        MicroRNA-181a-5p Curbs Osteogenic Differentiation and Bone Formation Partially Through Impairing Runx1-Dependent Inhibition of AIF-1 Transcription

        Background: Evidence has revealed the involvement of microRNAs (miRNAs) in modulating osteogenic differentiation, implying the promise of miRNA-based therapies for treating osteoporosis. This study investigated whether miR-181a-5p influences osteogenic differentiation and bone formation and aimed to establish the mechanisms in depth. Methods: Clinical serum samples were obtained from osteoporosis patients, and MC3T3-E1 cells were treated with osteogenic induction medium (OIM) to induce osteogenic differentiation. miR-181a-5p-, Runt-related transcription factor 1 (Runx1)-, and/or allograft inflammatory factor-1 (AIF-1)-associated oligonucleotides or vectors were transfected into MC3T3-E1 cells to explore their function in relation to the number of calcified nodules, alkaline phosphatase (ALP) staining and activity, expression levels of osteogenesis-related proteins, and apoptosis. Luciferase activity, RNA immunoprecipitation, and chromatin immunoprecipitation assays were employed to validate the binding relationship between miR-181a-5p and Runx1, and the transcriptional regulatory relationship between Runx1 and AIF-1. Ovariectomy (OVX)-induced mice were injected with a miR-181a-5p antagonist for in vivo verification. Results: miR-181a-5p was highly expressed in the serum of osteoporosis patients. OIM treatment decreased miR-181a-5p and AIF-1 expression, but promoted Runx1 expression in MC3T-E1 cells. Meanwhile, upregulated miR-181a-5p suppressed OIM-inducedincreases in calcified nodules, ALP content, and osteogenesis-related protein expression. Mechanically, miR-181a-5p targeted Runx1, which acted as a transcription factor to negatively modulate AIF-1 expression. Downregulated Runx1 suppressed the miR-181a-5p inhibitor-mediated promotion of osteogenic differentiation, and downregulated AIF-1 reversed the miR-181a-5p mimic-induced inhibition of osteogenic differentiation. Tail vein injection of a miR-181a-5p antagonist induced bone formation in OVX-induced osteoporotic mice. Conclusion: In conclusion, miR-181a-5p affects osteogenic differentiation and bone formation partially via the modulation of the Runx1/AIF-1 axis.

        Analysis of Local Surface Plasmon Resonance in Multilayered Au/Ag/Graphene Nanoshells

        "The localized surface plasmon resonance (LSPR) properties of Au/Ag/graphene nanoshells are studied by discrete dipole approximation (DDA). The coupled resonance wavelengths show a remarkable dependence on the graphene thickness as well as refractive index of the surrounding medium. The resonance wavelengths of Au/Ag/graphene nanoshells red-shift as the thickness of the graphene layer is increased, when the radii of the Au core and Ag interlayer are 40 nm and 45nm, respectively. Specifically, the longer wavelength red-shifts from 540 nm to 740 nm when the refractive index varies from 1.25 to 2.05. "

        Analysis of Localized Surface Plasmon Resonance in Ag/ITO/CdS/SiO2 Multilayered Nanostructured Composite

        Multilayered nanoshells have attracted much attention due to their unique optical, electronic and magnetic properties. In this work, numerical calculation using discrete dipole approximation (DDA) is conducted to investigate the quad-layered metal nanoshell consisting of a particle with a dielectric silica (SiO2) core, inner cadium sulfide (CdS) shell, middle indium tin oxide (ITO) shell and outer metal silver (Ag) shell. The phenomenon is interpreted by plasmon hybridization theory and the Ag–ITO–CdS–SiO2 multilayered nanoshells are studied by extinction spectra of localized surface plasmon resonance. The variation in the spectrum peak with nanoparticle thickness and refractive index of the surrounding medium is derived. The electric field enhancement contour around the nanoparticles under illumination is analyzed at the plasmon resonance wavelength. The |ω-->, |ω+->, and |ω-+> modes red-shift with the refractive index of the surrounding medium and increase in the layer thickness causes either blue-shift or red-shift as shown by the extinction spectra. The mechanism of the red-shift or blue-shift is discussed. The |ω--> blue-shifts and furthermore, the |ω-+> and |ω+-> modes of the Ag coated multilayered nanostructure are noticeable by comparing the extinction efficiency spectra of the Au–ITO–CdS–SiO2 and Ag–ITO–CdS–SiO2 multilayered nanoshells.

      • Soil Respiration and Its Effect Factors at Different Succession Stages during the Growing Season of Larix Gmelinii Forest in Great Xing’ an Mountain

        With Li-8100 chamber, the soil respiration rates in different succession stages of Larix gmelinii plantation in Great Xing’ an Mountains was measured from June to October, 2014. Results show that the variations of soil respiration can be expressed as a one-humped curve. The maximum of soil respiration rate occurs from July for early and mid-succession stage to August for late succession. Soil respiration rate varies from 1.2 to 4.6μmol ۬∙m-2∙s-1 with mean value of 2.76μmol ∙m-2∙s-1 for early succession, 1.5 to 5.6μmol ∙m-2∙s-1 with mean value of 3.17μmol ∙m-2∙s-1 for mid-succession, and 1.8 to 4.1μmol ∙m-2∙s-1 with mean value of 2.89μmol ∙m-2∙s-1 for late succession. Soil temperature is the main factor to affect soil respiration and there is a significant correlation between soil respiration and the soil temperature at 10cm depth in all different succession forests, but less correlation is observed between soil respiration and soil water content.

