        Polysaccharides from the Fleshy Fruits of Camellia oleifera Attenuate the Kidney Injury in High-Fat Diet/Streptozotocin-Induced Diabetic Mice

        The fleshy fruits of Camellia oleifera Abel are the immature fruits of C. oleifera, which are infected by Exobasidium vexans Massee and then turn to be intumescent and hollowed. They contain rich trace elements and vitamin C and are eaten directly as wild fruits in the Chinese countryside. Recent studies report that C. oleifera has anti-inflammatory and antioxidative effects. The current study, for the first time, evaluates the renal protective capacity of polysaccharides from the fleshy fruits of C. oleifera (CFFP) in streptozotocin-induced diabetic mice fed high-fat diets. The diabetic mice were orally administered CFFP for 3 months to evaluate the renoprotective function of CFFP. Our results indicated that 250 mg/kg CFFP significantly alleviated diabetes-induced renal injury by decreasing serum creatine, blood urea nitrogen levels, the kidney/body weight ratio, expression of fibronectin and collagen, as well as the secretion of tumor necrosis factor-α and interleukin-6. Additionally, 250 mg/kg CFFP could significantly ameliorate renal oxidative stress through increasing glutathione levels and lowering malondialdehyde contents. We confirmed that CFFP could exert antioxidative, anti-inflammatory, and antifibrosis activities. CFFP might be a potential therapeutic agent, and the fleshy fruits of C. oleifera might be a diet therapy for diabetic patients in the future.

      • Which Factors Help Visitors Convert Their Short-Term Pro-Environmental Intentions to Long-Term Behaviors?

        The theoretical perspective of this study is to integrate Visitor Environmental Learning (VEL) into the Theory of Planed Behavior (TPB). The Xiangjiang Safari Park, which is one of the largest theme parks in Asia,was selected to test the conceptual model to examine whether Post-Visit Action Resources (PVAR), Ease of Behavior (EB), and Sunk Cost (SC) have the mediate/moderate effect on the relationship between short-term pro-environment intentions and long-term behaviors. The sample consisted of 269 tourists who have travel experience with the Xiangjiang Safari Park. Findings from a Structural Equation Model analysis show that post-visit action resources partially mediate the relationship between tourists’ short-term pro-environmental intentions and behaviors, and that ease of behavior and sunk cost both have moderating effects on this relationship. These findings indicate that a high level of post-visit action resources, ease of behavior and sunk cost are instrumental in encouraging visitors’ pro-environmental intentions to turn into behaviors.

        Channel Capacity Analysis of DNA-based Molecular Communication with Length Encoding Mechanism

        The double helix structure of DNA makes it diverse, stable and can store information with high density, and these characteristics are consistent with the requirements of molecular communication for transport carriers. In this paper, a specific structure of molecular communication system based on DNA length coding is proposed. Transmitter (Tx) adopts the multi-layer golden foil design to control the release of DNA molecules of different lengths accurately, and receiver (Rx) adopts an effective and sensitive design of nanopore, and the biological information can be converted to the electric signal at Rx. The effect of some key factors, e.g., the length of time slot, transmission distance, the number of releasing molecules, the priori probability, on channel capacity is demonstrated exhaustively. Moreover, we also compare the transmission capacity of DNA-based molecular communication (DNA-MC) system and concentration-based molecular communication (MC) system under the same parameter setting, and the peak value of capacity of DNA-MC system can achieve 0.08 bps, while the capacity of MC system remains 0.025 bps. The simulation results show that DNA-MC system has obvious advantages over MC system in saving molecular resources and improving transmission stability.

Through these object of investigations we have came to three conclusions. Second, now specialty stores are Increase the spatial attributes and experience Space"s function and positions have been found. Third, by close to the life of the show, increase the choice of consumers. This is a new development trend and need to do more research in the future.

