        한국과 일본 대학생의 스포츠상황에 대한 도덕적 가치판단수준 비교

        The purpose of this study was to compare the differences of moral reasoning among general students, sports related majors students, and student athletes in Korea and Japan University. We collected total 612 out of 700 (315 from Korea, 297 from Japan) and 88 questionnaires were excluded questionnaire due to incompleteness and unreliable data. For this study, Hahm-Bellar Values Choice Inventory-16(HBVCI-16), which is a Moral Reasoning Questionnaire on Sports Activities, was used. The collected data was analyzed by t-test, one way ANOVA and two way ANOVA with using SPSS Var. 17.0 statistical package program and result were as follow. First, the Japan student`s overall score of moral reasoning were lower compare to Korean university students. Second, there were 1 % higher score of moral reasoning in woman against man and in younger subjects. Sports majored university students presented 1% higher score against other related students and higher grade students presented 5% higher score against lower grade students. Third, among student athletes of Korea and Japan, although there were no significant differences, it was 0.1% higher among those who engaged in individual sports and 5% higher score in those who have less sports career.

        A Model for Computer-Aided Process Planning System in Flexible Manufacturing Systems

        Most of computer-aided process planning(CAPP) systems have been developed to automate the process planning function. In this paper, we describe an analytical model for a CAPP system in order to improve the performance of production system in flexible manufacturing systems(FMSs) for computer intergrated manufacturing(CIM) architecture. This paper proposes an optimal process planning that minimizes the load time by minimizing the cycle time and the number of workstations using Kang and Hahm's heuristic approach so as to improve the performance of production system under the batch production of discrete products. We also perform simulation using SIMAN language to campare the line utilization of each for various product types. The proposed algorithm can be implemented in existing FMSs for on-line control of product quantity using programmable logic controllers(PLC) and communication devices.

      • Comparative Glycopeptide Analysis for Protein Glycosylation by Liquid Chromatography and Tandem Mass Spectrometry: Variation in Glycosylation Patterns of Site-Directed Mutagenized Glycoprotein

        <P>Glycosylation is one of the most important posttranslational modifications for proteins, including therapeutic antibodies, and greatly influences protein physiochemical properties. In this study, glycopeptide mapping of a reference and biosimilar recombinant antibodies (rAbs) was performed using liquid chromatography-electrospray ionization tandem mass spectrometry (LC-ESI-MS/MS) and an automated Glycoproteome Analyzer (GPA) algorithm. The tandem mass analyses for the reference and biosimilar samples indicate that this approach proves to be highly efficient in reproducing consistent analytical results and discovering the implications of different rAb production methods on glycosylation patterns. Furthermore, the comparative analysis of a mutagenized rAb glycoprotein proved that a single amino acid mutation in the Fc portion of the antibody molecule caused increased variations in glycosylation patterns. These variations were also detected by the mass spectrometry method efficiently. This mapping method, focusing on precise glycopeptide identification and comparison for the identified glycoforms, can be useful in differentiating aberrant glycosylation in biosimilar rAb products.</P>

        Breast Cancer Selective Disruption of Actin Cytoskeleton by Diallyl Trisulfide

        Diallyl trisulfide (DATS) is an attractive anti-cancer phytochemical with in vitro and in vivo growth inhibitory effects against different solid tumors including breast cancer. We have shown previously that an immortalized mammary epithelial cell line (MCF-10A) is resistant to growth inhibition by DATS. In this study, we performed RNA-seq analysis using a breast cancer cell line (SK-BR-3) and MCF-10A cells to gain insights into cancer selective effects of DATS. The Gene Ontology analysis revealed upregulation of genes associated with actin cytoskeleton but downregulation of mitochondria-related genes in the SK-BR-3 human breast cancer cell line but not in the non-oncogenic MCF-10A cell line upon treatment with DATS. Quantitative real-time reverse transcription polymerase chain reaction confirmed DATS-mediated upregulation of several actin cytoskeleton-related genes in the SK-BR-3 cell line. The DATS treatment dose-dependently disrupted actin cytoskeleton in the SK-BR-3 cell line, whereas the MCF-10A cell line was more resistant to this effect. The DATS treatment caused a marked increase in phosphorylation of dynamin-1-like (DRP1) protein in the SK-BR-3 cell line. However, the DATS-mediated apoptosis was not affected by genetic deletion of DRP1 protein. The Reactome pathway analysis showed downregulation of genes associated with citric acid cycle in the SK-BR-3 cell line but not in the MCF-10A cells. However, expression of aconitase 2 or dihydrolipoamide S-succinyltransferase was not affected by DATS treatment. In conclusion, this study reveals that actin cytoskeleton is a novel target of DATS in the SK-BR-3 cell line, which may explain its inhibitory effect on breast cancer cell migration.