        The extinction spectral properties based on localized surface plasmon resonance (LSPR) of the concentric dual-ring nanodisk (CDRN) structure are investigated by discrete dipole approximation (DDA) and plasmon hybridization theory. The CDRN nanostructure shows flexible tunable multipole resonances in the near-infrared regime and the coupled resonance wavelengths depend on the structural parameters of the nanostructure, which has great potential in multichannel LSPR-based bio-sensing applications.

        Analyzing and validating the prognostic value and immune microenvironment of clear cell renal cell carcinoma

        Tumor immune microenvironment (TIME) plays an important role in tumor diagnosis, prevention, treatment and prognosis. However, the correlation and potential mechanism between clear cell renal cell carcinoma (ccRCC) and its TIME are not clear. Therefore, we aimed to identify potential prognostic biomarkers related to TIME of ccRCC. Unsupervised consensus clustering analysis was performed to divide patients into different immune subgroups according to their single-sample gene set enrichment analysis (ssGSEA) scores. Then, we validated the differences in immune cell infiltration, prognosis, clinical characteristics and expression levels of HLA and immune checkpoint genes between different immune subgroups. Weighted gene coexpression network analysis (WGCNA) was used to identify the significant modules and hub genes that were related to the immune subgroups. A nomogram was established to predict the overall survival (OS) outcomes after independent prognostic factors were identified by least absolute shrinkage and selection operator (LASSO) regression and multivariate Cox regression analyses. Five clusters (immune subgroups) were identified. There was no significant difference in age, sex or N stage. And there were significant differences in race, T stage, M stage, grade, prognosis and tumor microenvironment. WGCNA revealed that the red module has an important relationship with TIME, and obtained 14 hub genes. In addition, the nomogram containing LAG3 and GZMK accurately predicted OS outcomes of ccRCC patients. LAG3 and GZMK have a certain correlation with the prognosis of ccRCC patients, and play an important role in the TIME. These two hub genes deserve further study as biomarkers of the TIME.

        External Fixation Using Femoral Less Invasive Stabilization System Plate in Tibial Proximal Metaphyseal Fracture

        Background: The locking plates are often used for internal fixation of closed tibial fractures. The use of a locking plate as an externalfixator is still controversial, particularly for closed fractures. The purpose of this study is to evaluate the results of externalfixation using the femoral less invasive stabilization system (LISS) plate in proximal metaphyseal fractures of the tibia. Methods: We prospectively evaluated 35 patients (26 males and 9 females) with a mean age of 42 years (range, 21 to 62 years)who presented with fresh tibial proximal metaphyseal fractures. According to the AO Foundation and Orthopaedic Trauma Association(AO/OTA) classification, the fractures were identified as type 41-A2 in 18 cases and type 41-A3 in 17 cases, including 25closed fractures and 10 open fractures. The femoral LISS plate was used to fix these fractures, which was placed on the anteromedialaspect of the tibia as an external fixator. The mean follow-up period was 18 months (range, 13 to 22 months). Results: All fractures healed in a mean time of 14 weeks (range, 10 to 20 weeks). There was no case of nonunion, deep infection,and loosening of screws and plates. One month after the appearance of cortical bridging on biplanar radiographs, the locking platewas removed within 3 minutes in the clinic without any difficulty. According to the Hospital for Special Surgery (HSS) knee scoringsystem and American Orthopaedic Foot & Ankle Society (AOFAS) ankle scoring system, the mean HSS score was 91 (range, 85 to100) and 98 (range, 93 to 100), and the mean AOFAS score was 94 (range, 90 to 100) and 98 (range, 95 to 100) at 4 weeks postoperativelyand final follow-up, respectively. Conclusions: For proximal metaphyseal fracture of the tibia, external fixation using the femoral LISS plate is a safe and reliabletechnique with minimal complications and excellent outcomes. Its advantages include ease of performing the surgery, use of a lessinvasive technique, and convenience of plate removal after fracture healing.

        Performance of Contra-Rotating Propellers for Stratospheric Airships

        Small advance ratio and low Reynolds number of stratospheric propulsion system bring lots of challenges to the design of propellers. Contra-rotating propeller configuration is proposed to improve the propulsion efficiency. In this paper, the feasibility of contra-rotating propeller for stratospheric airship has been assessed and its performance has been investigated by wind tunnel tests. The experimental results indicate, at relatively low Reynolds number, although the advance ratio is fixed, the performance of propellers is different with variation of Reynolds number. Moreover, at the same Reynolds number, the efficiency of contra-rotating propeller achieved appears to be a few percent greater than that for a standard conventional propulsion system. It can be concluded that contra-rotating propellers would be an efficient means to improve the performance of stratospheric airship propulsion system.
