한국인 ADPKD 환자에서의 TGF-β1 Gene Leader Sequence 유전자 다형성의 분포 및 임상경과와의 연관성

목 적 : 상염색체우성다낭신(ADPKD) 환자에서 가계간·가계내의 임상양상의 심한 차이는 원인유전자 돌연변이 외에 modifying gene의 존재를 시사한다. 본 연구는 한국인 ADPKD 환자에서 TGF-β1 유전자 다형성의 분포와 고혈압, 신부전 등 임상상과의 연관성을 알아보고자 하였다. 방 법 : 정상 성인 47명과 ADPKD 환자 114명을 대상으로 TGF-β1 g ene leader sequence에 대한 PCR 시행한 후 제한효소 MspA 1I, Bg lI로 각각 처리하여 T 869C(Leu 10Pro)과 G915C(Arg25Pro)의 다형성을 보았다. 신부전, 고혈압 발생과의 연관성을 검정하였다. 결 과 : 각 유전자군의 비율은 Hardy -Weinberg 등식에 부합하였으며, TGF - β1의 BglI 처리결과는 125명 모두 GG형으로 서구인과 차이를 보였다(G : C=90 : 10). T GF - β1의 MspA1I 처리결과 정상인에서의 T : C allele의 빈도(56 : 44)는 서구인(65 : 35)과 차이를 보이지 않았으며, 총 PKD 환자군의 빈도(52 : 48)와도 통계적 차이를 보이지 않았다(p=0.670). PKD 환자군을 말기신부전 및 조기 고혈압 발생에 따라 나누었을 때 T : C allele 빈도의 차이는 없었다(p=0.571, p=0.252). 결 론 : 한국인 ADPKD 환자에서 TGF - β1 leader sequence 유전자 다형성과 임상경과 간에 유의한 관련은 없었고, 이는 싸이토카인의 유전적 차이보다는 물리적 요인 등 이차적인 원인에 의해 신조직 섬유화가 진행하는 것을 시사한다. Background : Two genetic loci, PKD1 and PKD2, have been identified as being responsible for ADPKD, and PKD1 is known to be associated with poor prognosis. However, the presence of intrafamilial clinical diversity suggests the presence of disease-modifying loci. Because the mechanism of renal failure in ADPKD includes cystic growth and tubulointerstitial atrophy and fibrosis, we studied the associations between two cytokine gene polymorphisms in the TGF-β1 gene, which are known to be related with chronic tubulointerstitial inflammation, and ADPKD progression in Korean patients. Methods : 47 normal controls and 114 individuals with ADPKD were genotyped by PCR-RFLP, and the TGF-β1 gene leader sequence of T 869C(Leu10-Pro) variant was compared with M spA 1I and G915C (Arg25Pro) with B glI. Statistic significances were determined using the Chi-square test. Results : The distribution of alleles for the T GF-β1 Leu10Pro polymorphism in ADPKD was : T 52%, C 48%, which w as similar to the Korean (56 : 44, p=0.670) and Western controls (65 : 35), and in addition, no differences were found between the CRF and the non-CRF groups (p=0.571) or the early hypertension and the normotension groups (p=0.252). T he distribution of alleles for the TGF-β1 Arg25Pro polymorphism was all GG type, which w as different from Western controls (90 : 10, p=0.000). Conclusion : Our results suggest that the polymorphism at Arg25Pro of TGF-β1 in Korean population has different allele distribution from Western, and the polymorphism at Leu10Pro of TGF-β1 has no association with the renal progression of Korean ADPKD patients.
The use of immunosuppressive drugs and legal implications in xenotransplantation

Transplantation is recognized as one of the greatest triumphs of modern medical science, but we are confronting the severe shortage of organs for transplantation. Xenotransplantation, the transplantation of nonhuman organs, tissues or cells into humans could be an alternative to fundamentally resolve this problem. Despite the potential benefit, xenotransplantation also gives us a potential risk, which are rejection and infection. The potential immunological rejection means the possibility of the human immune system to reject any foreign object. The rejection mechanism is very similar to that by which the body resists infection. The rejection of foreign object may endanger the life of the recipient. In xenotransplantation, immunosuppressive drugs can be clinically used to prevent the rejection of foreign object. However, these toxic drugs have side effects: the immune system loses its ability to successfully resist infections. The potential risk of infection means xenozoonosis that might result from xenotransplantation. For example, a pig virus, especially PERV might infect the human recipient, mutate and spread first to the close contacts of the recipient and then to the general population. This article primarily focuses on warning the risk of rejection and infection. This article also discusses clinicaltrials, informed consent and legal liability as legal aspects of xenotransplantation. Especially, an in-depth study is further required on legal implications and toxicology of xenotransplantation because the use of immunosuppressants for resolving immune rejection may bring lots of potential toxicologic problems. While clinical trials and informed consent are pretransplant prevention against the potential risk as preconditions for xenotransplantation, legal liability is posttransplant protection from it.
Characterization Method of Electric Field Probe by Using Transfer Standard in GTEM Cell

