        Oridinal thermal spray process has developed into two ways, namely, temperature dominated represented by plasma spraying, and velocity dominated represented by HVOF. It is common for both that the particle materials sprayed are basically in melted or half melted condition. New process has developed recently, that is, Cold Spray and Aerosol Deposition. Particle's heating is limited in CS lower than half of the material's melting point. Moreover, exactly no heating is loaded in AD process. Through the investigation on common feature for these three spraying processes, potential of new material process - Particle Deposition, PD - is considered and proposed

      • β-Secretase Protein and Activity Are Increased in the Neocortex in Alzheimer Disease

        Context : Amyloid plaques, a major pathological feature of Alzheimer disease(AD), are composed of an internal fragment of amyloid precursor protein (APP): the 4-kd amyloid-β protein(Aβ). The metabolic processing of APP that results in Aβ formation requires 2 enzymatic cleavage events, а γ-secretase cleavage dependent on presenilin, and a β-secretase cleavage by the aspartyl protease β-site APP-cleaving enzyme(BACE). Objective : To test the hypothesis that BACE protein and activity are increased in regions of the brain that develop amyloid plaques in AD. Methods : We developed an antibody capture system to measure BACE protein level and BACE-specific β-secretase activity in frontal, temporal, and cerebellar brain homogenates from 61 brains with AD and 33 control branis. Results : In the brains with AD, BACE activity and protein were significantly increased (p<.001). Enzymatic activity increased by 63% in the temporal neocortex (P=.007) and 13% in the frontal neocortex (P=.003) in brains with AD, but not in the cerebellar cortex. Activity in the temporal neocortex increased with the duration of AD(P=.008) but did not correlate with enzyme-linked immunosorbent assay measures of insoluble Aβ in brains with AD. Protein level was increased by 14% in the frontal cortex of brains with AD (P=.004), with a trend toward a 15% increase in BACE protein in the temporal cortex (P=.07) and no difference in the cerebellar cortex. Immunohistochemical analysis demonstrated that BACE immunoreactivity in the brain was predominantly neuronal and was found in tangle-bearing neurons in AD. Conclusions : The BACE protein and activity levels are increased in brain regions affected by amyloid deposition and remain increased despite significant neuronal and synaptic loss in AD.

      • Activation-induced Apoptosis of Peripheral Lymphocytes Treated with 7-hydroxystaurosporin, UCN-01

        7-hydroxystaurosporine (UCN-01) is a new anticancer agent which exerts an inhibitory effect on cell cycle check points and is currently under phase I clinical trials in US and Japan. Preliminary clinical data indicated that UCN-01 remained in plasma at high concentrations for long periods of time. this unavoidable high plasma drug exposure is likely to lead to hematological toxicties in patients. In the present study, cultured human peripheral blood lymphocytes (PBLs) were used to evaluate the possible hematological toxicities of UCN-01 treatment. UCN-01 treatment. UCN-01 induces apoptosis, and the induction of apoptosis-related surface markers were also examined to investigate the involvement of these molecules in UCN-01-induced apoptosis in PBLs. In vitro viability of PBLs was decreased by high dose of UCN-01 (25 microM, 3-day exposure). This effect of UCN-01 was significantly suppressed by the presence of human serum, suggesting that some specific inhibitory factor(s) in human serum may antagonize the lympholytic effect of UCN-01. The percentage of annexin V-positive PI-negative cells increased with exposure to UCN-01 in a time- and dose-dependent manner; by up to 30.3% after exposure to 25 microM UCN-01 for 3 day. At the same time, the expression of both interleukin-2 receptor (IL-2R, CD25) and Fas (CD95), analyzed by flow cytometry, was induced. Con A-stimulated PBLs were more sensitive to UCN-01-induced apoptosis than non-stimulated lymphocytes and UCN-01 increased the sFas-L released into culture medium from con A-stimulated PBLs. Therefore, lymphocyte depletion mediated by activation-induced apoptosis is likely to occur in patients treated with UCN-01 at high doses. (Investigational New Drugs 17(4):335-341, 1999)

        Steel-beam-to-concrete-filled-steel-tube-column Moment Connections in Japan

        Diagnostic Modalities for FGF23-Producing Tumors in Patients with Tumor-Induced Osteomalacia

        Fibroblast growth factor 23 (FGF23) is a hormone that is produced by osteocytes and regulates phosphate and vitamin D metabolism through binding to the Klotho-FGF receptor complex. Excessive actions of FGF23 cause several kinds of hypophosphatemic rickets/osteomalacia. Tumor-induced rickets/osteomalacia (TIO) is a paraneoplastic syndrome caused by overproduction of FGF23 from the responsible tumors. Because TIO is cured by complete resection of the causative tumors, it is of great clinical importance to locate these tumors. Several imaging methods including skeletal survey by magnetic resonance imaging and octreotide scintigraphy have been used to identify the tumors that cause TIO. However, none of these imaging studies indicate that the detected tumors are actually producing FGF23. Recently, systemic venous sampling was conducted for locating FGF23-producing tumor in suspected patients with TIO and demonstrated that this test might be beneficial to a subset of patient. Further studies with more patients are necessary to establish the clinical utility of venous sampling in patients with TIO.

        Immigration Policy in Japan for a New Era of Community Development

        Japan has been struggling to deal with population decline issue which has brought labor shortage. Currently, it is warned that a large number of municipalities will lose their population, and some might disappear by 2040. In order to avoid the disappearance, the government has suggested countermeasures. Acceptance of foreign labor is one of the important means to maintain Japan`s labor force and economy, however, Japan has not succeeded in recruiting high-skilled foreign nationals who can be a potential permanent resident and contribute to Japan`s economy as well as low birth rate. Once, Japan was in need of cheap labor, the government renewed its immigration policy and began to receive people with Japanese links who are called Nikkeijin. They took part in factory jobs and contributed Japan`s economy, however, the economic crisis has brought their return to home country. Thus, Japan has lost the potential permanent residents. Currently, the acceptance of foreign workers is still one of the prescriptions for population decline. The government focuses on accepting “high-skilled workers” rather than importing “immigrants”, which resulting in “revolving door migration”. In order to attract high-skilled workers and progress their settlement in Japan, more strategic way or realistic immigration policy is needed.
