<P>Background:The objective of this study was to investigate the underlying molecular mechanisms and the therapeutic time window for preventing astrogliosis with erythropoietin (EPO) treatment after in vitro modeled spinal cord injury (SCI).Methods:Cultured rat spinal cord astrocytes were treated with kainate and scratching to generate an in vitro model of SCI. EPO (100U/mL or 300U/mL) was added immediately or 2, 4, or 8 hours after injury. Some cultures were also treated with AG490, an inhibitor of the EPO-EPO receptor (EpoR) pathway mediator Janus kinase 2 (JAK2). To evaluate neurite extension, rat embryonic spinal cord neurons were seeded onto astrocyte cultures and treated with EPO immediately after injury in the presence or absence of anti-EpoR antibody.Results:EPO treatment at up to 8 hours after injury reduced the expression of axonal growth inhibiting molecules (glial fibrillary acidic protein, vimentin, and chondroitin sulfate proteoglycan), cytoskeletal regulatory proteins (Rho-associated protein kinase and ephephrin A4), and proinflammatory cytokines (tumor necrosis factor-alpha, transforming growth factor-beta, and phosphorylated-Smad3) in a dosedependent manner (P<.001). Most effects peaked with EPO treatment 2-4hours after injury. Additionally, EPO treatment up to 4 hours after injury promoted expression of the EpoR (>2-fold) and JAK2 (>3-fold) in a dose-dependent manner (P<.001), whereas co-treatment with AG490 precluded these effects (P<.001). EPO treatment up to 4hours after injury also enhanced axonal b-III tubulin-immunoreactivity (>12-fold), and this effect was precluded by co-treatment with an anti-EpoR antibody (P<.001).Conclusions:EPO treatment within 8 hours after injury reduced astrogliosis, and EPO treatment within 4 hours promoted neurite outgrowth. EPO therapy immediately after spinal cord injury may regulate glia to generate an environment permissive of axonal regeneration.</P>
모체의 당뇨병이 흰 쥐 태자 폐세포의 glycosylation에 미치는 영향

Stretozotocin으로 유발된 모체의 당뇨병이 태자 폐세포의 당질화(glycosylation) 과정에 미치는 영향을 알아보기 위하여 태자기 19일과 21일째의 폐조직을 ConA, WGA, UEAL, RCAL, MPA등의 lectin으로 염색하여 결합양상을 조사하였으며, image analyzer를 사용하여 형태학적인 계측을 시행하여 얻은 결과는 아래와 같다. 태자기 21일째의 폐조직에서 bronchiolar epithelium이 차지하는 면적은 두 군데에서 비슷하였으나 alveolar epithelial cell과 mesenchymal cell이 차지하는 면적의 비율은 실험군이 74.2%, 정상군이 56.8%를 나타내었으며, air space의 면적비율은 실험군에서 낮게 나타났다. 다섯 종류의 lectin으로 조직화학염색을 시행한 결과 α-mannose, N-acetylgucosamine, fucose, β-galactose와 각각 결합하는 ConA, WGA, UEAI, RCAI의 binding affinities는 정상군과 큰 차이를 나타내지 않았으나 N-acetyl-D-galactosamine과 결합하는 MPA의 binding intensity는 실험군에서 감소되어 나타났다. 이는 stretozotocin으로 유발된 모체의 당뇨병이 태자 폐조직의 O-linked glycosylation과정 중 어느 단계를 저해시켜 나타난 결과가 아닌가 추측된다. This study was to investigate the effects of maternal diabetes on glycosylation in the lung of the fetal rat using lectin histochmistry. Maternal diabetes was induced by intraperitoneal injection of stretozotocin(75mg/kg the body weight) into pregnant Sprague-Dawley rats on the 7th day of geastation. Fetuses of streptozocin-induced diabetic rats exhibited delayed lung maturation and reduced air space. In lectin histochemistry, the binding of Maclura pomifera(MPA) to fetal lings from diabetic mothers was reduced, but no significant changes in the binding of Concanavalin A(ConA). Wheat germ agglutinin(WGA), Ricinus communis I (RSAI) and Ulex eupopaeus I (UEAI) were noted. Because the MPA has affinity to terminal N-acetyl-D-galactosamine residues con stantly linked O-glycosidically to serine or threonine, the present finding may indicate that maternal diabetes interfere with the processes of O-linked glycosylation in fetal rat lung.

The purpose of this research was bring to be communities on field research to activate and being progress to "sport for all program." For the research 6 major cities-Seoul, Inchon, Deagu, Pusan, and Kyangju-and 19 minor cities have been selected. The survey questionnaire developed by the investigtors was mailed to the administrative persons of the City Hall and Local Hall before one week aheard the actural survey. The contents of this research has been divided into three major fields; 1. administration and planning, 2. menagements 3. evaluation. The research show the actual conditions of the local communities` sports for all is still limited. The results of this research are as following; 1. For improvement and progress of for all, the organization should have human power by stationing of sport 2. Currently the total government spending alloctates for soprts for all is approximate 2% but it needs to be increase to be effective. 3. Adimistration of long range plan of various fields is necessary. 4. Rather than to build new facilities, the current facilities needs to be revamped to meet the necessary functions. 5. Presently the possessor of liscences are not fully accounted for, thus there needs a was to account for them I hus tobe useful for sports for all programs. 6. Program should be diverse and complete to give the participants various selections. 7. Club of sports for all sould be formed not limited only to adult memberships. 8. Advertisement and continued education is necessary to bring about the understanding of sports for all. 9. Evaluation is necessary to assessment methodology and contents. Evalustion is to be able to be used as a feedback for future evaluation of sports.
