Relationship between the gut microbiota and bile acid composition in the ileal mucosa of Crohn’s disease

        Shigeki Bamba,Osamu Inatomi,Atsushi Nishida,Masashi Ohno,Takayuki Imai,Kenichiro Takahashi,Yuji Naito,Junichi Iwamoto,Akira Honda,Naohiro Inohara,Akira Andoh 대한장연구학회 2022 Intestinal Research Vol.20 No.3

Background/Aims: Crosstalk between the gut microbiota and bile acid plays an important role in the pathogenesis of gastrointestinal disorders. We investigated the relationship between microbial structure and bile acid metabolism in the ileal mucosa of Crohn’s disease (CD).Methods: Twelve non-CD controls and 38 CD patients in clinical remission were enrolled. Samples were collected from the distal ileum under balloon-assisted enteroscopy. Bile acid composition was analyzed by liquid chromatography-mass spectrometry. The gut microbiota was analyzed by 16S rRNA gene sequencing. Results: The Shannon evenness index was significantly lower in endoscopically active lesions than in non-CD controls. β-Diversity, evaluated by the UniFrac metric, revealed a significant difference between the active lesions and non-CD controls (<i>P</i>=0.039). The relative abundance of <i>Escherichia</i> was significantly higher and that of <i>Faecalibacterium</i> and <i>Roseburia</i> was significantly lower in CD samples than in non-CD controls. The increased abundance of <i>Escherichia</i> was more prominent in active lesions than in inactive lesions. The proportion of conjugated bile acids was significantly higher in CD patients than in non-CD controls, but there was no difference in the proportion of primary or secondary bile acids. The genera <i>Escherichia</i> and <i>Lactobacillus</i> were positively correlated with the proportion of conjugated bile acids. On the other hand, <i>Roseburia</i>, <i>Intestinibacter</i>, and <i>Faecalibacterium</i> were negatively correlated with the proportion of conjugated bile acids. Conclusions: Mucosa-associated dysbiosis and the alteration of bile acid composition were identified in the ileum of CD patients. These may play a role in the pathophysiology of ileal lesions in CD patients.

RESTRICTION BY MOTHERWORT (Leonurus sibiricus L) OF LACTATION SUPPRESSED BY PREGNANCY-DEPENDENT MAMMARY TUMORS IN GR/A MICE

        Nagasawa, H.,Suzuki, M.,Inatomi, H.,Hibino, A.,Yamamuro, Y.,Sensui, N. Asian Australasian Association of Animal Productio 1991 Animal Bioscience Vol.4 No.1

As a possible step to evaluate in the mammary gland the role of motherwort (Leonurus sibiricus L), a representative medicinal plant used traditionally for the therapy of gynecologic diseases, the effects of the agent on lactation which was suppressed by preganacy-dependent mammary tumors (PDMT) were studied in GR/A mice. Beginning the day of placing with males at 45-50 days of age, female mice were given 60% methanol-extract of the aerial part of motherwort as drinking water at the concentration of 0.5% throughout the experiment. Mice developing PDMT during pregnancy [PDMT(+)] and given motherwort were similar to mice developing no PDMT [PDMT(_)] with or without motherwort treatment and were significantly higher than PDMT(+) mice given tap water in litter growth and mammary RNA/DMA ratio on day 12 of the 2nd lactation. Mammary DNA and RNA contents were also elevated by motherwort in PDMT(+) mice. The results suggest that motherwort can ameliorate lactation suppressed by PDMT through its stimulation of both growth and function of the mammary glands.

