        Feeding and related morphology of salivary glands of Bradysia odoriphaga (Diptera: Sciaridae)

        The amount of Bradysia odoriphaga larval feeding at each developmental stage and the related morphology of salivary glands were investigated in this study. The amount of daily feeding in the third instar was greatest among all the developmental stageswhile the amount of cumulative feeding in the fourth-instar accounted for 53.56% of the total amount of larval feeding. The daily amount of feeding by B. odoriphaga during the whole larval stage peaked in the 8th and 14th day after hatching. The paired and transparent salivary glands of B. odoriphaga were characterized by two distinct regions: an anterior region and a posterior region. At the fourth-instar and pupal stage, the length of salivary glands decreased significantly compared with that of the third-instar, and the integrity of epithelium cytoplasm in the salivary glands also decreased markedly because of the formation of cytoplasmic vacuoles and degradation of rough endoplasmic reticulum and mitochondria. We concluded that the decrease of daily feeding amount in the last instar was related to the structural change of salivary gland closely. The function of salivary glands in B. odoriphaga was mainly secretion of silk and cocooning-like materials during the last larval instar.

        Impact of selected fungi from an artificial diet on the growth and development of Drosophila suzukii (Diptera: Drosophilidae)

        Drosophila suzukii (Diptera: Drosophilidae) is one of the very few Drosophila species which are able to lay eggs and feed on healthy ripening fruit. Adults and larvae are all able to obtain the nutrition in the decaying food, which will generate many microorganisms. However, the relationship between D. suzukii and microorganism is poorly understood. In this study, 13 species of fungi were identified in an artificial diet fed by D. suzukii. Short-term and continuously life table experiments were conducted to determine the impact of three of these fungus species including Geotrichum candidum, Talaromyces minioluteus and Actinomucor elegans on the growth and development of Drosophila suzukii. Results revealed that, compared to the control, G. candidum, T.minioluteus and A. elegans increased the mortality of D. suzukii adults in the short time, while extended the developmental time of pupal D. suzukii by 18.00%, 16.22% and 26.44%, respectively in the life table experiment. T. minioluteus reduced the total longevity of D. suzukii by 15.52%, while A. elegans enhanced the total longevity by 25.96%. G. candidum and A. elegans increased the fecundity bymore than two folds. T. minioluteus elongated the mean generation time (T) by 31.34%, whereas G. candidum shortened it by 15.26% but increased the net reproductive rate (R0) by 217.76%, intrinsic rate of increase (r) by 88.89% and finite rate of increase (λ) by 9.17%. It was concluded that G. candidumand A. elegans significantly had the beneficial effect on the growth and reproduction of D. suzukii, however, T. minioluteus had the negative effect on D. suzukii. Our results could provide a new integrated pest management strategy for D. suzukii which would be discussed in this study.

      • Bevacizumab Regulates Cancer Cell Migration by Activation of STAT3

        There are numerous clinical cases indicating that long-term use of bevacizumab may increase the invasiveness of tumors. However, to date, little is known about underlying molecular mechanisms. Therefore, the purpose of our study was to investigate effects of bevacizumab in four cancer cells lines (WSU-HN6, CAL27, Tca83, and HeLa). It was found to promote migration and invasion in the WSU-HN6 and Tca83 cases, while exerting inhibitory effects in CAL27 and HeLa cells. The signal transducer and activator of transcription (STAT) 3 inhibitors niclosamide and S3I-201 inhibited the STAT3 signal pathway, which is activated by bevacizumab. These inhibitors also substantially blocked bevacizumab-induced migration of WSU-HN6 and Tca83 cells. Bevacizumab upregulated interleukin (IL)-6 and phosphorylated (p)-STAT3 expression time-dependently. Therefore, we propose that bevacizumab has differential effects on the migration of different cancer cell lines and promotes migration via the IL-6/STAT3 signaling pathway.

        Construction of a New Agrobacterium tumefaciens-Mediated Transformation System based on a Dual Auxotrophic Approach in Cordyceps militaris

        Cordyceps militaris is a significant edible fungus that produces a variety of bioactive compounds. We have previously established a uridine/uracil auxotrophic mutant and a corresponding Agrobacterium tumefaciens-mediated transformation (ATMT) system for genetic characterization in C. militaris using pyrG as a screening marker. In this study, we constructed an ATMT system based on a dual pyrG and hisB auxotrophic mutant of C. militaris. Using the uridine/uracil auxotrophic mutant as the background and pyrG as a selection marker, the hisB gene encoding imidazole glycerophosphate dehydratase, required for histidine biosynthesis, was knocked out by homologous recombination to construct a histidine auxotrophic C. militaris mutant. Then, pyrG in the histidine auxotrophic mutant was deleted to construct a ΔpyrG ΔhisB dual auxotrophic mutant. Further, we established an ATMT transformation system based on the dual auxotrophic C. militaris by using GFP and DsRed as reporter genes. Finally, to demonstrate the application of this dual transformation system for studies of gene function, knock out and complementation of the photoreceptor gene CmWC-1 in the dual auxotrophic C. militaris were performed. The newly constructed ATMT system with histidine and uridine/uracil auxotrophic markers provides a promising tool for genetic modifications in the medicinal fungus C. militaris.

