2019 LHI journal of land, housing, and urban affairs Vol.10 No.3
In this study, the author suggests utilizing unsold and unused land as temporary commercial parking lots as a way to raise funds for active public transport use. In addition, reasonable parking fee scheme is suggested by estimating marginal parking fee and marginal parking distance for the commercial parking lots based on the CVM. The author conducts a survey to investigate citizens' opinion on utilizing unsold and unused land in the urban development project area as temporary commercial transport facilities such as parking lots. Based on survey outcome, travel behavior and requirements are analyzed and marginal parking fee and marginal parking distance are estimated through the Contingent Valuation Method (CVM). The results are as follows: in the single-bounded dichotomous choice model, people are willing to pay 216 KRW for 165m in average for 10 minutes and their willingness to pay goes down against reaching distance to the parking lots from 365 KRW for 50m, 295 KRW for 100m, 173 KRW for 200m, and 51 KRW for 300m; in the double-bounded dichotomous choice model, people are willing to pay 285 KRW for 165m in average for 10 minutes and their willingness to pay goes down as well against reaching distance to the parking lots from 310 KRW for 50m, 297 KRW for 100m, 272 KRW for 200m and 248 KRW for 300m. It is clearly appeared that people's willingness to pay goes higher as distance to reach the parking lots goes shorter.

Clioquinol induces autophagy in cultured astrocytes and neurons by acting as a zinc ionophore
Park, Mi-Ha,Lee, Sook-Jeong,Byun, Hyae-ran,Kim, Yunha,Oh, Young J.,Koh, Jae-Young,Hwang, Jung Jin Elsevier 2011 Neurobiology of disease Vol.42 No.3

Abstract

Recent studies have demonstrated that clioquinol, an antibiotic with an anti-amyloid effect, acts as a zinc ionophore under physiological conditions. Because increases in labile zinc may induce autophagy, we examined whether clioquinol induces autophagy in cultured astrocytes in a zinc-dependent manner.

Within 1h of exposure to 0.1–10μM clioquinol, the levels of microtubule-associated protein 1 light chain 3 (LC3)-II, a marker of autophagy, began to increase in astrocytes. Confocal live-cell imaging of GFP-LC3-transfected astrocytes showed the formation of LC3(+) autophagic vacuoles (AVs), providing a further indication that clioquinol induced autophagy. Addition of 3-methyladenine or small-interfering RNA against autophagy-related gene 6 (ATG6/Beclin-1) blocked clioquinol-induced increases in LC3-II. FluoZin-3 fluorescence microscopy showed that, like the zinc ionophore pyrithione, clioquinol increased intracellular zinc levels in the cytosol and AVs in an extracellular zinc-dependent manner. Zinc chelation with N,N,N′,N′-tetrakis-(2-pyridylmethyl) ethylenediamine (TPEN) reduced, and addition of zinc increased the levels of LC3-II and LC3(+) puncta, indicating that zinc influx plays a key role therein. Moreover, astrocytes and SH-SY5Y cells expressing mutant huntingtin (mHttQ74) accumulated less aggregates when treated with clioquinol, and this effect was reversed by TPEN. These results indicate that clioquinol-induced autophagy is likely to be physiologically functional.

The present study demonstrates that clioquinol induces autophagy in a zinc-dependent manner and contributes to clearance of aggregated proteins in astrocytes and neurons. Hence, in addition to its metal-chelating effect in and around amyloid beta (Aβ) plaques, clioquinol may contribute to the reduction of Aβ loads by activating autophagy by increasing or normalizing intracellular zinc levels in brain cells.

Research Highlights

► Clioquinol acts as a zinc ionophore and introduces zinc into autophagic vacuoles. ► Clioquinol induces autophagy in astrocytic cultures in a zinc-dependent manner. ► Autophagy induced by clioquinol reduces accumulation of huntingtin aggregates. ► Excessive clioquinol induces autophagic cell death especially in neurons. ► In conclusion, adequate doses of clioquinol may have beneficial actions in the Alzheimer's Diseases brains.

The Immunosuppressive Potential of Cholesterol Sulfate Through T Cell Microvilli Disruption
Park Jeong-Su,Chung Ik-Joo,Kim Hye-Ran,전창덕 대한면역학회 2023 Immune Network Vol.23 No.3
Cholesterol (CL) is required for various biomolecular production processes, including those of cell membrane components. Therefore, to meet these needs, CL is converted into various derivatives. Among these derivatives is cholesterol sulfate (CS), a naturally produced CL derivative by the sulfotransferase family 2B1 (SULT2B1), which is widely present in human plasma. CS is involved in cell membrane stabilization, blood clotting, keratinocyte differentiation, and TCR nanocluster deformation. This study shows that treatment of T cells with CS resulted in the decreased surface expression of some surface T-cell proteins and reduced IL-2 release. Furthermore, T cells treated with CS significantly reduced lipid raft contents and membrane CLs. Surprisingly, using the electron microscope, we also observed that CS led to the disruption of T-cell microvilli, releasing small microvilli particles containing TCRs and other microvillar proteins. However, in vivo, T cells with CS showed aberrant migration to high endothelial venules and limited infiltrating splenic T-cell zones compared with the untreated T cells. Additionally, we observed significant alleviation of atopic dermatitis in mice injected with CS in the animal model. Based on these results, we conclude that CS is an immunosuppressive natural lipid that impairs TCR signaling by disrupting microvillar function in T cells, suggesting its usefulness as a therapeutic agent for alleviating T-cell-mediated hypersensitivity and a potential target for treating autoimmune diseases.

Transparent In-doped (1 at.%) zinc oxide (IZO) thin films are deposited by pulsed DC magnetron sputtering with H2 mixed Ar atmosphere on glass substrate without any heating process. Even at room temperature, highly c-axis oriented IZO thin films were grown in perpendicular to the substrate. The hydrogenated IZO (IZO:H) film isolated in H2 atmosphere for 30 min exhibited an average optical transmittance higher than 85% and low electrical resistivity of less than 2.7 × 10?3 Ω·cm. These values are comparable with those of commercially available ITO. Each of the IZO films was used as an anode contact to fabricate organic light-emitting diodes (OLEDs) and the device performances studied. At the current density of 1 × 103 A/m2, the OLEDs with IZO:H (H2) anode show excellent efficiency (11 V drive voltage) and a good brightness (8000 cd/m2) of the light emitted from the devices, which are as good as the control device built on a commercial ITO anode.

Park, Pil-Man,Jeong, Myeung-Il,Yae, Byeong-Woo,Kim, Mi-Sun,Lee, Young-Ran,Park, Pue-Hee,Yoo, Bong-Sik Korean Society of Horticultural Science 2012 원예과학기술지 Vol.30 No.3
A new grafted cactus cultivar, called 'Hwangseon', Gymnocalycium mihanovichii, is a hybrid crossed between two orange color cultivars 'Geoseong' and 'Huhwang'. Mating was performed in the greenhouse of National Institute of Horticultural & Herbal Science (NIHHS) in 2005. The harvested seeds were sown at 100 mL volumetric flasks filled with 'Kyoto' medium. Seedling had been grafted into a young rootstock, Hylocereus trigonus, and cultured in vitro in 100 mL test tube for more than six months. After re-grafting and planting in the green house in 2006, their characteristics were evaluated three times until 2009. The new cultivar has a flat round shape and bright yellow skin color. In addition, it has 7 to 9 ribs with gray and straight spines on the body and produces an average of 12.3 offsets.
