        A variant acute promyelocytic leukemia with t(11;17) (q23;q12); <i>ZBTB16-RARA</i> showing typical morphology of classical acute promyelocytic leukemia

        <P>A subgroup of acute leukemia with morphology resembling acute promyelocytic leukemia (APL) shows variant translocations involving <I>RARA</I> and has a different morphology from that of classical APL. The variant APL with t(11;17)(q23;q12); <I>ZBTB16-RARA</I> subgroup has been reported to have leukemic cells with regular nuclei, many granules, absence of Auer rods, an increased number of Pelgeroid neutrophils, strong myeloperoxidase (MPO) activity, and all-trans-retinoic-acid (ATRA) resistance. Here, we report a case of variant APL with t(11;17)(q23;q12); <I>ZBTB16-RARA</I> showing typical morphological features of classical APL, including numerous Auer rods and faggot cells. The leukemic cells expressed CD13, CD33, CD117, human leukocyte antigen (HLA)-DR, and cytoplasmic-MPO on the immunophenotyping study. The diagnosis was confirmed by cytogenetic and molecular studies. To distinguish variant APL cases from classical APL cases, regardless of whether morphologically the findings are consistent with those of classical APL, combining morphologic, immunophenotypic, cytogenetic, and molecular studies before chemotherapy is very important.</P>

      • Interphase Cytogenetics of Non-Hodgkin's Lymphoma Using Non-Fluorescent in Situ Hybridization in Paraffin Embedded Tissue

        Paraffin-embedded tissue samples from 30 cases of non-Hodgkin's lymphoma(NHL) and 10 of reactive hyperplasia, were processed for interphase cytogenetic chromosomal study. We performed non-fluorescent in situ hybridization(NFISH) using enzymatic method with digoxigenin-labeled DNA centromeric probes for chromosome 7, 12, 18 and X, and painting probe for chromosome 18. Chromosomal aberrations were observed in 27(90%) out of 30 cases of NHL. The most commonly observed numerical aberratiort was extracopy of X chromosome. There were some characteristic aberrations corresponding to each grades and groups of NHL by International Working Formulation: In low grade NH]L(9 cases), a third was associated with extracopy of chromosome 12, and disomy X was frequently found in small Iymphocytic Iymphoma(75%). With intermediate grade(16 cases), tetraploidy(25%), translocation of chromosome 18(25%), and extracopy of chromosome 18(19%) were chararteristically associated. These result suggest that interphase NFISH is a easily performable method in retrograde cytogenetic study of archival materials. Some specifically correlated chromosomal aberrations corresponding to the histopathologic grades and groups could provide us more valuable information for determining pathologic diagnosis and assessing the clinical outcome of NHL.

        Characteristics of hematologic malignancies with coexisting t(9;22) and inv(16) chromosomal abnormalities

        <P><B>Background</B></P><P>The coexistence of t(9;22)(q34;q11.2) and inv(16)(p13q22) chromosomal abnormalities is extremely uncommon, and only a small number of such cases have been reported. Here, we characterized 7 cases of hematologic malignancy exhibiting t(9;22) and inv(16) coexistence.</P><P><B>Methods</B></P><P>We reviewed the cytogenetic data for hematologic malignancies treated at the Catholic Blood and Marrow Transplantation Center between January 2004 and June 2013. We identified 7 cases exhibiting t(9;22) and inv(16) coexistence. In addition, we analyzed mutations in the <I>IKZF1</I>, <I>NPM1</I>, <I>FLT3</I>, <I>N-RAS</I>, <I>K-RAS</I>, <I>c-KIT</I>, and <I>TP53</I> genes.</P><P><B>Results</B></P><P>Four cases of chronic myelogenous leukemia (CML; 1 chronic phase, 2 accelerated phase, and 1 blast phase) and 3 cases of acute myeloid leukemia (AML; 1 de novo and 2 therapy-related) were identified. The percentages of circulating blasts and bone marrow eosinophils were higher in AML cases than in CML cases (53% vs. 5% and 30% vs. 5.5%, respectively). The proportions of each chromosomal abnormality were used along with follow-up karyotyping results to identify secondary changes. In <I>BCR/ABL</I>, a p210 fusion transcript was associated with CML, whereas a p190 fusion transcript was associated with AML. One patient with AML harbored 2 mutations: <I>c-KIT</I> D816V and <I>TP53</I> E11Q. All patients except 1 with CML blast phase sustained clinical remission after treatment, which included an imatinib mesylate regimen.</P><P><B>Conclusion</B></P><P>This study shows that observations of bone marrow morphology, initial and follow-up cytogenetic studies, and karyotyping of <I>BCR/ABL1</I> and <I>CBFB/MYH11</I> provide valuable information for characterizing hematologic malignancies exhibiting t(9;22) and inv(16) coexistence.</P>

