with the conventional CFT and TR-CFT columns.

This paper presents experimental results for the deformation capacity of circular TR-CET (Transversely Reinforced - Concrete Filled Steel Tubes) reinforced by carbon fiber sheets(CFSs). Nine specimens were made and axial compression tests were conducted. The various parameters are diameter-thickness ratio and setting a cushion gap between surface of steel tube and CFSs. From compression tests, load-displacement curves of the specimens can be obtained. Finally, the deformation capacity is evaluated and compared with the conventional CIT, TR-CIT and TR-CIT with a gap.
둥근성게(Strongylocentrotus nudus)의 수정 및 배 발생률을 이용한 해양배출 폐기물의 독성평가

Toxicity of ocean dumping wastes (dye waste, urban sewage, food waste) were examined by observing fertilization and embryo development rates of the Sea Urchin, Strongylocentrotus nudus. Spawning was induced by injecting 1 ml, of 0.5 M KCl into coelomic cavity. Males released white or cream-colored sperms and females released yellow or orange-colored eggs. Experiments were began within 30 min after the collection of both gametes. The fertilization and embryo development rates tests were performed for 10 min and 48 h after fertilization, respectively. The fertilization and embryo development rates in the control condition (not including ocean dumping wastes sludge elutriate) were greater than 90%, but markedly decreased with increasing concentrations of ocean dumping waste sludge elutriate. The fertilization and normal embryogenesis rates were significantly inhibited in all waste sludge elutriate from dye waste (EC??=5.76; EC??=4.53), urban sewage (EC??=9.82; EC??=9.67) and food waste (EC??=3.90; EC??=3.27), respectively. The NOEC (>3.13%) and LOEC(3.13%) of fertiliztion and normal embryogenesis rates very similar in all waste sludge elutriate. These results suggest biological assay using the fertilization and embryo development rates of S. nudus are very useful test method for the ecological toxicity assessment of ocean dumping wastes.
