        Porphyromonas endodontalis의 침투에 따른 혈관 내피세포의 유전자 발현

        During the last two decades, there has been an increasing interest in the impact of oral health on atherosclerosis and subsequent cardiovascular disease (CVD). To date, some periodontal pathogens including Porphyromonas gingivalis (P. gingivalis) have been reported to be relevant to CVD. Porphyromonas endodontalis (P. endodontalis), which shares approximately 87% sequence homology with P. gingivalis, is mostly found within infected root canals. However, recent studies reveal that this pathogen also resides in the dental plaque or periodontal pocket in patients with periodontitis. It has been shown that P. endodontalis invades human coronary artery endothelial cells (HCAEC) and coronary artery smooth muscle cells (CASMC). To evaluate whether P. endodontalis can participate in the progression of atherosclerosis and CVD, we examined the changes in transcriptional gene expression profiles of HCAEC responding to invaion by P. endodontalis in this study. The following results were obtained. 1. Porphyromonas endodontalis was invasive of HCAEC. 2. According to the microarray analysis, there were 625 genes upregulated more than two-folds, while there were 154 genes downregulated by half. 3. Upregulated genes were relevant to inflammatory cytokines, apoptosis, coagulation and immune response. Enhanced expression of MMP-1 was also noticeable. 4. The transcription profiles of the 10 selected genes examined by real-time PCR agreed well with those observed in the microarray analysis. Thus, these results show that P. endodontalis presents the potential to trigger and augment atherosclerosis leading to CVD. 본 연구에서는 Porphyromonas endodontalis와 죽상경화증 및 심혈관질환과의 관계를 알아보기 위해, P. endodontalis가 사람의 관상동맥 내피세포에 침투했을 때 나타나는 유전자 발현의 변화를 microarray와 real-time PCR로 측정하였고, 발현이 증가된 유전자 중에서 죽상경화증과 연관된 유전자들이 관련 KEGG pathway 상에서 유의성 있는 영향을 미치는지를 분석하여 다음과 같은 결과를 얻었다. 1. Porphyromonas endodontalis는 사람의 관상동맥 내피세포에 침투하였다. 2. Microarray 분석결과, 대조군보다 발현이 2배 이상 증가된 유전자는 625개였고, 1/2이하로 감소된 유전자는 154개였다. 3. 발현이 2배 이상 증가된 유전자 중에는 염증성 cytokine 및 chemokine, 세포자멸사, 혈액응고와 면역반응 관련 유전자들이 포함되었다. 4. Microarray 분석결과를 확인하기 위해 발현이 2배 이상 증가된 유전자 중에서 10개의 유전자를 선택하여 real-time PCR을 시행하였고, 그 결과 microarray에서와 마찬가지로 발현 정도가 대조군보다 모두 높게 나타났다. 따라서 P. endodontalis가 사람의 관상동맥 내피세포에 만성적으로 작용했을 때, 심혈관질환에서 중요한 부분을 차지하는 죽상경화증을 유발하는 위험 요소 중의 하나로 작용할 수 있을 것으로 판단된다. 이와 관련된 자세한 기전을 이해하기 위해서는 앞으로 더 많은 연구가 필요할 것으로 보인다.

      • 검정금파리의 第二生殖週期 誘導

        Induction of the second reproductive cycle and ovarian maturation, and changes of protein were investigated in the black blowfly, Phormia regina. A height in between hardboard and bottom of the experimental device based on the length of mouthpart and ovipositor was 2.5cm. Ovipositional rate of the first reproductive cycle in the experimental device was 81.2±6.1% without liver juice in the crop and midgut. During the second reproductive cycle terminal oocyte was 385, 977, 1121㎛ and ovary was 1732, 3819, 3946 ㎛ in length at 24, 48, and 60h after the second liver-feeding, respectively. Haemolymph protein increased 36h after the second liver-feeding and decreased thereafter while ovarian protein increased continuously until 60h in the second reproductive cycle. This result should be applicable as reference data for the embryo-physiological research of the second reproductive cycle in other Dipteran insects.