        Preparation and molecular characterization of a polyclonal antibody as an efficient cutworm reference protein

        Antibodies, which are wildly used in molecular researches, are proteins secreted by plasma cells that can specifically recognize specific antigens. To obtain a stable reference antibodies are important for protein analysis. In this study, a capable glyceraldehyde phosphate dehydrogenase (GAPDH) antiserum against most noctuid larval protein samples was prepared. Firstly, the full-length gapdh was amplified from Spodoptera exigua larvae to construct the prokaryotic expression vector. The purified His-tag fused GAPDH protein was used to immunize rabbits for the antiserum preparation. Protein samples extracted from 11 insect species distributed across seven families and three orders were used to perform the Western bolt analysis. The bright specific bands were detected in S. exigua, S. litura, Helicoverpa armigera, and Mythimna seperata protein samples, indicating that this antiserum may be capable of detecting noctuid larval encoded GAPDH. Further immunofluorescence identification of H. armigera and S. exigua paraffin section slides with GAPDH antiserum exhibited an identical cytostained image. The GAPDH antiserum prepared in this study is useful and efficient as reference antibodies in studies involving noctuid larval protein samples in other related fields.

        Analysis of the Interdecadal and Interannual Variability of Autumn Extreme Rainfall in Taiwan Using a Deep-Learning-Based Weather Typing Approach

        This study sought to assess the interdecadal and interannual variability of autumn extreme rainfall (ER) in Taiwan from 1979to 2019. Three types of ER events were identified based on a clustering analysis augmented by a deep autoencoder-basedneural network model. This method outperforms other methods in obtaining the optimal number of clusters by extractingthe synoptic features in advance. The patterns associated with these three types include a tropical cyclone covering Taiwan(TC), a TC-like circulation in the South China Sea (SCS) accompanied by northeasterly near northern Taiwan (TC-NE), andnortheasterly near northern Taiwan (NE). The differences in the rainfall pattern caused by the three types were discernableover Taiwan. How the PDO or ENSO modulates the regional large-scale environment to favor the occurrence of these ERevents was investigated. The occurrence of TC-NE events was simultaneously correlated with the negative phases of PDO/ENSO in the interdecadal/interannual scale. In the negative phases of PDO/ENSO, a low-level anomalous cyclone overSCS accompanied by background northeasterly favored the regional TC activities and may cause more TC-NE events. Theoccurrence of NE events is simultaneously correlated with the cold phase of ENSO. An anomalous low-level anticyclone inNortheast Asia strengthened the northeasterly toward northern Taiwan, and with the seasonal background moisture, providedfavorable conditions for the occurrence of the NE events. Overall, the occurrence of the TC events did not correlate with thePDO or ENSO signals; the reasons for the lack of correlation were discussed herein.