        Comparative Transcriptomics Identifies Different Gene Expression Networks Regulating Gametic Development in Arabidopsis

        Gamete development is a complex process during the plant reproductive life cycle. A global vision of the molecular mechanisms regulating gametic development would enhance our understanding of developmental similarities and differences between male and female reproductive pathways in plants. In this study, we applied comparative transcriptomics to characterize gene expression during sporogenesis and gametogenesis in Arabidopsis. Compared with genes expressed in leaves, 11,112 and 10,133 differentially expressed genes (DEGs) were identified in the anthers and ovules, respectively. The shared DEGs exhibited a similar expression pattern regarding cell cycle and chromatin dynamics, but DEGs associated with cell wall formation and pectin modifications were prevalent during male development, reflecting dynamic changes during pollen wall development in anthers. In contrast, DEGs related to DNA repair, homologous recombination, and RNA-directed DNA methylation (RdDM) were comparatively over-represented in ovules, probably indicating a relatively intensified mechanism for supervising genome fidelity during female gametogenesis. In addition, DEGs related to brassinolide (BR) synthesis and signaling pathway were also highly enriched in ovules, suggesting a pivotal role in female gamete development. Collectively, our results provide a contrasting transcriptional atlas in developing anthers and ovules, and shed new light on our global understanding of gamete development in plants.

        Glasgow prognostic score and combined positive score for locally advanced rectal cancer

        Purpose: This study was performed to investigate the association of Glasgow prognostic score (GPS), combined positive score (CPS), and clinicopathological characteristics of locally advanced rectal cancer. Methods: Between February 2012 and February 2018, 103 patients with locally advanced rectal cancer treated by neoadjuvant chemoradiotherapy and total mesorectal excision (TME) were retrospectively evaluated. Results: According to the classification of the GPS, 85 (82.5%), 13 (12.6%), and 5 patients (4.9%) were classified as a score of 0, 1, and 2, respectively. Patients were classified into the GPS-low group (GPS of 0, n = 85) and GPS-high group (GPS of 1 or 2, n = 18) with an area under the curve of 0.582 for overall survival (OS). The mean programmed death-ligand 1 (PD-L1) CPS of the whole group was 2.24 (range, 0–70). The PD-L1 CPS of the GPS-high group was higher than the GPS-low group (P < 0.001). Multivariate analysis by Cox proportional hazards model indicated that GPS was associated with OS and disease- free survival (DFS). Furthermore, PD-L1 CPS was associated with DFS (hazard ratio, 1.050; 95% confidence interval, 1.017– 1.083; P = 0.003). Conclusion: Elevated GPS was related to the PD-L1 CPS. GPS and PD-L1 CPS were associated with the prognosis of locally advanced rectal cancer treated with neoadjuvant chemoradiotherapy followed by TME.

        Transcriptome analysis of response strategy in Hemerocallis fulva under drought stress

        Background Hemerocallis fulva is an important ground cover plant widely used in urban greening. The analysis of the molecular mechanism underlying the drought response of H. fulva can lay a foundation for improving its adaptability and expanding its planting area. Objective To reveal the drought response mechanisms of H. fulva, identify candidate unigenes associated with drought response, and lay a foundation for further unigenes functional study and drought resistance improvement of H. fulva via genetic engineering. Methods RNA was isolated from H. fulva under different experimental conditions. De novo transcriptomic analysis of the samples was performed to screen drought response unigenes. The transcriptional changes of candidate drought response unigenes were verified by quantitative real-time PCR. Results The differentially expressed unigenes and their functions were analyzed after H. fulva treated by PEG-simulated drought stress and rewatering. The candidate unigenes, associated with H. fulva drought response, were identified after transcriptome analysis. Then, the transcription level of drought response unigenes of H. fulva under different conditions was further verified. Abscisic acid, protein phosphorylation, sterol biosynthesis and ion transport were involved in drought response with quick restore in H. fulva. The response unigenes, involved in hormone (ABA, JA, CK and GA) signaling pathways, defense response, high light response, karrikin response and leaf shaping, can maintain at changed expression levels even after stress withdraw. Conclusion Hemerocallis fulva has unique drought response mechanism. Negative regulation mechanism may play more important roles in drought response of H. fulva. The analysis of candidate unigenes, associated with drought response, lays a foundation for further drought resistance improvement of H. fulva.