        정신재활 치료가 정신분열병 환자의 재발율, 입원횟수, 입원일수 그리고 치료 경비에 미치는 효과 : 2년 추적조사 연구

        목 적 : 사회기술훈련, 환자교육, 가족교육, 집단치료, 예술치료, 작업치료와 같은 포괄적인 정신재활 치료를 약물치료와 병행할 경우 약물치료만 시행할 때에 비해 정신분열병 환자의 재발율, 입원횟수, 입원일수, 직접 치료경비 등에 어떤 효과가 있는 지를 알아보기 위함이다. 연구방법 : 1993년 9월부터 1995년 2월까지 양산 신경정신병원의 폐쇄병동에서 퇴원한 20∼35세 사이의 정신분열병 환자 173명을 대상으로 46명에게는 6개월동안 정신재활 치료를 시행하였고 127명에게는 통상적인 외래치료만 시행한 후 연구시작 시점을 포함하여 2년동안 양군에 속한 환자들을 대상으로 재발율, 입원횟수, 입원일수, 직접 치료경비 등을 1년 단위로 추적 조사하였으며, 연구시작 시점을 기준으로 전과 후의 매년 평균 입원횟수, 입원일수, 직접 치료경비도 서로 비교하였다. 결 과 : 정신재활 치료를 약물치료와 병행할 경우 약물치료만 시행할 때에 비해 재발율, 입원횟수, 입원일수, 직접 치료비용이 현저히 감소되었다. 그리고 정신재활 치료를 받은 군에서는 치료 전에 비해 치료 후에 재발율, 입원횟수, 입원일수, 직접 치료비용이 유의하게 감소하였지만 통상적인 외래치료만 받은 군에서는 치료 전과 후간에 유의한 변화를 보이지 않았다. 결 론 : 정신분열병 환자들에게 정신재활 치료를 약물치료와 병행할 경우 약물치료만 시행할 때에 비해 환자의 임상적 경과를 호전시킬 뿐 아니라 가족의 경제적 부담도 줄일 수 있다. Objective : We evaluated the clinical efficacy of a comprehensive psychiatric rehabilitation of schizophrenia, including family psychoeducation, patient education, social skills training, group psychotherapy, art therapy, and occupational therapy. Method : From September 1993 to February 1995, 173 chronic schizophrenics who met DSM-IV criteria and were discharged from a locked ward were assigned into the psychiatric rehabilitation service group(psychiatric rehabilitation group, n=46) or the customary out-patient service group(out-patient group, n=127) prospectively, but not randomly. Assignment was based on preferences of patients and their relatives, Each subject included in the psychiatric rehabilitation group received twice weekly patient education, social skills training, family psychoeducation, group psychotherapy, art therapy, occupational therapy and maintenance chemotherapy during a 6-month period. Those in the out-patient group received individual supportive psychotherapy and maintenance chemotherapy according to their clinical needs for 10-20 minutes once per week. After entry into the study, all subjects were assessed for the number of readmission, days of hospitalization, and the direct treatment costs through the first and second year of follow-ups. These outcome variables were also compared pre-and post-2 years at the time point of entry into the study. Aftercare chemotherapy on a weekly to monthly basis continued for the remainder of the two year follow-up period. Results : With regard to the mean number of hospital admissions per year, the mean days of hospitalization per year, and the direct treatment costs per year, the psychiatric rehabilitation group had significantly decreased all clinical outcomes 2 years after compared with before receiving psychiatric rehabilitation. But there were no significant differences in the all clinical outcomes between before and after study intake in the out-patient group. For the number of relapse regardless of drug compliance, 4 patients(9%) during first year and 14 patients(30%) during second year were relapsed in the 46 subjects of the psychiatric rehabilitation group. In contrast, 75 patients(59%) during first year and 90 patients (71%) during second year relapsed in the out-patient group. When considering the effect of drug noncompliance to relapse, 4(9%) and 14 patients(30%) were relapsed while on medication respectively during the first and second year in the 46 of the psychiatric rehabilitation group, vs. 27(34%) of 79 patients on medication during the first year and 31(46%) of 68 patients on medication during the second year were relapsed in the out-patient group. Conclusions : These results indicate that the comprehensive psychiatric rehabilitation can be useful therapeutic intervention both to improve the clinical outcomes of schizophrenic patients and to reduce the economical burden of their relatives.

