Ginsenoside Rh2 inhibiting HCT116 colon cancer cell proliferation through blocking PDZ-binding kinase/T-LAK cell-originated protein kinase

        Yang, Jianjun,Yuan, Donghong,Xing, Tongchao,Su, Hongli,Zhang, Shengjun,Wen, Jiansheng,Bai, Qiqiang,Dang, Dongmei The Korean Society of Ginseng 2016 Journal of Ginseng Research Vol.40 No.4

Background: Ginsenoside Rh2 (GRh2) is the main bioactive component in American ginseng, a commonly used herb, and its antitumor activity had been studied in previous studies. PDZ-binding kinase/T-LAK cell-originated protein kinase (PBK/TOPK), a serine/threonine protein kinase, is highly expressed in HCT116 colorectal cancer cells. Methods: We examined the effect of GRh2 on HCT116 cells ex vivo. Next, we performed in vitro binding assay and in vitro kinase assay to search for the target of GRh2. Furthermore, we elucidated the underlying molecular mechanisms for the antitumor effect of GRh2 ex vivo and in vivo. Results: The results of our in vitro studies indicated that GRh2 can directly bind with PBK/TOPK and GRh2 also can directly inhibit PBK/TOPK activity. Ex vivo studies showed that GRh2 significantly induced cell death in HCT116 colorectal cancer cells. Further mechanistic study demonstrated that these compounds inhibited the phosphorylation levels of the extracellular regulated protein kinases 1/2 (ERK1/2) and (H3) in HCT116 colorectal cancer cells. In vivo studies showed GRh2 inhibited the growth of xenograft tumors of HCT116 cells and inhibited the phosphorylation levels of the extracellular regulated protein kinases 1/2 and histone H3. Conclusion: The results indicate that GRh2 exerts promising antitumor effect that is specific to human HCT116 colorectal cancer cells through inhibiting the activity of PBK/TOPK.

Adaptive Finite Time Control for Wearable Exoskeletons Based on Ultra-local Model and Radial Basis Function Neural Network

        Jianjun Sun,Jie Wang,Peng Yang,Yan Zhang,Lingling Chen 제어·로봇·시스템학회 2021 International Journal of Control, Automation, and Vol.19 No.2

This paper investigates an adaptive finite time control scheme for wearable exoskeletons to realize trajectory tracking control. The main feature of the proposed scheme is model-free in which no dynamic models are required except for the input and output data. Firstly, a second order ultra-local model is employed to replace the complex dynamic model of wearable exoskeleton as the controlled object, which is a novel designed model. In addition, a nonsingular fast terminal sliding surface is proposed to design the controller to guarantee the finite time convergence of tracking errors, and a radial basis function (RBF) neural network is developed to approximate the lumped disturbance in the ultra-local model. Then the stability of closed-loop system is proved by Lyapunov theory. Finally, to validate the proposed control scheme, virtual prototype is designed in SolidWorks and transferred to MATLAB, then visual simulation program is implemented based on SimMechanics. What’s more, reference trajectories are extracted from the measured data of DELSYS Electromyography (EMG) recording system. Theeffectiveness of proposed scheme is demonstrated by the simulation results.

Individual DC Voltage Balancing Method at Zero Current Mode for Cascaded H-bridge Based Static Synchronous Compensator

Yang, Zezhou,Sun, Jianjun,Li, Shangsheng,Liao, Zhiqiang,Zha, Xiaoming The Korean Institute of Electrical Engineers 2018 Journal of Electrical Engineering & Technology Vol.13 No.1

Individual DC voltage balance problem is an inherent issue for cascaded H-bridge (CHB) based converter. When the CHB-based static synchronous compensator (STATCOM) is operating at zero current mode, the software-based individual DC voltage balancing control techniques may not work because of the infinitesimal output current. However, the different power losses of each cell would lead to the individual DC voltages unbalance. The uneven power losses on the local supplied cell-controllers (including the control circuit and drive circuit) would especially cause the divergence of individual DC voltages, due to their characteristic as constant power loads. To solve this problem, this paper proposes an adaptive voltage balancing module which is designed in the cell-controller board with small size and low cost circuits. It is controlled to make the power loss of the cell a constant resistance load, thus the DC voltages are balanced in zero current mode. Field test in a 10kV STATCOM confirms the performance of the proposed method.

