No effective antiviral therapy is proven yet, but clinical use of ribavirin (RBV) has been tried. We investigated the antiviral effect of RBV against SFTSV in vitro. Methods: To test for cytotoxicity of RBV, Vero cells were treated with different concentrations of RBV (3.90 to 500 μg/mL, two-fold dilution) and analyzed by cell viability MTS assay 48h post-infection. To determine antiviral activity of RBV against SFTSV, Vero cells were infected with SFTSV strain Gangwon/Korea/2012 at 100 TCID50 (50% tissue culture infective dose) per well in a 96-well plate, and RBV was added at the concentrations showing no or minimal cytotoxicity. Viral RNAs were extracted from the culture supernatants and quantifi ed using one-step real-time reverse transcription- PCR to amplify the partial large segment of SFTSV. Statistical analysis was done by one-way ANOVA with Tukey`s post hoc test. Results: Cytotoxicity due to RBV was not observed at RBV concentration =31.3 μg/ mL. Viral RNAs at 24h post-RBV treatment were reduced with increasing RBV concentrations (1-32 μg/mL), compared with those of mock-treated cells (P <0.01, Figure). Half maximal inhibitory concentration (IC50) of RBV was 3.69 μg/mL at 24h post-RBV treatment. Conclusions: Our study shows that RBV has antiviral effect against SFTSV in a dose-dependent manner. Further studies are required to evaluate the effi cacy of RBV in SFTS.
Effects of γ-Irradiation on the Antiallergic Activity of Alginate

This study was performed to determine the effects of γ-irradiation on the antiallergic activity of alginate. An alginate aqueous solution was γ-irradiated at 3, 5, 7, 10, 20, and 100 kGy. First, the molecular weight (Mw) of alginate rapidly decreased as the γ-irradiation dose increased up to 20 kGy. Then, the antiallergic activity of the γ-irradiated alginate was measured. Interlukin (IL)-4 cytokine and IgE level were significantly decreased in the γ-irradiated alginate groups as compared to the control group in vitro. The mice intraperitoneally administered with the γ-irradiated alginate exhibited lower ovalbumin-specific IgE and IgG1 level in serum than the control mice. Furthermore, the γ-irradiated alginate suppressed total and ovalbumin-specific IgE secretions in the splenocytes. Increased IL-2 level was observed in the culture supernatants of the splenocytes that were obtained from the mice administerd with the γ-irradiated alginate, while IL-4 level decreased. The present study indicates that γ-irradiated alginate can suppress allergy in a mouse allergy model.
90-Day Inhalation Toxicity of Dimethylamine in F344 Rats

Dimethylamine (DMA) is a widely used commodity chemical with few toxicity data. Groups of 10 male and female F-344 rats were exposed by inhalation to 0, 5, 10, 20, 40 and 80 ppm of DMA for 6 hrs/day, 5 days/week for 90 days. The changes of body weight, organ weight, hematology, clinical chemistry, and histopathological changes were evaluated after the exposure. As the results, the body weight was significantly decreased at 80 ppm in male and female rats (p<0.05). The absolute lung weight showed no statistically significant changes in any group. In contrast, the relative lung weight significantly increased at 80 ppm in male and female rats (p<0.05). Erythrocytes, mean cell hemoglobin, leukocytes, neutrophil, and platelet numbers were significantly increased in male and female at 40 or 80 ppm of DMA (p<0.05, p<0.01). In addition, the serum values of total protein, urea nitrogen were increased in male and creatine kinase, total protein were increased in female rats at 40 or 80 ppm (p<0.05, p<0.01). Histopathological examinations of the male and female lung samples showed slight hyperplasia and congestion at 80 ppm. Taken together, our study revealed that maximum tolerated dose of DMA would be over 40 ppm.

Inhalation toxicity, mutagenicity, and immunotoxicity tests were performed using a smoke generation system to investigate the safety of Herbrette, a tobacco substitute made with the leaves of Perilla frutescens. ICR mice were exposed to nicotine-free Herbrette smoke with concentrations of 0 (control), 4.08 $\pm$ 1.32 mg/$m^3$ (low dose), 7.72 $\pm$ 2.14 mg/$m^3$ (medium dose) and 12.83 $\pm$ 1.69 mg/$m^3$ (high dose) total particulate matters (TPM) for 4 weeks. When compared to the control group, the body weights, organ weights in the exposed groups did not show any significant differences. However, certain change of several serum chemical data and biochemical parameters were observed, however, the changes were within normal physiological ranges. Moreover, no changes in organ weight, and no gross/microscopic changes were observed between the exposed and control groups. Salmonella typhimurium reverse mutation, in vivo chromosomal aberration and micronucleus assays revealed that Herbrette did not induce mutagenicity. Upon evaluation of peripheral cellular immunity of mice through in vitro lymphocyte proliferation assay, no significant difference was observed in mean stimulation index between the exposed and control groups. Taken together, our results strongly suggest that Herbrette may not cause toxicity on mice under current condition.
식품 알레르기 마우스 모델에서 감태 물 추출물의 IgE 분비 억제 효과