        필사본 중국어 학습서 《니니귀성(你呢貴姓)》문헌 연구

        이 글의 목적은 조선 후기 필사본 중국어 학습서 《你呢貴姓》의 자료적 가치를 발굴해 내는 데 있다. 이를 위해 먼저 책의 체재에 대한 면밀한 분석을 통해 구성상의 특징을 확인해야 했고 또 회화 내용 분석을 통해 필사 시기를 확정해야만 했다. 그리고 표기에 대한 종합적인 고찰이 필요했다. 이러한 기본적인 논의 없이 《你呢貴姓》이 갖는 자료의 가치를 논하는 것은 불가능하다고 판단되었기 때문이다. 본 연구의 주요 성과들을 제시하면 아래와 같다. 먼저 구성 체재 분석으로부터 《你呢貴姓》은 《華語類抄(목판본)》을 참고했을 가능성이 크다는 사실을 확인할 수 있었다. 또 동시대의 중국어 학습서들의 회화의 내용을 살펴본 바로는 《學清》외에 내용상의 공통점을 갖는 학습서를 찾아볼 수 없었다. 따라서 현재까지 밝혀지지 않은 제3의 模本이 존재한다고 보는 것보다 이본으로 알려진 필사본 《學清》이 《你呢貴姓》의 模本일 가능성이 있다고 주장하였다. 회화 본문에 등장하는 두 인물의 대화 내용을 통해 그 필사연대를 1903년 정도로 추정할 수 있었다. 그리고 《你呢貴姓》에서 나타나는 ‘你呢’ 표기는 ‘您[nín]’의 反切표기가 아닌 이인칭 대명사에 어기조사 ‘呢’가 결합한 것일 뿐, 어기조사 ‘呢’의 기능은 현대 중국어의 것과 다름이 없음을 주장했다. The purpose of this paper is to discover the value of the manuscript Chinese study book NiNiGuiSeong(你呢貴姓), that was personal compiled in the late Joseon Dynasty. In order to accomplish it, first, it was necessary to check the compositional characteristics through a detailed analysis of the format, and also to determine the compilation period through the analysis of the conversational content. And a comprehensive review of the notation was necessary. Because it was considered impossible to discuss the value of the material of NiNiGuiSeong without such a basic discussion. The main achievements of this paper are presented below. First, from the analysis of the basic compositional format, it could be confirmed that NiNiGuiSeong most likely referred to HwaEoYuCho(華語類抄). Also, looking at the contents of book, we could not find any textbooks that had anything in common other than HakCheong(學清). Therefore, it was argued that there would be a possibility that the manuscript HakCheong known to be a different version of NiNiGuiSeong is more likely than that a third master copy version that has not been revealed until now exists. From the conversation between the two characters appearing in the main text, it is possible to estimate the time of the transcription as 1903. And it was confirmed that ‘NiNi(你呢)’ appearing in NiNiGuiSeong is simply the use of the word ‘Ni(呢)’, not the ‘FanQie(反切)’ form of ‘您[nín]’. Hence, it was argued that the function was no different from that of Modern-Chinese.

        A practical method for simultaneous determination of Poisson’s ratio and Young’s modulus of elasticity of thin films

        In this paper, based on the exact analytical solution of axisymmetric deformation of the circular membrane fixed at its edge under the action of uniformly-distributed loads, we propose a new method to be able to simultaneously determine Poisson's ratio and Young's modulus of elasticity for thin films. We also present a set of exact formulas used for simultaneously determining Poisson's ratio and Young's modulus of elasticity for free-standing thin films or coating thin films without residual stresses or pre-tension. This method has the advantages of simplicity and reliability, and it doesn’t involve the substrate effect.

        Desulfurization of Model Oil via Adsorption by Copper(II) Modified Bentonite

        In order to further reduce the sulfur content in liquid hydrocarbon fuels, a desulfurization process by adsorption for removing dimethyl sulfide (DMS) and propylmercaptan (PM) was investigated. Bentonite adsorbents modified by $CuCl_2$ for the desulfurization of model oil was investigated. The results indicated that the modified bentonite adsorbents were effective for adsorption of DMS and PM. The bentonite adsorbents were characterized by X-ray diffraction (XRD) and thermal analysis (TGA). The acidity was measured by FT-IR spectroscopy. Several factors that influence the desulfurization capability, including loading and calcination temperature, were studied. The maximum sulfur adsorption capacity was obtained at a Cu(II) loading of 15 wt %, and the optimum calcination temperature was $150^{\circ}C$. Spectral shifts of the ${\nu}$(C-S) and ${\nu}$(Cu-S) vibrations of the complex compound obtained by the reaction of $CuCl_2$ and DMS were measured with the Raman spectrum. On the basis of complex adsorption reaction and hybrid orbital theory, the adsorption on modified bentonite occurred via multilayer intermolecular forces and S-M (${\sigma}$) bonds.

      • Study the Thermal Behaviours for Asphalt Concrete Mixed with Basic Oxygen Furnace Slags

        In Taiwan, the basic oxygen furnace (BOF) slags are characterized as porous material and with better heat insulation than that of the natural aggregates. The BOF slags have been applied to the pavement as parts of the natural aggregate replacement. The asphalt concrete (AC) with the BOF slags replacements were well recognized and performed. In this study, parts of natural aggregates were replaced by the BOF slags in the AC helping resolve the gradual shortage of the natural resources and save energy and reduce the produce of carbon. In this study, parts of the natural aggregates were replaced by the coarse and fine BOF slags in the construction of AC. Because the thermal insulation and heat absorption are different between BOF slags and natural aggregates, the infrared thermal conductivity measuring instrument was applied to obtain the surface temperatures presented as thermal infrared images in AC. Then, the infrared temperature distribution images were obtained by using the image transformation techniques. The results show that the thermal conductivity for the AC containing with fine BOF slag was larger than that for the coarse BOF slag. The BOG slag replacement in AC helped improve the thermal insulation in AC. Moreover, the temperature was more uniformly distributed for AC with the fine BOF slag than that for the coarse BOF slag replacement. However, the coarse BOF slag was better in the preserve of energy for AC than that of the fine BOF slag.