Prototype single-balloon enteroscopy with passive bending and high force transmission improves depth of insertion in the small intestine

( Yasuhiro Morita ),( Shigeki Bamba ),( Osamu Inatomi ),( Kenichiro Takahashi ),( Takayuki Imai ),( Masaki Murata ),( Masashi Ohno ),( Masaya Sasaki ),( Tomoyuki Tsujikawa ),( Akira Andoh ) 대한장연구학회 2020 Intestinal Research Vol.18 No.2

Background/Aims: We retrospectively analyzed Crohn’s disease (CD) patients with small intestinal strictures who underwent single-balloon enteroscopy (SBE) to ascertain whether prototype SBEs with a passive bending mechanism and high force transmission insertion tube had better insertability in the small intestine than a conventional SBE. Methods: Among 253 CD patients who underwent SBE, we identified 94 CD patients who had undergone attempted endoscopic balloon dilatation (EBD) for small intestinal stenosis for inclusion in this study. We analyzed whether the type of scope used for their initial procedure affected the cumulative surgery-free rate. For the insertability analysis, patients who underwent SBE at least twice were divided into 3 groups according to the type of scope used: conventional SBE only, prototype SBE only, and both conventional and prototype SBEs. For each group, depth of insertion, procedure time, and number of EBDs were compared in the same patient at different time points. Results: The success rate of EBD was 88.3%. The 5- and 10-year cumulative surgery-free rate was 75.7% and 72.8%, respectively. Cox regression analysis indicated that the factors contributing to surgery were long stricture (≥2 cm), EBD failure, and elevated Crohn’s Disease Activity Index, but not the type of scope used for EBD. The prototype SBEs significantly improved the depth of insertion (P=0.03, Wilcoxon’s signed-rank test). Conclusions: In CD patients with small intestinal stenosis, the prototype SBEs with a passive bending mechanism and high force transmission insertion tube did not improve long-term EBD outcome but did improve deep insertability. EFFECTS OF CHRONIC INGESTION OF ANTHRANILIC ACID ON MAMMARY GLAND GROWTH IN SHN MICE

        Nagasawa, H.,Konishi, R.,Sakagami, N.,Inatomi, H. Asian Australasian Association of Animal Productio 1988 Animal Bioscience Vol.1 No.3

Effects of anthranilic acid on normal mammary gland growth were examined in SHN/Mei female virgin mice. Anthranilic acid was given to the experimental groups as drinking water at the concentrations of 0.01, 0.02 or 0.04% for 21 days beginning 2-3 months of age. The control group received tap water only. RNA content and RNA/DNA ratio in mammary glands were significantly higher in mice given 0.04% anthranilic acid than in the control, while not mammary DNA content. The results indicate that chronic ingestion of anthranilic acid can induce an enhancement of proliferation and differentiation of mammary cells.

EFFECTS OF CHRONIC INGESTION OF ANTHRANILIC ACID ON LACTATION IN MICE

        Nagasawa, H.,Suzuki, M.,Sakagami, N.,Inatomi, H.,Yamamoto, K. Asian Australasian Association of Animal Productio 1989 Animal Bioscience Vol.2 No.1

Treatment of mice with 0.04% anthranilic acid (AnA) as drinking water resulted in an apparent stimulation of pup's growth and food intake of mothers in their first lactations associated with an increased rearing rate and no alteration in plasma prolactin level. AnA showed no significant effects on the day of vaginal opening, the pattern of estrous cycles, plasma growth hormone level and endocrine organ weights of the female and male offspring. The growth of male offspring was significantly retarded by AnA, however, their reproductivity was quite normal. The results indicate the AnA can simulate lactation of mice through little modulation of endocrine systems.

Genetically Independent Tetranucleotide to Hexanucleotide Core Motif SSR Markers for Identifying Lentinula edodes Cultivars

        ( Teruaki Saito ),( Genki Sakuta ),( Hitoshi Kobayashi ),( Kenji Ouchi ),( Satoshi Inatomi ) 한국균학회 2019 Mycobiology Vol.47 No.4

For the purpose of protecting the rights of Lentinula edodes breeders, we developed a new simple sequence repeat (SSR) marker set consisting only of genetically independent tetranucleotide or longer core motifs. Using available genome sequences for five L. edodes strains, we designed primers for 13 SSR markers that amplified polymorphic sequences in 20 L. edodes cultivars. We evaluated the independence of every possible marker pair based on genotype data. Consequently, eight genetically independent markers were selected. The polymorphic information content values of the markers ranged from 0.269 to 0.764, with an average of 0.409. The markers could distinguish among 20 L. edodes cultivars and produced highly repeatable and reproducible results. The markers developed in this study will enable the precise identification of L. edodes cultivars, and may be useful for protecting breeders’ rights.