<P>In this paper, we introduce a simple and wideband characterization method for electric field probes using a transfer standard. As a transfer standard, a thin disk-type reference probe that can operate from 50 to 1000 MHz is used and calibrated using a micro transverse electromagnetic (mu-TEM) cell. A gigahertz transverse electromagnetic (GTEM) cell is used to generate a reference electric field. It has been shown that the uncertainty of the proposed method can be increased due to the imperfection of the field condition in the cell. According to the measurement result, the proposed characterization method for the electric probes agrees within 6.3%, compared with the method that uses the TEM cell and standard antennas in a fully anechoic chamber.</P>

For the production method, stamp base is increasing the market share. But also, by the manufacturing method, stiffness of the stamp base is small. Many studies have been carried out in order to increase the stiffness of the stamp base. In this study, we optimally designed according to the position of bolting using a screening method in order to increase the stiffness base. After establishing a simulation model, the maximum deformation and frequency of the first mode based having relevance with stiffness of the base, were optimized. There for, it was possible to expect of increasing the stiffness of the stamp base.
여성노인의 올바른 보행자세 평가를 위한 측정평가도구 개발

The purpose of this study was to develop a measurement and evaluation tool for analysis of posture and movement during gait, predict abnormal gait pattern, and suggest the basic data of correct position for the elderly women. For this study, 30 elderly women(76.73 ± 4.86(yrs), 152.57 ± 3.93(cm), 53.15 ± 6.34(kg)) who live in C city were participated. The developed gait tool was classified according to frontal and lateral gait movement. 13 items of classification and 14 questions of miner contents in frontal, 12 items of classification and 14 questions of miner contents in lateral were completed by literature reviews and experts consultation. Data analysis was used Cronbah-alpha and Intra-class correlation coefficient(ICC). The results were as follows; First, Frontal posture Cronbach-alpha in reliability analysis of the evaluated questions was .81. Second, Lateral posture Cronbach-alpha was .80. Third, ICCs of most questions by evaluator were significant in both frontal and lateral gait movements. Therefore, this study may suggest that this developed measurement instrument analyze gait posture correctly and be an effective guideline on their detailed movement for elderly women.

Alpha-fetoprotein(AFP) is a plasma protein produced in the fetal liver, yolk sac and gastrointestinal tract. The plasma level of AFP decreases markedly 1 year after birth. The AFP level is usually increased in hepatocellular carcinoma and yolk sac tumor but is rare in a primary lung cancer. We report a case of primary adenocarcinoma of lung producing high levels of AFP. (Tuberc Respir Dis 2006; 61: 70-73)

]Aliphatic aldehyde polyoxyethylene glycol acetals were synthesised through the reaction of aliphatic aldehydes such as caproic aldehyde, capryl aldehyde, capric aldehyde and lauric aldehyde with excess diethylene glycol, triethylene glycol and tetraethylene glycol, respectively. The acetal formation, in which water was azeotropically distilled by adding benzene to the reaction system, was gained a good yield of acetal type compounds. This reaction is found pseudo first order reaction at various temperatures such 70, 80, 90 and 97℃. Also these activation energies of reaction of acetal type products such as caproic aldehyde diethylene glycol acetal, capryl aldehyde diethylene glyco acetal, capric aldehyde diethylene glycol acetal, lauric aldehyde diethylene glycol acetal, caproic aldehyde triethylene glycol acetal and caproic aldehyde tetraethylene acetal were 17.3, 19,6, 21.2, 21.6, 15.5 and 14.7 Kcal/mole.
Antileishmanial Mechanism of Diamidines Involves Targeting Kinetoplasts

<B>ABSTRACT</B><P>Leishmaniasis is a disease caused by pathogenicLeishmaniaparasites; current treatments are toxic and expensive, and drug resistance has emerged. While pentamidine, a diamidine-type compound, is one of the treatments, its antileishmanial mechanism of action has not been investigated in depth. Here we tested several diamidines, including pentamidine and its analog DB75, againstLeishmania donovaniand elucidated their antileishmanial mechanisms. We identified three promising new antileishmanial diamidine compounds with 50% effective concentrations (EC50s) of 3.2, 3.4, and 4.5 μM, while pentamidine and DB75 exhibited EC50s of 1.46 and 20 μM, respectively. The most potent antileishmanial inhibitor, compound 1, showed strong DNA binding properties, with a shift in the melting temperature (Δ<I>Tm</I>) of 24.2°C, whereas pentamidine had a Δ<I>Tm</I>value of 2.1°C, and DB75 had a Δ<I>Tm</I>value of 7.7°C. Additionally, DB75 localized inL. donovanikinetoplast DNA (kDNA) and mitochondria but not in nuclear DNA (nDNA). For 2 new diamidines, strong localization signals were observed in kDNA at 1 μM, and at higher concentrations, the signals also appeared in nuclei. All tested diamidines showed selective and dose-dependent inhibition of kDNA, but not nDNA, replication, likely by inhibitingL. donovanitopoisomerase IB. Overall, these results suggest that diamidine antileishmanial compounds exert activity by accumulating toward and blocking replication of parasite kDNA.</P>
