Participation of Protein Synthesis in in vitro Oocyte Maturation and Fertilization in Cattle

Bovine oocytes with compact and complete cumulus cells were cultured for up to 24h in TCM199 buffered with 25mmol/l hepes and supplemented with 10% FBS (fetal bovine serum), 1mg/ml $17{\beta}$-estradiol, 20IU/ml hCG(human chorionic gonadotropin). All of the oocytes were divided into at 6 groups depending upon incubation times (control, 0 hour, 6 hours, 12 hours, 16 hours, 18 hours). To all experimental media, $200{\mu}g/ml$ puromycin was added at different incubation times mentioned above. Following these culture times, in vitro insemination was conducted with frozen-thawed bovine spermatozoa in medium BO (Brackett and Oliphant medium for in vitro insemination) with $10{\mu}g/ml$ BSA(bovine serum albumin) and 10 mg/ml heparin added. After 22h culture, the oocytes were fixed with acetic alcohol solution and stained with orcein acetic solution to evaluate sperm nuclear progression. Addition of puromycin after 0, 6 and 12 h of culture resulted in near of oocyte maturation at the M1 stage. Contrariwise, puromycin addition after 12 h of culture led to restoration of nuclear progression to M2 stage. On the one hand, puromycin affected the synthesis of Cyclin B protein that may be involved in the oocyte maturation and sperm capacitation for in vitro fertilization. The present study suggests the participation of protein synthesis, cyclin B, in the oocyte development from M1 to M2 stages in vitro.

Alignment failure sometimes occurs during the rubbing process because the rubbing cloth comes in direct contacts with the surface of the alignment film. A number of researches observed and evaluated the surface of the alignment film after the rubbing process had been reported. The real-time rubbing process has not been observed directly yet, though. In this study, the movement of the piles of the rubbing cloth during the rubbing process was observed with a high-speed camera. Furthermore, the relationship between the rubbing scratch on the alignment films and the movement of the pile was investigated. It was found that the movement of the pile affected the rubbing scratches.

<P>Cosmic microwave background (CMB) is an important source of information about the origin of our universe. In particular, odd-parity large angular scale patterns in the CMB polarization, the primordial B-modes, are strong evidence for an inflationary universe, related to the accelerating expansion of the metric. We are developing a unique telescope, GroundBIRD, to take CMB polarization measurements. The telescope combines novel techniques: high-speed rotation scanning, cold optics, and microwave kinetic inductance detectors (MKIDs). We evaluated the response of MKIDs on the rotation stage. Method of shielding from the geo-magnetic field is established. We have also developed a receiver cryostat. We are able to maintain a sufficient cold status for observations on the optical configuration. We plan to start commissioning the system by observing CMB in Japan in 2015-2016. We will then deploy GroundBIRD in the Canary Islands for further scientific observations.</P>
The Livelihood of Riverine People in Rural Bangladesh: Insights from climate vulnerable context

This study assesses the vulnerabilities and determines the livelihood status of individuals living in riverine areas. Additionally, it explores the relationships between selected characteristics of the riverine population and their vulnerability-based livelihood status. Data were collected from 70 heads of households residing in the Narail district of Bangladesh. To develop a scale for assessing vulnerability, a Focus Group Discussion was conducted to identify potential vulnerability indicators. Pearson's product-moment correlation coefficient (r) was calculated to investigate the connections between respondents' characteristics and their livelihood status within the context of vulnerability. Results revealed that the majority of respondents (90 percent) faced a moderate level of vulnerability in their living conditions. The primary vulnerabilities observed in the area included feelings of insecurity due to unsafe infrastructure, displacement caused by riverbank erosion, and disruptions to income opportunities. Regarding livelihood status, the majority of respondents (82.9%) fell into the medium category. Notably, there was considerable variation in different livelihood assets, with a significant proportion (80%) of respondents having low economic capital. Among the selected characteristics of the riverine population, including household size, household assets, household farm size, annual household income, and credit received, significant positive relationships were found with their vulnerability-based livelihood status. However, no significant relationships were observed between the remaining characteristics, namely age, years of schooling, organizational participation, decision-making ability, innovativeness, and vulnerability-based livelihood status. This study emphasizes the urgent need for interventions to address the vulnerabilities faced by riverine communities, including improving infrastructure safety, reducing riverbank erosion, and developing sustainable earning opportunities.
