The effects of additions/deletions in glycosylated residues of recombinant human EPO (rhEPO) produced in CHO-K1 on their secretion were examined. hEPO cDNA was amplified from human liver mRNA and cloned into the pCR2.1 TOPO. Using overlapping-extension site-directed mutagenesis method, glycosylation sites at 24th, 38th, 83rd, and 126th were respectively or accumulatively removed by substituting its asparagine (or serine) with glutamine. To add novel glycosylation sites, 69 and 105th leucine was mutated to asparagine. Mutant and wild type rhEPO constructs were cloned into the pcDNA3 expression vector with CMV promoter and transfected into CHO cell line, CHO-K1, to produce mutant rhEPO mutant rhEPO proteins. Enzyme-linked immunosorbant assay (ELISA) and Western analysis with monoclonal anti-EPO antibody were performed using supernatants of the cultures showing transient and stable expressions respectively. Addition of novel glycosylation reduced rhEPO secretion dramatically while deletion mutants had little effect except some double deletion mutants (△24/83 and △38/83) and triple mutant (△24/38/83). This fact suggests that not single but combination of changes in glycosyl groups affect secretion of rhEPO in cell culture, possibly via changes in their conformations.
Control of Thermoelectric Properties through the addition of Ag in the Bi0.5 Sb1.5 Te3Alloy

In this study, the thermoelectric properties of the Ag-doped Bi0.5Sb1.5Te3 compounds were investigated in the temperature range from 323 K to 573 K. Ingots were fabricated by a conventional melting process and the powder crushed from ingots was then sintered using a hot-pressing method. The temperature dependence of the Seebeck coefficient and the electrical conductivity of the Ag-doped Bi0.5Sb1.5Te3compound are characteristic of degenerate semiconductors, which is fairly different from the conventional Bi0.5Sb1.5Te3 compound. The power factor (α 2 σ) of the quaternary compound was larger than that of the ternary, which is mainly due to the increase in the electrical conductivity with doping content of Ag. The thermal conductivity was greater than that of the Ag-freeBi0.5Sb1.5Te3compound in the temperature range from 323 K to 523 K. The lattice thermal conductivity showed low values throughout the temperature range. The maximum value of the dimensionless figure of merit (ZT) of the 0.05 wt. % Ag-doped compound and the ternary alloy were 1.2 at 373 K and 0.88 at 323 K, respectively. Each of the maximum peak ZT shifts to a higher temperature region with increases in the doping content of Ag. This is likely due to the control of the lattice thermal conductivity by the twin structure, which had a nano-ordered layer.

Objective : To evaluate that genetic factor(s) in chromosome 11q13 may control the expression of basophil histamine release (BaHR) after anti-IgE stimuli, linkage analysis between this phenotype and gene marker of chromosome 11q13 was performed. Material and methods : BaHR after anti-IgE stimuli and genotyping chromosome 11q13 using microsatellite marker (D11S97) was performed in 56 probands with asthma and 59 their sibs. The linkage between the phenotype and the genotype was evaluated by affected and quantitative trait locus (QTL) sib-pair analysis. Results : Maximal BaHR after anti-IgE were 43.3±3.5% in probands and 29.5±2.6% in sibs, respectively. Of 20 sib-pairs with the maximal BaHR more than 33%, 11 (55%) shared two D11S97 alleles, 9 (45%) shared one allele, neither sib-pair shared identical allele. The sharing rate of D11S97 alleles was 75.5%, which indicates linkage of the phenotype and the genotype (p= 0.026). Difference of the maximal BaHR between probands and their sibs was smaller in sib-pairs with two identical alleles than in those with one identical allele and with no identical allele (14.1±2.5% vs. 25.8±3.1% vs. 40.9±4.9%). Conclusion : Expression of basophil histamine release after anti-IgE stimuli was linked to gene marker of chromosome 11q13.

근래 국내 수력/화력발전소 보호제어반의 국산화를 통해 기술을 축적하였다. 확보된 핵심기술은 IEC 61850기반 발전기 보호를 위한 갑종 보호 IED(Intelligent Electronic Device) 시스템이다. 최근 원자력 발전소용 발전기 보호 IED를 위한 RTDS를 이용한 시뮬레이터 기반 보호계전기 및 계통의 동특성 현상 과제가 준비되고 있다. 원자력 발전소의 선진화, 현대화 및 IED의 설정 및 기능을 검토하기 위하여, 본 논문에서는 발전기 보호를 위한 거리계전기의 동작 특성을 해석하고자 한다. PSCAD/EMTDC를 이용하여 발전기 시스템을 모델링하고 선로의 사고를 시뮬레이션하여 거리계전 알고리즘에 의한 동작 특성을 해석하고자 한다.
