        MUC16 facilitates cervical cancer progression via JAK2/STAT3 phosphorylation‑mediated cyclooxygenase‑2 expression

        Objectives MUC16 (mucin 16, also known as CA-125, cancer antigen 125, carcinoma antigen 125, or carbohydrate antigen 125) has been predicted as tumor biomarker for therapy. We determined to investigate effects and regulatory mechanism of MUC16 on cervical tumorigenesis. Methods Expression levels of MUC16 in cervical cancer cell lines was analyzed via qRT-PCR (quantitative real-time polymerase chain reaction). Knockdown of MUC16 was conducted via shRNA (Short hairpin RNA) transfection. MTT and colony formation assays were used to investigate effect of MUC16 on cell proliferation. Wound healing assay was utilized to detect migration and transwell assay to detect invasion. The underlying mechanism was demonstrated via western blot analysis. Results MUC16 was elevated in cervical cancer cell lines. MUC16 knockdown inhibited cell proliferation, invasion and migration. Gain- and loss-of functional assays revealed that over-expression of MUC16 activated Janus Kinase 2 (JAK2)/ signal transducer and activator of transcription 3 (STAT3) via phosphorylation, thus facilitating cyclooxygenase-2 (COX-2) expression, while knockdown of MUC16 demonstrated the reverse effect on JAK2/STAT3 activation and COX-2 expression. Moreover, inhibition of JAK2/STAT3 attenuated the regulation of MUC16 on COX-2. Conclusions MUC16 enhanced proliferation and invasion of cervical cancer cells via JAK2/STAT3 phosphorylation-mediated cyclooxygenase-2 expression, suggesting the potential therapeutic target ability of MUC16 to treat cervical cancer.

        Rheological kinetics of thermo-sensitive supramolecular assemblies from poly(N-isopropyl acrylamide) and adenine-functionalized poly(ethylene oxide) stabilized by complementary multiple hydrogen bonds

        In this study, we synthesized a poly(N-isopropylacrylamide) (PNIPAm) through the polymerization of N-isopropylacrylamidein distilled water with azodiisobutyronitrile as the initiator and a bisadenine-functionalizedpoly(ethylene oxide) (A-PEO-A) from the reaction of adenine with a difunctionalized toluenesulfonyl-PEO. When blended together in distilled water, PNIPAm and A-PEO-A formed supramolecular aggregates stabilizedthrough complementary multiple hydrogen bonds between the amide groups of PNIPAm and the adenineunits of A-PEO-A. Agrawal integral equation and rheometry revealed the rheological kinetics ofsupramolecular assemblies, which were influenced significantly by the spherical micelles, large associatedaggregates of spherical micelles, network structures, and toroid structures formed in aqueous solutions.

        Using Nano Hexagonal Boron Nitride Particles and Nano Cubic Silicon Carbide Particles to Improve the Thermal Conductivity of Electrically Conductive Adhesives

        To satisfy the high electrical and thermal conductivity required for the continuous development of electronic products, nano hexagonal boron nitride (BN) particles and nano cubic silicon carbide (SiC) particles were added into electrically conductive adhesives (ECAs) to improve the thermal conductivity. BN and SiC had little negative effect on the electrical conductivity, but improved the thermal conductivity significantly. When their content was 1.5 wt. %, the thermal conductivity at 100°C, 150°C and 200°C was increased by 71%(100°C), 78% (150°C) and 70% (200°C), and 114% (100°C), 110% (150°C) and 98% (200°C) respectively for BN and SiC comparing with those of the ECAs with no thermal conductive fillers. This method is simple,easy to do, and can be used practically in electronic packaging.

        Using Polyurethane, Ethylene-Vinyl Acetate Hotmelt, and Nano Hexagonal Boron Nitride Particles to Electrospin High Surface Adhesion Polymer Fibers

        In this study, we presented the use of polyurethane, ethylene-vinyl acetate hotmelt, and nano hexagonal boron nitride particles to prepare high surface adhesion polymer fibers via an elertrospinning method. The shear strength, dynamic tensile properties, and surface morphology have been investigated. These polymer fibers were found to have high shear strength, high tensile stress, and high tensile strain, which may have a good potential application in the matrix materials for thermal interface materials. Polymer fibers with and without nano hexagonal boron nitride particles showed the shear strength of 6.52 MPa and 5.44 MPa respectively on being heated up at 150°C for 45 min.

        Using a Functional Epoxy, Micron Silver Flakes, Nano Silver Spheres, and Treated Single-Wall Carbon Nanotubes to Prepare High Performance Electrically Conductive Adhesives

        In this study, a matrix resin containing a functional epoxy, a reactive diluent, a silane-coupling agent, and a curing agent was used to fabricate three modal electrically conductive adhesives (ECAs) with micron silver flakes, nano silver spheres, and treated single-wall carbon nanotubes (CNT). Results showed that too many micron silver flakes reduced the bulk resistivity and adhesion strength of uni-modal ECAs (matrix resin and micron silver flakes). As the nano silver spheres increased, the bulk resistivity of bi-modal ECAs (matrix resin, micron silver flakes, and nano silver spheres) firstly decreased, and then increased again. The adhesion strength decreased also. The bulk resistivity and adhesion strength of tri-modal ECAs (matrix resin, micron silver flakes, nano silver spheres, and treated CNT) both were reduced by the treated CNT greatly. These ECAs could be cured at 120°C or any higher temperature than this with different curing time. They all had high temperature stability with a pyrolysis temperature above 350°C and a glass transition temperature around 180°C.