      • 토양환경지도 자료를 이용한 콩밭 토양의 화학성 비교

        It is important to interpret the soil chemical properties in order to effectively manage the farmland. This study was conducted to investigate the soil chemical properties of upland soybean field according to the soil series at 32 upland fields located in the parts of Gangwon-do. Soil sampling sites were selected by using the National Statistics DB and KSIS DB that is a soil environment information system provided by the Rural Development Administration. Soil samples were collected from 0~15 cm of top soil before seeding or transplanting, air-dried, passed through a 2 mm sieve, and analyzed for soil pH, EC, organic matter, available phosphorus (P2O5), exchangeable potassium, exchangeable calcium and exchangeable magnesium. The average chemical properties have exceeded the each nutrient optimum range level of RDA, except the organic matter in experimental soil 2015. And despite application of conventional fertilization, each soil series nutrient contents were change respectively. The results of this study suggest that conventional fertilization should be applied based on the analysis about the amount of accumulated nutrient contents in the soil.

        Recently with the spread of smart phones is increasing the importance of handwriting recognition technology. Therefore, we studied these online handwriting recognition technology for real-time input. In this research process we study the distance filtering method, the catching feature points, chain code, and similarity matching techniques and make possible the actual coded. We implemented the on-line symbol recognition method which recognize up to 91% of the capital letter. That you can use handwriting recognition to target programs, the children learn to play program planning and production.

      • Comparative nutritional analysis for marker-free transgenic Bt rice and non-transgenic counterparts

        The selectable marker-free rice plants containing mcry1Ac insecticidal gene isolated from Bacillus thuringiensis (Bt) were generated using a non-selection approach by Agrobacterium tumefaciens-mediated transformation. The nutritional composition of two lines of transgenic rice plants (RTB5 and RTB11) was compared with that of its non-transgenic counterpart. The results showed that, except for small differences in dietary fiber and some minerals, there was no significant difference between transgenic rice and conventional counterpart variety with respect to their nutrient composition. Most of measured levels of nutrients were within the range of values reported for other commercial cultivars, showing substantial equivalency. Therefore, the insertion of transgenes did not affect the nutritional composition of transgenic RTB5 and RTB11 rice grains.

      • PCR Detection of Disease Resistant Transgenic Rice for the Biosafety Assessment

        Regulations in the EU, Japan, Korea, etc. require that foods and feeds made of or derived from genetically modified organisms (GMOs) should be approved and labeled according to a threshold. Recently, disease resistant transgenic rice was developed in Korea, which resulted from the transformation events involving choline kinase gene, OsCK1. In order to monitor unintended release of the developed GM rice in the near future, as well as to meet GM-labeling requirements, the development of a reliable method for detection of disease resistant GM rice is requisite. Here, specific primer pairs for the detection of GMO was designed on the basis of a introduced gene and the flanking junction sequences between a plant DNA and a integrated gene construct, and also SPS gene was used as an endogenous reference material. Specificities of all designed primers were tested through qualitative PCRs. Clearly, target specific amplicons could be detected from disease resistant GM rice event. In addition, the limits of detection (LOD) using the event-specific primers were approximately 0.1% for the disease resistant GM rice line. This result indicated that the developed detection method is suitable for the traceability of disease resistant GM rice, because of using the primer specifically corresponded to the junction site between plant genomic DNA and inserted DNA.       • Development of Transgenic Rice Lines Expressing Coleoptera- Resistant Genes and Insect Bioassay on Rice Water Weevil