        보육시설의 영유아 건강관리 순회방문 서비스 모형개발

        Objectives: The number of children at child day-care centers is rising and the entering age is getting younger, and it requires better management of health care by health professionals. The purpose of this study was to develop visiting nurse's service model in child day-care centers for promoting child's health and preventing and detecting health problems. Methods: Visiting nurse's service program in child day-care centers was developed based on the survey result on child health need, and nonequivalent control group pretest-posttest design study was performed to analyze the service effect. Three facilities were assigned to each experimental and control group. About 100 children of each group, their mothers, and their teachers were participated. Finally this program was evaluated with structure, process, and outcome framework. Results: This program was served once a week for 12 weeks. Service took about 115 minutes and routine check up, providing health information, infection prevention occupied nurse's activities mostly. This service program was effective in finding health problems, reducing accident and safety risk factors. Parents' and teachers' satisfaction on this program were high. Facilitating factors were independent space for exam, collaboration with staff in providing this program. On the other hand, barrier was interrupting the regular program when nurse visited daycare centers. Conclusion: This study showed the possibilities of effective child health care model in child day-care centers and suggests this new model as a strategy in policy making for child health promotion.

        최승희춤의 아시아적 후원환경론<sup>+</sup>

        본 연구는 2011년 현재 105건(단행본, 석ㆍ박사학위 논문, 학술지 논문, 학술대회집 포함)에 이르는 최승희의 연구성과를 토대로 최승희춤이 시대를 앞서간 근대 무용의 선구자로 자리매김 하기까지의 후원환경에 초점을 맞추어 고찰하였다. 최승희춤의 후원환경에 대하여 국내와 국외를 구분하여 아시아의 지역으로 한정하였다. 국내의 후원환경으로는 가족구성원 중에서 가장 중요한 역할을 차지했던 최승일과 안막을 둘러싼 문화 예술인과 단체, 언론 기관 등에 대한 긍정적 후원환경이 형성되었다. 국외의 환경으로는 일본에서 결성된 최승희 후원회의 공식 명칭인 “백십자회”의 구성에 대하여 고찰하였으며 그 이외 중국과 북한의 정치 문화와 예술적 후원환경을 살폈다. 이를 토대로 최승희는 전시상황과 정치적 혼란기를 극복하는 공연활동을 전개할 수 있었다고 사료된다. <최승희 제2회 신작무용발표회>를 기점으로 후원회 활동을 시작한 백십자회(白十字會)의 발기인 가운데는 일본에서 일류로 꼽히는 이시이바꾸나 가와바다 야스나리와 같은 예술가, 정치가, 잡지사 사장, 문화인들이 다수 포함되었으며, 특히 조선독립을 위해 활동한 민족지도자들도 포함되어 주목을 끈다. 결과적으로 최승희춤이 세계적으로 주목받는 무용 활동을 할 수 있게 된 기저에는 문학가정 환경하에서 개방적 신식교육을 출발점으로 하였고, 또 진보적이고 문학적인 토대가 수반된 후원회, 정치와언론, 군부의 조력이 배경적 역학구도를 형성하였다. 그와 같은 후원 환경아래 보다 확고한 무용철학을 정립하여 작품 창작의 체계와 공연환경을 구축할 수 있었던 것으로 판단된다. 본연구의 사적 측면에서 가장 의미 있는 결과는 무엇보다 최승희 후원회의 명칭이 백십자회 (白十字會)라는 점을 밝힌 것이다. 그에 더하여 가을 들판에서 투명한 베일을 들고 도약하는 잘 알려진 작품사진의 제목이 「적구흔무(赤丘欣舞)」라는 점을 최초로 밝혔다. This study has considered focusing on the patronage environment of the process where Choi Seung-hee dance had settled down as the pioneer in modern dance ahead of its time based on her performance of current 105 research in 2011 including books, master`s and doctorate degree theses, academic and symposium journals. The patronage environment of Choi Seung-hee dance has been limited to Asian region while classified into domestic and overseas. The domestic patronage environment includes Choi Seung-il who has played the most important role in her family members, cultural artists and association surrounding An-mak (An Pil-seung), and media organization and so on, developing a positive patronage environment. In overseas environment, this study has concentrated on the formation of `the White Cross Institute` which is an official title of the Choi Seung-hee supporters association in Japan, and based on the political culture and supporters` environment in North Korea and China, the performance that described overcoming the wartime condition and the period of political tumult could be staged. Among the initiators of White Cross Institute (白十字會) who had begun the patronage activity at the onset of `the Second Recital of New Choi Seung-hee Dance,` many artists, politicians, presidents of magazine publishers, and cultured individuals who are chosen as the top-notch in Japan such as Baku Ishii or Yasnari Kawabata. Especially, many of the nationalist leaders who had worked for the independence of Joseon Dynasty were included and gained public attention. As a result, at the base of the fact that Choi Seung-hee dance has become internationally famous, the supporters` association, politics and media, and military support involving progressive and literary foundation starting from the open and modern education in the literary family background have formed a dynamic structure in the background. At the groundwork of such patronage environment, one could figure out that more solid philosophy of dance was founded to establish the system of creating works and the performance environment. In a personal aspect, the most meaningful result from this study is that this research is the first to disclose the fact that the title of the Choi Seung-hee supporters association is `the White Cross Institute (白十字會)` and the title of the well-known photograph in which a woman is jumping into the air with a transparent veil on her hands in the field in autumn is `Jeokgu Heunmu (赤丘欣舞).`

