        Identification of novel rheumatoid arthritis-associated MiRNA-204-5p from plasma exosomes

        Rheumatoid arthritis (RA) is an autoimmune disease characterized by infiltration of immune cells in the synovium. However, the crosstalk of immune cells and synovial fibroblasts is still largely unknown. Here, global miRNA screening in plasma exosomes was carried out with a custom microarray (RA patients vs. healthy controls = 9:9). A total of 14 exosomal miRNAs were abnormally expressed in the RA patients. Then, downregulated expression of exosomal miR-204-5p was confirmed in both the replication (RA patients vs. healthy controls = 30:30) and validation groups (RA patients vs. healthy controls = 56:60). Similar to the findings obtained in humans, a decreased abundance of exosomal miR-204-5p was observed in mice with collagen-induced arthritis (CIA). Furthermore, Spearman correlation analysis indicated that plasma exosomal miR-204-5p expression was inversely correlated with disease parameters of RA patients, such as rheumatoid factor, erythrocyte sedimentation rate, and C-reactive protein. In vitro, our data showed that human T lymphocytes released exosomes containing large amounts of miR-204-5p, which can be transferred into synovial fibroblasts, inhibiting cell proliferation. Overexpression of miR-204-5p in synovial fibroblasts suppressed synovial fibroblast activation by targeting genes related to cell proliferation and invasion. In vivo assays found that administration of lentiviruses expressing miR-204-5p markedly alleviated the disease progression of the mice with CIA. Collectively, this study identified a novel RA-associated plasma exosomal miRNA-204-5p that mediates the communication between immune cells and synovial fibroblasts and can be used as a potential biomarker for RA diagnosis and treatment.

      • Macrophages Promote Coal Tar Pitch Extract-induced Tumorigenesis of BEAS-2B Cells and Tumor Metastasis in Nude Mice Mediated by AP-1

        Background: We sought to evaluate the role of tumor associated macrophages (TAMs) on the promotion of coal tar pitch extract (CTPE)-induced tumorigenesis of human bronchial epithelial cells (BEAS-2B) and tumor metastasis in nude mice, and related mechanisms. Materials and Methods: BEAS-2B cells were first treated with 2.4 mg/mL CTPE for 72 hours. After removal of CTPE, the cells were continuously cultured and passaged using trypsin-EDTA. THP-1 cells were used as macrophage-like cells. BEAS-2B cells under different conditions (n=6/group) were injected into the back necks of nude mice, and alterations of tumor xenograft growth, indicative of tumorigenicity, and tumor metastasis were determined. Pathological changes (tumor nests and microvascular lesions) of HE-stained tumor tissues were also evaluated. The expression of AP-1(c-Jun) in xenografts and metastatic tumors was determined using immunohistochemistry. Results: Tumor size and weight in nude mice transplanted with the mixture of CTPE-induced passage 30 BEAS-2B and THP-1 cells (2:1) were increased compared to those from the CTPE-treated BEAS-2B cells at passage 30 alone at different observation time points. Tumor metastasis to lymph nodes and liver was only detected after transplantation of a mixture the two kinds of cells. The numbers of tumor nests and microvascular lesions, and the expression levels of AP-1 (c-Jun) in tumors from the mixture of two kinds of cells were increased apparently in contrast to those in tumor from the CTPE-treated BEAS-2B cells of passage 30 alone. In addition, there was positive correlation between AP-1 (c-Jun) expression level and the number of microvascular lesions, or between AP-1 (c-Jun) expression level and tumor metastasis in these two groups. Conclusions: TAMs not only facilitate tumorigenesis transformation of CTPE-induced BEAS-2B cells, but also promote tumor growth, angiogenesis and metastasis in nude mice in vivo, which may be mediated by AP-1.