Magnified single-balloon enteroscopy in the diagnosis of intestinal follicular lymphoma: a case series

( Kenichiro Takahashi ),( Shigeki Bamba ),( Masahiro Kawahara ),( Atsushi Nishida ),( Osamu Inatomi ),( Masaya Sasaki ),( Tomoyuki Tsujikawa ),( Ryoji Kushima ),( Mitsushige Sugimoto ),( Katsuyuki Kit 대한장연구학회 2018 Intestinal Research Vol.16 No.4

The objective of this study was to evaluate the magnified endoscopic findings in the diagnosis of follicular lymphoma in the small intestine in comparison with those of intestinal follicular lymphoma and lymphangiectasia. Four patients with follicular lymphoma and 3 with lymphangiectasia in the small intestine were retrospectively analyzed. A prototype magnifying single-balloon enteroscope was used. The findings of the intestinal follicular lymphoma and lymphangiectasia were retrospectively analyzed to determine the magnified endoscopic findings of follicular lymphoma in the small intestine. Opaque white granules were observed in 3 of the 4 patients with follicular lymphoma. Magnified narrow-band imaging (NBI) of the opaque white granules showed stretched microvessels, which had a diminutive tree-like appearance. The remaining patient had no opaque white granules and only displayed whitish villi. Magnified NBI observation of the whitish villi revealed the absence of marginal villus epithelium, which was confirmed by histology. The magnified NBI enteroscopy revealed the diminutive tree-like appearance on the opaque white granules and the absence of marginal villus epithelium of the whitish villi in intestinal follicular lymphoma. These findings may be useful in diagnosing follicular lymphoma. (Intest Res 2018;16:628-634)

      • Study of Trehalase during Autolysis of the Fruit-body in Pleurotus sp.

        Alireza Arastoo,Masami Nakazawa,Tatsuji Sakamoto,Mizuho Kusuda,Hitoshi Kobayashi,Kenji Ouchi,Satoshi Inatomi,Mitsuhiro Ueda 한국버섯학회 2017 버섯 Vol.21 No.2

Fruiting bodies were degraded themselves by the several glycoside hydrolases after spore releasing from mature fruiting bodies or harvesting. The enzymes involved in autolysis such as glucanase and chitinase have been studied. However, there are almost no information about the relationship between several glycoside hydrolases and autolysis. In this study, we studied to obtain the enzymatic properties of trehalase, and also to get the new information on the relationship between trehalase and autolysis. Crude enzymes were prepared from each fruiting body of Pleurotus sp. (from the immature stage to the autolysis stage) and the trehalase activities were measured at each growth stage. Trehalase activities sharply increased in autolysis stage. Trehalase was partially purified from fruiting bodies of the autolysis stage using various column chromatography and its properties were examined. The optimum temperature was 50 °C and the optimum pH was 4.5. In order to elucidate the localization of trehalase, fruit bodies of the autolysis stage were divided into the stipes and the pileuses, and the each trehalase activity was measured. High trehalase activities were found in the pileuses. Furthermore, in order to elucidate trehalase activities in autolysis more detail, the each fruiting body of autolysis progressing stages was finely divided into the stipes and the pileuses, and their activities were measured. The activities in the outer part of the pileuses were highest at the initial stage of autolysis and the activities shifted from the outer side to the inner side of the pileuses according to the progress of autolysis.