        High-ionic-strength pre-concentration via ion concentration polarization for blood-based biofluids

        <P><B>Abstract</B></P> <P>Ion concentration polarization (ICP) preconcentration is a pretreatment method that is actively utilized in the analysis of low-abundance biomolecules. In biofluids (e.g. urine, sweat, blood, serum, plasma, saliva, etc.), low-concentrated target analytes presented among large number of background molecules under a high ionic concentration, there still exist several hurdles in the utilization of ICP preconcentration in the such high ionic fluids. This is especially true in the case of blood-based biofluids, such as plasma and serum, wherein on account of the abundance biomolecules as well as high ionic buffer concentration makes the ICP preconcentration process difficult to realize. In this study, we have demonstrated the realization of ICP preconcentration of blood-based biofluids (serum) using a paper-based preconcentrator. We acquired preconcentration factors (PF) for 25.5, 127.5 and 255 mM of human serum were 40, 10, and 5.5 within 20 min. The squared fluorescence intensity is also found to be inversely proportional to ionic concentration, showing that the PF are strongly correlated to the Debye length of the buffer solution. Using the paper-based preconcentrator, we were able to acquire effective preconcentration factor (PF<SUB>eff</SUB>) of 5.17, 4.88, 4.58, and 4.23 for the 1X, 0.75X, 0.5X, and 0.1X diluted solutions, demonstrating approximately 5 times increase in PF<SUB>eff</SUB> for all prepared human serum samples.</P> <P><B>Highlights</B></P> <P> <UL> <LI> Ion concentration polarization (ICP) preconcentration platform of high-ionic-concentration sample is demonstrated for blood-based samples. </LI> <LI> The squared fluorescence intensity is inversely proportional to ionic concentration, showing that the PFs are correlated to the Debye length. </LI> <LI> We demonstrated approximately 5 times increase in effective preconcentration factor (PF<SUB>eff</SUB>) for all prepared human serum samples. </LI> </UL> </P>

      • Bioactivity and molecular characterization of bombolitins from Bombus ardens, B. consobrinus, B. terrestris and B. ussurensis

        Biological activities of bombolitins from Bombus ardens, B. consobrinus, B. terrestris and B. ussurensis (bombolitinsA, C, T and U, respectively) were examined using hemolytic, anti-microbial, anti-fungal and anti-tumor activity assays.Among the four bombolitins tested, bombolitin T showed the highest hemolytic and anti-tumor activities. All bombolitinsexhibited strong anti-microbial and anti-fungal activities, and bombolitin A specifically possessed the highest anti-microbialactivity against the Gram-negative bacteria Escherichia coli. Circular dichroism spectrometry analysis revealed that allfour bombolitins had over 61.7% and 45.5% of α-helicity in 30 mM sodium dodecyl sulfate and 50% trifluoroethanolbuffers, respectively, which form lipid-membrane-mimicking environments. Bombolitin T showed the lowest IC50 valuesof 8.5 μM and 8.8 μM against SK-OV-3 and NIH-OVCAR-3 cell lines, respectively, after 72 h of treatment, but itsrelative hemolytic activity at a concentration of 200 μM was 2.3-fold higher than that of 0.1% Triton X-100. Thisstudy provides new information on the biological and molecular properties of venom peptides of bumblebees. However,further studies on reducing cytotoxicity of bombolitins are needed for designing selective anti-tumor peptides.

      • Characterization of biological activities of bombolitins from bumblebees Bombus ardens and Bombus ussurensis

        Bombolitin is a venom peptide originally isolated from bumblebees and possesses various biological activities, including hemolytic activity. Bombolitins exhibit amphipathic α-helical structure in lipid-membrane-mimicking environments. To investigate their pharmacological and toxicological properties, anti-tumor, anti-microbial and cytotoxic activities of bombolitins from Bombus ardens and Bombus ussurensis were evaluated. Bombolitins of the two species exhibited extremely high anti-tumor activity against ovarian tumor cells SK-OV-3 and NIH; OVCAR-3 at 25-50 μM, which is 2-fold more potent than other wasp venom peptides studied to date (Yoon et al., 2015; Yoon et al., 2016). The two bombolitins showed significantly high antimicrobial activity. However, bombolotin of B. ussurensis showed no antimicrobial activity against Escherichia coli. In addition to their high levels of anti-tumor activity, bombolitins showed considerable levels of hemolytic activity. Thus, to utilize bombolitins as a potential candidate for anti-tumor peptide drugs, further studies for reducing cytotoxic properties of bombolitns is essential.