        Self-management Experience of Middle-aged and Older Adults With Type 2 Diabetes: A Qualitative Study

        Purpose: Diabetes mellitus has been either the fourth or fifth leading cause of death among Taiwanese adults during 1995e2015. Older adults with diabetes are at higher risk of developing diabetic macrovascular and micro-vascular complications. The purpose of this study explored the self-management experiences of middle-aged and older adults with diabetes through a focus group. Methods: Purposive sampling was used to recruit patients with diabetes from the metabolic outpatient clinics of medical centers and regional hospitals in Taiwan. Two focus groups, comprising a total of 23 participants, were employed to collect data, and group discussions were held a total of four times in an education room that was distant from clinical areas. Results: Three themes were generated from analysis of the collected data: (1) “listening to the voice of the body and observing physical changes,” (2) “re-recognizing diabetes and challenges,” and (3) “selfmanagement implementation dilemmas.” This study provided new insights into the experiences of middle-aged and older adults in Taiwan regarding their self-management of diabetes. Conclusion: Healthcare teams should be involved in the self-management education of patients with diabetes as early as possible to reduce patients’ anxiety and to develop more patient-centered, culturesensitive clinical skills. In addition to monitoring patients’ self-management, healthcare professor should pay more attention to patients’ successful adaptation to and coexistence with the disease.

        Effects of Matching Conditions Between Crocheted Mesh Tube and Metal Tube on Crushing Behaviour and Energy Absorption of Interlayer Tubes

        This paper focuses on comparing the quasi-static crushing mechanical behaviour and energy absorption characteristics of four types of interlayer tubes formed by combining mesh tube and 304 tube with two diff erent interface matching methods (tight fi t and metallurgical bonding). The deformations and energy absorptions of interlayer tubes are still dominated by 304 tube. The mesh tube and metallurgical points are able to induce and improve the deformation of 304 tube by changing its axial crushing stress state, thus, aff ecting the shapes and quantities of its folds, changing its deformation behaviours and modes, and fi nally forming interlayer tubes with diff erent mechanical behaviours and energy absorption characteristics. The tight fi t interlayer tubes exhibit better mechanical behaviour and energy absorption compared with the hollow 304 tube. The sintering interlayer tubes matching by metallurgical bonding can further improve the mechanical behaviours and energy absorptions of interlayer tubes and present more stable crushing deformations and stronger bearing capacities. It is an eff ective method to improve the mechanical behaviours and energy absorption characteristics of interlayer tubes by changing interface matching modes.

        Concentrated Trading and Order Execution Costs

        Prior studies indicate that investors who hold more concentrated investment portfolios per-form better. In our paper, we examine whether concentration benefits occur in trading set-tings. We find that US traders who focus on fewer stocks pay lower order execution costs. Concentrated trading behavior is correlated across different dimensions. Traders who concen-trate on a smaller number of stocks also concentrate in fewer market centers, at particulartimes of the day, and on certain order types. Our results suggest that trading costs varyacross market participants, and that by focusing on a smaller subset of securities, traders gainadvantages for order execution.

        Do Stock Prices Underreact to Information Conveyed by Investors’ Trades? Evidence from China

        We examine the process of stock prices adjusting to information conveyed by the trading process. Using the price impact of a trade to measure its information content, we show that the weekly price impact has significant cross-sectional predictive power for returns in the subsequent week. The effect is sensitive to the level of informational asymmetry. We find that the price impact contains information that is not fully captured by public order flows and that a lead–lag effect exists regarding the arrival of information to different groups of investors. Our finding suggests a price under-reaction to trading information, which can be explained by the gradual-information-diffusion theory.

      • Wave propagation in double nano-beams in thermal environments using the Reddy's high-order shear deformation theory

        We study the bending wave, shear wave and longitudinal wave characteristics in the double nanobeams in this paper for the first time, in the process of research, based on the Reddy's higher-order shear deformation theory and considering shear layer stiffness, linear stiffness, inter-laminar stiffness, the pore volume fraction, temperature variation, functionally graded index influence on wave propagation, based on the nonlocal strain gradient theory and Hamilton variational principle, the wave equation of the double-nanometer beams are derived. Since there are three different motion states for the double nanobeams, which includes the cases of "in phase", "out of phase" and "one nanobeam fixed", the propagation characteristics of shear-, bending-, and longitudinal- waves in these three cases are discussed respectively, and some valuable conclusions are obtained.