        The rice water weevil (RWW), Lissorhoptrus oryzophilus are major pests of aquatic rice plant in Korea as well as throughout the country. Larvae of RWW sucking the nourishment on roots, causes a stunted root system and reduces grain yields. To prevent these damages, we constructed various plant expression vectors, which were harbored by insecticidal genes, cryBP1 and cryIIIa, and fused with the actin promoter and/or the modified RCg2 root-preferential promoters for expressing the insect-toxic genes in leaves and roots. A cryBP1 was cloned from Bacillus popilliae, producing crystal toxin against Japanese beetle, and CryIIIa was modified from the δ-endotoxin gene of Bacillus thuringiensis ssp. tenebrionis, encoding the coleoptera-specific toxin. The vectors containing the insecticidal genes were transferred into Oryza sativa japonica cultivar, Nakdong, by Agrobacterium -mediated transformation method. Several independent transgenic lines were selected by Southern blotting and Western blotting, confirming that cryBP1 and cryIIIa genes were stably integrated into the plant genomes and were expressed in transgenic plants. Upon insect bioassay using RWW, the mortality of insect larvae on cryBP1 and cryIIIa transgenic rice lines recorded up to 41% and 34%, respectively. These results suggested that the transgenic lines can be used to develop Coleoptera-resistant cultivars and could be valuable for later application in crop breeding for insect resistance.

      • Comparative Whole-Genome Analysis of Tall Transgenic Bt Line and wild-type Line

        Natural and artificially induced mutants have provided valuable resources for plant genetic studies and crop improvement. Some variations induced in the process of plant transformation have often been observed in regenerated plants. In this study, we investigated the insertion number of transgene and the flanking sequences of T-DNA in tall-induced line BP23, which was unexpectedly gained in the process of transformation of insect-resistant rice with cryBP1 gene, and also analyzed the whole-genome sequencing by using the NGS technologies to gain a better understanding of the sequence and structural changes between tall line or natural cultivar and rice reference. than others, was confirmed with two copies of foreign gene insertion, which was inserted in one genomic site facing each other between the position 2,430,152~2,430,151 of rice chromosome 12 without any deletion of genomic sequences. Sequencing analysis also revealed that 18bp-unknown sequences were added in the 5′ insertion site of T-DNA. This position in rice genome was confirmed with none of expressed gene sites. By the NGS analysis, we detected 86560 SNPs and 1091/1472 large insertion/deletion (indel) sites (100bp) between BP23 and rice reference, and 84743 SNPs and 1094/1451 large indels between natural cultivar Nagdong and rice reference. The possible mechanisms for the gene mutation, the developmental and tissue expression of the taller height in BP23 line may need to be scrutinized a few more.

        Piplartine induces caspase-mediated apoptosis in PC-3 human prostate cancer cells.

        <P>The present study examined the anti-proliferative effects of piplartine on the human prostate cancer cell line PC-3. This is the first report demonstrating the piplartine anti-cancer activity toward prostate cancer cell lines, although its precise mechanism of action is still not completely defined. In MTT assays, it preferentially inhibited growth of androgen-independent PC-3 cells in a dose-dependent (3-30 microM) and time-dependent (12-48 h) manner. In PC-3 cells, it showed an IC50 of 15 microM after 24 h of treatment. After a 24-30 microM treatment for 24 h, there were some reduction of cell volume, cell vacuolization, chromatin condensation and increased number of apoptotic cells visible by light and fluorescence microscopy. Agarose gel electrophoresis revealed that cells treated with piplartine exhibited DNA fragmentation. In addition, growth inhibition of PC-3 cells was associated with G2/M arrest and sub-G1 accumulation. Higher concentrations (24-30 microM) of piplartine modulated apoptosis-related protein expression by down-regulating cdc-2 expression and up-regulating PARP/procaspase-3 cleavage. Also, PC-3 cells treated with piplartine demonstrated caspase-3 activation, as observed with an in vitro caspase-3 colorimetric assay kit. Taken together, these results demonstrated that high concentrations of piplartine exhibited anti-proliferative and anti-cancer effects on PC-3 cells and that caspase-3-mediated PARP cleavage and cell cycle arrest at G2/M phase are involved in the underlying cellular mechanism of the apoptosis process.</P>