Vibration Characteristics of Cutter-Head in Soft-Hard Mixed Stratum: An Experimental Case Study on Su’ai Tunnel

        Zhenxing Yang,Dongjiang Pan,Jianjun Zhou,Jian Chen,Zhenchuan Sun,Haining Liu 대한토목학회 2020 KSCE Journal of Civil Engineering Vol.24 No.4

This paper conducts a simulation experiment research on cutter-head vibration characteristics by evaluating a soft-hard mixed stratum (the upper layer is silty soil, whereas the lower layer is moderately weathered granite of maximum uniaxial compressive strength (UCS) 142.0 MPa and slightly weathered granite of maximum 210.0 MPa). The ratios of hard rock and soil within the tunnel face were designed to be 1:1 and 1:2. Then, the cutter-head and stratum model were subjected to vibration analysis by reasonably deployed accelerometers. Results show that: 1) different conditions of heterogeneous strata mainly affected the acceleration amplitude and frequency distribution of cutter-head, in other words, the stronger hard rock, the larger the strength difference between the hard rock and soil within a tunnel face, the larger the vibration acceleration and the more concentrated the vibration frequency distribution; 2) the radial and circumferential vibration acceleration of the cutter-head in different positions were not largely different; however, axial accelerations were largely different. The axial vibration accelerations first increased and then decreased along the radial direction of cutter-head. Considering the difference between Multi-Mode Boring Test System and real cutter-head structure, the radius of joint between the cutter-head and the flange plate should be reasonably increased according to the ratio of hard rock in tunnel face; 3) with the increasing difference between the strength of the hard rock and soil, the circumferential and axial vibration acceleration multiplied. Thus, the soil remarkably reduces the vibration of the cutter-head. These results can provide some evidence of potential application for advancing manufacturing practice for cutter-head and disc cutter of shield machine.

BK Channel Deficiency in Osteoblasts Reduces Bone Formation via the Wnt/β-Catenin Pathway

        Jiang, Lan,Yang, Qianhong,Gao, Jianjun,Yang, Jiahong,He, Jiaqi,Xin, Hong,Zhang, Xuemei Korean Society for Molecular and Cellular Biology 2021 Molecules and cells Vol.44 No.8

Global knockout of the BK channel has been proven to affect bone formation; however, whether it directly affects osteoblast differentiation and the mechanism are elusive. In the current study, we further investigated the role of BK channels in bone development and explored whether BK channels impacted the differentiation and proliferation of osteoblasts via the canonical Wnt signaling pathway. Our findings demonstrated that knockout of Kcnma1 disrupted the osteogenesis of osteoblasts and inhibited the stabilization of β-catenin. Western blot analysis showed that the protein levels of Axin1 and USP7 increased when Kcnma1 was deficient. Together, this study confirmed that BK ablation decreased bone mass via the Wnt/β-catenin signaling pathway. Our findings also showed that USP7 might have the ability to stabilize the activity of Axin1, which would increase the degradation of β-catenin in osteoblasts.

Chinese Society of Allergy and Chinese Society of Otorhinolaryngology-Head and Neck Surgery Guideline for Chronic Rhinosinusitis

        Zheng Liu,Jianjun Chen,Lei Cheng,Huabin Li,Shixi Liu,Hongfei Lou,Jianbo Shi,Ying Sun,Dehui Wang,Chengshuo Wang,Xiangdong Wang,Yongxiang Wei,Weiping Wen,Pingchang Yang,Qintai Yang,Gehua Zhang,Yuan Zhan 대한천식알레르기학회 2020 Allergy, Asthma & Immunology Research Vol.12 No.2