        TRIM22 promotes the proliferation of glioblastoma cells by activating MAPK signaling and accelerating the degradation of Raf-1

        The tripartite motif (TRIM) 22 and mitogen-activated protein kinase (MAPK) signaling pathways play critical roles in the growth of glioblastoma (GBM). However, the molecular mechanism underlying the relationship between TRIM22 and MAPK signaling remains unclear. Here, we found that TRIM22 binds to exon 2 of the sphingosine kinase 2 (SPHK2) gene. An ERK1/2-driven luciferase reporter construct identified TRIM22 as a potential activator of MAPK signaling. Knockout and overexpression of TRIM22 regulate the inhibition and activation of MAPK signaling through the RING-finger domain. TRIM22 binds to Raf-1, a negative regulator of MAPK signaling, and accelerates its degradation by inducing K48-linked ubiquitination, which is related to the CC and SPRY domains of TRIM22 and the C1D domain of Raf-1. In vitro and in vivo, an SPHK2 inhibitor (K145), an ERK1/2 inhibitor (selumetinib), and the nonphosphorylated mutant Raf-1S338A inhibited GBM growth. In addition, deletion of the RING domain and the nuclear localization sequence of TRIM22 significantly inhibited TRIM22-induced proliferation of GBM cells in vivo and in vitro. In conclusion, our study showed that TRIM22 regulates SPHK2 transcription and activates MAPK signaling through posttranslational modification of two critical regulators of MAPK signaling in GBM cells.

      • Network Traffic Identification Algorithm Based on Neural Network

        In this paper, a network traffic identification model is established using a multilayer excitation function quantum neural network which is suitable for data classification. Because the conventional quantum neural network has different target function in the training of the weights of the network and the sigmoid function of the neurons in the hidden layer, the coupling effect of the two parameters is not processed. This will result in the middle and later stage of the training iteration process, and it may be possible to reduce the objective function value of a kind of parameter, and make the objective function value of another kind of parameter increase. In order to avoid this situation, using LM algorithm to optimize, using the same objective function not only as the target function of the network weight, but also the function of translational spacing of sigmoid function of neurons in the hidden layer, and the training objective is to minimize the sum of squared error of the neural network output and the desired value. Finally, the recognition performance of the proposed algorithm is compared with that of the conventional quantum neural network and LM-BP neural network. The results show that the convergence rate of the proposed algorithm is the fastest and the convergence accuracy is the highest.

      • High Mobility Group Box 1 Protein Is Methylated and Transported to Cytoplasm in Clear Cell Renal Cell Carcinoma

        Background: The high mobility group box 1 (HMGB1) protein is a widespread nuclear protein present in most cell types. It typically locates in the nucleus and functions as a nuclear cofactor in transcription regulation. However, HMGB1 can also localize in the cytoplasm and be released into extracellular matrix, where it plays critical roles in carcinogenesis and inflammation. However, it remains elusive whether HMGB1 is relocated to cytoplasm in clear cell renal cell carcinoma (ccRCC). Methods: Nuclear and cytoplasmic proteins were extracted by different protocols from 20 ccRCC samples and corresponding adjacent renal tissues. Western blotting and immunohistochemistry were used to identify the expression of HMGB1 in ccRCC. To elucidate the potential mechanism of HMGB1 cytoplasmic translocation, HMGB1 proteins were enriched by immunoprecipitation and analyzed by mass spectrometry (MS). Results: The HMGB1 protein was overexpressed and partially localized in cytoplasm in ccRCC samples (12/20, 60%, p<0.05). Immunohistochemistry results indicated that ccRCC of high nuclear grade possess more HMGB1 relocation than those with low grade (p<0.05). Methylation of HMGB1 at lysine 112 in ccRCC was detected by MS. Bioinformatics analysis showed that post-translational modification might affect the binding ability to DNA and mediate its translocation. Conclusion: Relocation of HMGB1 to cytoplasm was confirmed in ccRCC. Methylation of HMGB1 at lysine 112 might the redistribution of this cofactor protein.