        Uterine Intravenous Leiomyomatosis with Intracardiac Extension and Pulmonary Benign Metastases on FDG PET/CT: A Case Report

        A 48-year-old woman presented with a 50-day history of irregular vaginal bleeding and lower abdominal pain. Ultrasound indicated an extremely large occupying lesion in the pelvic cavity that was highly suggestive of malignancy. Fluorodeoxyglucose (FDG) positron emission tomography/computed tomography (PET/CT) was performed to further assess the nature of pelvic abnormality. PET/CT images demonstrated a diffusely lobulated mass ranging from cervix up to the inferior pole of kidneys with mild FDG uptake. Simultaneously, multiple nodules in bilateral lungs and a hypodense lesion in the right ventricle were shown without FDG-avidity. Based on the imaging results, the presumptive diagnosis was uterine intravenous leiomyomatosis with intracardiac extension and pulmonary benign metastases, which was subsequently confirmed by MRI and the lesion biopsy.

        A FACILE SYNTHESIS OF SHAPE- AND SIZE-CONTROLLED α-Fe_2O_3 NANOPARTICLES THROUGH HYDROTHERMAL METHOD

        α-Fe_2O_3 nanoparticles have wide-ranging applications such as in catalysis, sensoring, painting, etc. This is the reason to study their controlled synthesis. Here we have investigated the synthesis of uniform α-Fe_2O_3nanoparticles using amino acids as morphology control agents. The products were characterized by transmission electron microscopy (TEM), X-ray diffraction (XRD), thermogravimetry (TG) and differential thermal analysis (DTA). It was found that the type and the amount of amino acids as well as the reaction temperatures have significant influence on the shape and size of the obtainedα-Fe_2O_3nanoparticles. The use of acidic amino acids (always contain C=O in the side chain) typically leads to the formation of α-Fe_2O_3nanoparticles with spindle shape. However, rhombohedrally shaped α-Fe_2O_3nanoparticles were formed in presence of basic amino acids (always contain -NH_2 in the side chain). Increasing the amount of amino acid generally results in α-Fe_2O_3nanoparticles with decreasing particle sizes.

        Gonadotropins Improve Porcine Oocyte Maturation and Embryo Development through Regulation of Maternal Gene Expression

        The present study assessed the effect of FSH and LH on oocyte meiotic, cytoplasmic maturation and on the expression level and polyadenylation status of several maternal genes. Cumulus-oocyte complexes were cultured in the presence of FSH, LH, or the combination of FSH and LH. Significant cumulus expansion and nuclear maturation was observed upon exposure to FSH alone and to the combination of FSH and LH. The combination of FSH and LH during entire IVM increased the mRNA level of four maternal genes, C-mos, Cyclin B1, Gdf9 and Bmp15, at 28 h. Supplemented with FSH or LH significantly enhanced the polyadenylation of Gdf9 and Bmp15; and altered the expression level of Gdf9 and Bmp15. Following parthenogenesis, the exposure of oocytes to combination of FSH and LH during IVM significantly increased cleavage rate, blastocyst formation rate and total cell number, and decreased apoptosis. In addition, FSH and LH down-regulated the autophagy gene Atg6 and upregulated the apoptosis gene Bcl-xL at the mRNA level in blastocysts. These data suggest that the FSH and LH enhance meiotic and cytoplasmic maturation, possibly through the regulation of maternal gene expression and polyadenylation. Overall, we show here that FSH and LH inhibit apoptosis and autophagy and improve parthenogenetic embryo competence and development.

      • Integration Sites and Genotype Distributions of Human Papillomavirus in Cervical Intraepithelial Neoplasia

        Objectives: To analyse HPV integration prevalence and genotype distributions in cervical intraepithelial neoplasia (CIN) in east part of China, furthermore to assess preferential sites for common HPV integrations and provide baseline information for cervical abnormality screening and prevention. Methods: Integration of HPV in 113 paraffin-embedded cervical intraepithelial neoplasia samples was assessed using Gencap technology in Key Laboratory of Biotechnologies in BGI-Shenzhen. Results: 64 samples were HPV-integrated and as the cervical lesions increased, the integration rate became higher significantly (P=0.002). Fifteen different HPV genotypes were detected, 14 high-risk (16, 18, 31, 33, 51, 52, 56, 58, 66, 68) and 1 low-risk (11). The most common genotypes were HPV-16, 58, 33, 52, 66, and 56. Thirteen patients had co-integration involving mainly HPV-16 and 58. The frequency of HPV gene disruption was higher in L1 and E1 genes than in other regions of the viral genomes. Conclusion: Some 56.6% of CIN lesions in Qingdao had HPV integrations, and 67.2% of HPV-integrated patients were HPV-16 and 58, more prone to be integrated in younger patients below 45 years old. There exist preferential sites for HPV-16 and HPV-58 integration, and they are more likely to be disrupted in the L1 and E1 loci.