OVA를 항원으로 한 식품알레르기 마우스 모델을 이용하여 감태 물 추출물의 알레르기 억제 효과를 확인하였다. 먼저, OVA로 면역한 마우스의 비장세포에 감태 물 추출물을 첨가하여 배양한 후, 배양 상층액의 IgE antibody와 IL-4 및 IFN-γ cytokine 분비량을 측정하였다. 그 결과, 10~100 ㎍/mL 농도의 감태 물 추출물 첨가로 비장세포의 IgE 분비량이 농도 의존적으로 감소하였고, IL-4 및 IFN-γ 분비량도 감태 물 추출물 첨가 농도가 증가할수록 감소하였다. 감태의 알레르기 억제능을 in vivo 실험을 통해 확인하기 위해 OVA로 알레르기를 유발한 마우스에 감태 물 추출물을 복강주사 한 후 혈청과 비장세포의 IgE 분비능을 확인하였다. 혈청의 항체 분비량 측정 결과, 1, 5 및 10 ㎎/㎏?BW 농도의 감태 물 추출물 투여군이 PBS 투여군에 비해 유의적으로 낮은 OVA-specific IgE 분비량을 보였다. 반면, IgG1과 IgG2a 분비량은 추출물 투여군과 PBS 투여군 사이에 유의적인 차이를 보이지 않았다. 또한 비장세포의 IgE 분비량이 5 및 10 ㎎/㎏?BW 농도의 감태 물 추출물의 복강주사에 의해 감소함을 확인하였다. 이상의 결과를 통해 감태 물추출물이 IgE 분비량의 유의적인 감소 효과를 통해 식품알레르기 반응을 효과적으로 억제할 수 있을 것으로 사료된다. This research was done to verify the effect of Ecklonia cava water extracts (ECWE) on inhibition of allergic reactions using ovalbumin (OVA)-immunized food allergy mouse model. For in vitro test, 10~100 ㎍/mL of ECWE and OVA were added to splenocytes obtained from OVA-immunized mice. The significant reduction of IgE antibody level in culture supernatants of splenocytes was shown in ECWE adding group at all tested concentrations. In addition, ECWE decreased IL-4 and IFN-γ levels in supernatants of splenocytes. To confirm the effect of ECWE in in vivo test, ECWE was injected to peritoneal cavity of OVA-immunized mice. Subsequently, IgE level was measured in serum and cultured supernatants of splenocytes. As a result, the injection of ECWE (5 and 10 ㎎/㎏?BW) significantly attenuated the secretion of IgE antibody in both serum and splenocytes. In conclusion, the present study indicates that ECWE could suppress in a food allergy mouse model through the inhibition of IgE secretion.
커피박 추출물이 간고등어의 저장성과 품질에 미치는 영향

커피박의 항산화능을 확인하고 커피박의 새로운 용도를 개발하고자 지질산화로 인한 선도저하가 문제시 되는 간고등어의 제조에 커피박 열수 추출물을 천연항산화제로 이용하였다. 먼저, 커피박 열수 추출물의 항산화 활성을 측정하기 위해 DPPH radical 소거능을 측정한 결과, 4~0.4 ㎎/mL 농도에서 약 92%의 높은 라디칼 소거능을 유지하였다. 항산화 활성을 확인한 커피박 열수 추출물 5, 10 및 15%를 염수에 첨가하여 간고등어를 제조한 후, 4℃에 15일간 저장하면서 산화도, VBN, pH 및 관능적 특성의 변화를 측정하였다. 그 결과, 15% 커피박 열수 추출물 처리구가 저장기간 내내 무처리구에 비해 낮은 산화도 값을 나타내었으며, VBN 함량은 10일까지는 시험구간의 차이를 보이지 않다가 급격한 산화가 시작된 저장 15일에 유의적으로 낮은 값을 보였다. pH는 저장기간이 증가할수록 증가하는 경향을 보였으나 실험구간의 유의적인 차이는 나타나지 않았다. 저장 0일과 5일에 관능평가를 실시한 결과, 향, 맛 그리고 전체적인 호감도 항목에서 10과 15% 커피박 열수 추출물 처리구가 높은 값을 보였다. 이상의 결과를 통해 15% 커피박 열수 추출물 처리를 통해 간고등어의 산화도가 억제됨을 확인하였으며 동시에 관능적 품질이 증진됨을 알 수 있었다. 따라서 커피박열수 추출물을 간고등어와 같이 빠른 지질산화로 인한 저장성 및 품질저하를 유발하는 식품에 천연 항산화제로써 사용할 수 있을 것으로 사료된다. To develop a novel application for roasted ground coffee residue, hot water extract of roasted ground coffee residue was used in manufacturing salted mackerels. First, DPPH radical scavenging effect of roasted ground coffee residue extract was measured. As a result, roasted ground coffee residue extract showed high activity of about 92% in the concentration of 0.4 to 4 ㎎/mL. Thereafter, the salted mackerel was treated with 5, 10 and 15% of roasted ground coffee residue extract and its shelf-life and quality was investigated. The salted mackerels treated with 15% roasted ground coffee residue extract showed significantly low TBARS and VBN as compared to the control. In sensory evaluation, the salted mackerels treated with 10 and 15% roasted ground coffee residue extracts scored higher as compared to the control. In conclusion, roasted ground coffee residue extract increased the shelf-life of salted mackerel and improved the sensual quality by inhibiting lipid oxidation.