        Gene Expression Changes by Diallyl Trisulfide Administration in Chemically-induced Mammary Tumors in Rats

        Diallyl trisulfide (DATS) was shown to be a potent inhibitor of luminal-type MCF-7 xenograft growth in vivo. The present study was conducted to determine the preventive effect of DATS administration using an N-methyl-N-nitrosourea (MNU)-induced rat mammary tumor model, which shares molecular resemblance to luminal-type human breast cancers. The DATS administration (50 mg/ kg body weight, 5 times/week) was safe, but did not reduce mammary tumor latency, incidence, burden or multiplicity. Therefore, we conducted RNA-seq analysis using mammary tumors from control and DATS-treated rats (n = 3 for each group) to gain insights into lack of mammary tumor prevention by this phytochemical. The gene ontology and the Kyoto encyclopedia of genes and genomes pathway analyses of the RNA-seq data revealed upregulation of genes associated with ribosomes, translation, peptide biosynthetic/metabolic process, and oxidative phosphorylation but downregulation of genes associated with mitogen-activated protein kinases. A total of 33 genes associated with ribosomes were significantly upregulated by DATS treatment, including RPL11 and RPS14. Western blotting confirmed upregulation of RPL11 and neurofascin protein expression in mammary tumors from DATS-treated rats when compared to controls. A statistically significant increase in protein level of c-Jun N-terminal kinase 2 was also observed in tumors from DATS-treated rats when compared to controls. On the other hand, expression of complex I subunits NDUFV1 or NDUFS1 was not affected by DATS treatment. These results offer potential explanations for ineffectiveness of DATS in the chemically-induced rat mammary tumor model. Inhibitors of the proteins upregulated by DATS may be needed to improve chemopreventive efficacy of this phytochemical.

        APPROXIMATION ORDER TO A FUNCTION IN $C^1$[0, 1] AND ITS DERIVATIVE BY A FEEDFOWARD NEURAL NETWORK

        We study the neural network approximation to a function in $C^1$[0, 1] and its derivative. In [3], we used even trigonometric polynomials in order to get an approximation order to a function in $L_p$ space. In this paper, we show the simultaneous approximation order to a function in $C^1$[0, 1] using a Bernstein polynomial and a feedforward neural network. Our proofs are constructive.

      • Electroacupuncture Enhancement of Natural Killer Cell Activity Suppressed by Anterior Hypothalamic Lesions in Rats

        Background/Objective: Neuroendocrine hormones are derived from the hypothalamus. The central nervous system, particularly the hypothalamus, is capable of modulating the cytolytic activity of adherent natural killer (NK) cells. In addition, electroacupuncture (EA) stimulation of the Zusanli (ST36) acupoint enhances splenic NK cell and cytokine activities in rats. However, it is still unclear whether the anterior hypothalamus affects this immunomodulation. Therefore, the aim of the present study was to examine the effect of EA stimulation at the Zusanli acupoint on the NK cell activity modulated by an anterior hypothalamic area lesion. Methods: Male Sprague-Dawley rats were used. Lesions were placed by means of a direct current through a concentric electrode. The electric acupuncture stimulation was delivered for 30 min per each experiment at the right ST36 acupoint with an electrical stimulator. The NK cell activity of the spleen was measured by a fluorescence assay. Results: The NK cell activity was significantly reduced on the 2nd day after the lesion, but was restored to that of the sham group by the 7th day. However, when EA was applied for 2 days after the operation, the NK cell activity of the lesion group was restored to that of the sham group. After 7 days of EA, the NK cell activity of the lesion group was slightly higher than that of the sham group. Conclusion: From these results, we can suggest that EA enhances or restores the NK cell activity suppressed by an anterior hypothalamic area lesion.

        Effects of Gamma Irradiation on Nutrient Composition, Anti-nutritional Factors, In vitro Digestibility and Ruminal Degradation of Whole Cotton Seed

        Whole cotton seed (WCS) has become one of the major feed ingredients in TMR for dairy cattle in Korea, and WCS for feed use is mostly imported from abroad. Since this genetically modified oil seed is usually fed to the animal in raw state, its germination ability, if last long, often causes concerns about ecological disturbances. In the process of looking for effective conditions to remove germination ability of WCS this study had the objectives to evaluate the nutritional effects of gamma irradiation at doses of 8, 10 and 12 kGy on changes in nutrient contents, anti-nutritional factors, in vitro digestibility and ruminal degradability. No significant differences were found in proximate analysis of nutrients between raw WCS and gamma irradiated one. Glycine and threonine contents significantly increased when the WCS was exposed to gamma ray as compared to untreated WCS (p<0.05). As for fatty acid composition, no significant differences were observed with the irradiation treatment. Free gossypol in WCS was decreased (p<0.05) by gamma irradiation treatment. Of the 3 different levels of gamma irradiation, a dose of 12 kGy was found to be the most effective in reducing free gossypol concentration. Results obtained from in situ experiment indicated that gamma irradiation at a dose of 10 kGy significantly (p<0.05) lowered rumen degradability of both dry matter and crude protein as compared with raw WCS. However, there were no significant differences in rapidly degradable and potentially degradable fractions of crude protein due to 10 kGy gamma irradiation. Overall, this study show that gamma irradiation at a dose of 10 kGy is the optimum condition for removing germination ability of WCS, and could improve nutritive value for the ruminant with respect to the decrease in both ruminal protein degradability and gossypol content of WCS.
