        FGD5-AS1 promotes growth and mobility of trophoblast cells by regulating IGF2 via sponging miR-16-5p in pre-eclampsia

        Background PE is a common pregnancy disease that can cause various pathologies in pregnant women. GEO microarray data revealed low expression of FGD5-AS1 in placenta tissues from patients with severe PE. As predicted by ENCORI, FGD5-AS1 sponges miR-16-5p, and IGF2, and promotes cell proliferation, differentiation, and metabolism. Objectives This study aimed to interrogate the interaction of FGD5-AS1/miR-16-5p/IGF2 axis and the mechanism of FGD5-AS1 in hypoxia-induced trophoblast cell injury. Results Our findings revealed that FGD5-AS1 and IGF2 were substantially decreased in PE patients than that in normal pregnant females, whereas miR-16-5p was increased in PE patients than that in normal pregnant females. In vitro studies showed that FGD5-AS1 were reduced in trophoblast cells after hypoxia treatment, accompanied by decreased viability and increased apoptosis, also reduced invasion, migration and angiogenesis. However, FGD5-AS1 overexpression substantially reversed these effects. Further mechanistic analysis showed that FGD5-AS1 could target miR-16-5p to regulate IGF2. FGD5-AS1 promoted the expression of IGF2 by sponging miR-16-5p, thereby increasing the viability of trophoblast cells after hypoxia treatment and promoting invasion and migration. Conclusion Overall, our findings demonstrated that FGD5-AS1 promoted growth and mobility of trophoblast cells by regulating miR-16-5p/IGF2 axis in PE.

        Screening nephroprotective compounds from cortex Moutan by mesangial cell extraction and UPLC

        A method for screening nephroprotective compoundsin cortex Moutan, a common traditional Chinesemedicine (TCM) in treating diabetic nephropathy with renalmesangial cell extraction and ultra performance liquidchromatography technique was described in this paper. Wehypothesize that the compounds which bind to cell membranesunder pathological conditions may be the bioactivecompounds in TCMs. Mesangial cells were cultured inmedium containing 5 mM (physiological, NG) or 30 mM(pathological, HG) glucose for 48 h and then incubated withcortex Moutan extract. After the unbound substances werewashed off, the cell membrane-bound compounds weredissociated and concentrated by an SPE column. By comparingthe chromatograms of NG and HG cultured-cellextractions and cortex Moutan extract, three compoundsbound to both NG and HG-cultured mesangial cells wereidentified as paeoniflorin, pentagalloylglucose (PGG) andpaeonol. In vitro studies showed that paeoniflorin, PGG andpaeonol reduced the activity of nicotinamide-adenine dinucleotidephosphate oxidase (NADPH) activity, anddecreased the level of reactive oxygen species (ROS),transforming growth factor-b1 (TGF-b1) and fibronectin inhigh glucose cultured mesangial cells. The results indicatethat paeonol, paeoniflorin and PGG may be the nephroprotectivecompounds from cortex Moutan. This study isexpected to provide a more reliable and effective method forscreening bioactive compounds from the complex TCM systems.

        External Fixation Using Femoral Less Invasive Stabilization System Plate in Tibial Proximal Metaphyseal Fracture

        Background: The locking plates are often used for internal fixation of closed tibial fractures. The use of a locking plate as an externalfixator is still controversial, particularly for closed fractures. The purpose of this study is to evaluate the results of externalfixation using the femoral less invasive stabilization system (LISS) plate in proximal metaphyseal fractures of the tibia. Methods: We prospectively evaluated 35 patients (26 males and 9 females) with a mean age of 42 years (range, 21 to 62 years)who presented with fresh tibial proximal metaphyseal fractures. According to the AO Foundation and Orthopaedic Trauma Association(AO/OTA) classification, the fractures were identified as type 41-A2 in 18 cases and type 41-A3 in 17 cases, including 25closed fractures and 10 open fractures. The femoral LISS plate was used to fix these fractures, which was placed on the anteromedialaspect of the tibia as an external fixator. The mean follow-up period was 18 months (range, 13 to 22 months). Results: All fractures healed in a mean time of 14 weeks (range, 10 to 20 weeks). There was no case of nonunion, deep infection,and loosening of screws and plates. One month after the appearance of cortical bridging on biplanar radiographs, the locking platewas removed within 3 minutes in the clinic without any difficulty. According to the Hospital for Special Surgery (HSS) knee scoringsystem and American Orthopaedic Foot & Ankle Society (AOFAS) ankle scoring system, the mean HSS score was 91 (range, 85 to100) and 98 (range, 93 to 100), and the mean AOFAS score was 94 (range, 90 to 100) and 98 (range, 95 to 100) at 4 weeks postoperativelyand final follow-up, respectively. Conclusions: For proximal metaphyseal fracture of the tibia, external fixation using the femoral LISS plate is a safe and reliabletechnique with minimal complications and excellent outcomes. Its advantages include ease of performing the surgery, use of a lessinvasive technique, and convenience of plate removal after fracture healing.

        Georgenia faecalis sp. nov. isolated from the faeces of Tibetan antelope

        Two aerobic, Gram-stain-positive, non-motile, non-sporulating coccoid strains, designated ZLJ0423T and ZLJ0321, were isolated from the faeces of Tibetan antelope (Pantholops hodgsonii). Their optimal temperature, NaCl concentration and pH for growth were 28°C, 0.5% (w/v) NaCl and pH 7.5, respectively. Phylogenetic analysis based on 16S rRNA gene sequences revealed that strains ZLJ0423T and ZLJ0321 were very similar to each other (99.8%) and had a sequence similarity of 97.0% with Georgenia satyanarayanai NBRC 107612T and Georgenia subflava CGMCC 1.12782T. Phylogenomic analysis based on 688 core genes indicated that these strains formed a clade with G. satyanarayanai NBRC 107612T and Georgenia wutianyii Z294T. The predominant cellular fatty acids were anteiso-C15:0, anteiso-C15:1 A and C16:0. The major menaquinone was MK-8(H4). The cell-wall amino acids consisted of alanine, lysine, glycine and aspartic acid, with lysine as the diagnostic diamino acid. Diphosphatidylglycerol, phosphatidylglycerol, phosphatidylinositol, phosphatidylinositol mannosides and two unidentified lipids formed the polar lipid profile. The DNA G + C content of both isolates was 73.9 mol%. The digital DNA–DNA hybridization value between strains ZLJ0423T and ZLJ0321 was 91.2%, but their values with closely related species and other available type strains of the genus Georgenia were lower than the 70% threshold. On the basis of polyphasic taxonomic data, strains ZLJ0423T and ZLJ0321 represent a novel species within the genus Georgenia, for which the name Georgenia faecalis sp. nov. is proposed. The type strain is ZLJ0423T (= CGMCC 1.13681T = JCM 33470T).