        Effect of serotonin on the cell viability of the bovine mammary alveolar cell-T (MAC-T) cell line

        5-Hydroxytryptamine (5-HT), a monoamine, as a local regulator in the mammary gland is achemical signal produced by the mammary epithelium cell. In cows, studies have shown that5-HT is associated with epithelial cell apoptosis during the degenerative phase of the mammarygland. However, studies in other tissues have shown that 5-HT can effectively promotecell viability. Whether 5-HT could have an effect on mammary cell viability in dairy cows isstill unknown. The purpose of this study was to determine: (1) effect of 5-HT on the viability ofbovine mammary epithelial cells and its related signaling pathways, (2) interaction betweenprolactin (PRL) and 5-HT on the cell viability. The bovine mammary alveolar cell-T (MAC-T)were cultured with different concentrations of 5-HT for 12, 24, 48 or 72 hours, and then wereassayed using cell counting kit-8, polymerase chain reaction (PCR) and immunobloting. Theresults suggested that 20 μM 5-HT treatment for 12 or 24 h promote cell viability, which wasmainly induced by the activation of 5-HT receptor (5-HTR) 1B and 4, because the increasecaused by 5-HT vanished when 5-HTR 1B and 4 was blocked by SB224289 and SB204070. And protein expression of mammalian target of rapamycin (mTOR), eukaryotic translationelongation factor 2 (eEF2), janus kinase 2 (JAK2) and signal transducer and activator of transcription5 (STAT5) were decreased after blocking 5-HT 1B and 4 receptors. When MAC-Tcells were treated with 5-HT and PRL simultaneously for 24 h, both the cell viability and thelevel of mTOR protein were significantly higher than that cultured with 5-HT or PRL alone. Inconclusion, our study suggested that 5-HT promotes the viability of MAC-T cells by 5-HTR1B and/or 4. Furthermore, there is a reciprocal relationship between PRL and 5-HT.

        A fluorescent probe for colorimetric detection of bisulfite and application in sugar and red wine

        A new fluorescent probe made from (E)-2-(benzo[d]thiazol-2-yl)-3-(6-hydroxynaphthalen-2-yl) acrylonitrile(Probe 1) was synthesized for the determination ofbisulfite concentrations in real food samples (red wine andsugar). Adding bisulfite to a Probe 1 solution caused amarked decrease in fluorescence intensity and a visualcolor change from yellow to light yellow. This distinctcolor response indicates that Probe 1 could be used as avisual sensor for bisulfite. Probe 1 can detect bisulfitequantitatively in the range 0–400 lM with a detection limitof 0.10 lM. This makes Probe 1 a convenient signalinginstrument for determining bisulfite levels in sugar and redwine samples.

        A study on the bearing effect of pier column backfilling in the goaf of a thin coal seam

        The pier column backfilling method in the goaf of a thin coal seam introduced in this paper is the latest backfilling technology in China, which has the combined advantages of high efficiency and low cost. In order to fully verify the load bearing effect of this backfilling method, a comprehensive evaluation model which combined the actual measurement, numerical simulation and physical simulation is implemented in this paper. First, the stress bearing formula and backfilling volume rate of a pier column backfilling body is obtained by theory analysis. Then, through field monitoring, it was concluded that the bearing capacity when the backfilling body reached a stable state was similar to the results of theoretical calculation, and no obvious deformation occurred by observation the roadway deformation with the site. Finally, based on the drilling columnar and lithological parameters of the site, corresponding physical and numerical models were established and used to study the bearing effect of this backfilling method. It could be found that the stresses and displacements data that resulted from the simulation relatively confirmed data obtained from the site. After comprehension of above researches, it is concluded that the pier column backfilling method in a goaf has good bearing effect and a good economic benefit for mining thin coal seams.