The current document is based on a consensus reached by a panel of experts from the Chinese Society of Allergy and the Chinese Society of Otorhinolaryngology-Head and Neck Surgery, Rhinology Group. Chronic rhinosinusitis (CRS) affects approximately 8% of Chinese adults. The inflammatory and remodeling mechanisms of CRS in the Chinese population differ from those observed in the populations of European descent. Recently, precision medicine has been used to treat inflammation by targeting key biomarkers that are involved in the process. However, there are no CRS guidelines or a consensus available from China that can be shared with the international academia. The guidelines presented in this paper cover the epidemiology, economic burden, genetics and epigenetics, mechanisms, phenotypes and endotypes, diagnosis and differential diagnosis, management, and the current status of CRS in China. These guidelines—with a focus on China—will improve the abilities of clinical and medical staff during the treatment of CRS. Additionally, they will help international agencies in improving the verification of CRS endotypes, mapping of eosinophilic shifts, the identification of suitable biomarkers for endotyping, and predicting responses to therapies. In conclusion, these guidelines will help select therapies, such as pharmacotherapy, surgical approaches and innovative biotherapeutics, which are tailored to each of the individual CRS endotypes.

Chinese Society of Allergy Guidelines for Diagnosis and Treatment of Allergic Rhinitis

        Lei Cheng,Jianjun Chen,Qingling Fu,Shaoheng He,Huabin Li,Zheng Liu,Guolin Tan,Zezhang Tao,Dehui Wang,Weiping Wen,Rui Xu,Yu Xu,Qintai Yang,Chonghua Zhang,Gehua Zhang,Ruxin Zhang,Yuan Zhang,Bing Zhou,Do 대한천식알레르기학회 2018 Allergy, Asthma & Immunology Research Vol.10 No.4

Allergic rhinitis (AR) is a global health problem that causes major illnesses and disabilities worldwide. Epidemiologic studies have demonstrated that the prevalence of AR has increased progressively over the last few decades in more developed countries and currently affects up to 40% of the population worldwide. Likewise, a rising trend of AR has also been observed over the last 2-3 decades in developing countries including China, with the prevalence of AR varying widely in these countries. A survey of self-reported AR over a 6-year period in the general Chinese adult population reported that the standardized prevalence of adult AR increased from 11.1% in 2005 to 17.6% in 2011. An increasing number of original articles and imporclinical trials on the epidemiology, pathophysiologic mechanisms, diagnosis, management and comorbidities of AR in Chinese subjects have been published in international peer-reviewed journals over the past 2 decades, and substantially added to our understanding of this disease as a global problem. Although guidelines for the diagnosis and treatment of AR in Chinese subjects have also been published, they have not been translated into English and therefore not generally accessible for reference to non-Chinese speaking international medical communities. Moreover, methods for the diagnosis and treatment of AR in China have not been standardized entirely and some patients are still treated according to regional preferences. Thus, the present guidelines have been developed by the Chinese Society of Allergy to be accessible to both national and international medical communities involved in the management of AR patients. These guidelines have been prepared in line with existing international guidelines to provide evidence-based recommendations for the diagnosis and management of AR in China.

Sparsity Adaptive Expectation Maximization Algorithm for Estimating Channels in MIMO Cooperation systems

( Aihua Zhang ),( Shouyi Yang ),( Jianjun Li ),( Chunlei Li ),( Zhoufeng Liu ) 한국인터넷정보학회 2016 KSII Transactions on Internet and Information Syst Vol.10 No.8

We investigate the channel state information (CSI) in multi-input multi-output (MIMO) cooperative networks that employ the amplify-and-forward transmission scheme. Least squares and expectation conditional maximization have been proposed in the system. However, neither of these two approaches takes advantage of channel sparsity, and they cause estimation performance loss. Unlike linear channel estimation methods, several compressed channel estimation methods are proposed in this study to exploit the sparsity of the MIMO cooperative channels based on the theory of compressed sensing. First, the channel estimation problem is formulated as a compressed sensing problem by using sparse decomposition theory. Second, the lower bound is derived for the estimation, and the MIMO relay channel is reconstructed via compressive sampling matching pursuit algorithms. Finally, based on this model, we propose a novel algorithm so called sparsity adaptive expectation maximization (SAEM) by using Kalman filter and expectation maximization algorithm so that it can exploit channel sparsity alternatively and also track the true support set of time-varying channel. Kalman filter is used to provide soft information of transmitted signals to the EM-based algorithm. Various numerical simulation results indicate that the proposed sparse channel estimation technique outperforms the previous estimation schemes.
