        The impacts of resveratrol on the retinal degeneration in a rat model of retinitis pigmentosa induced by alkylation: an in-vivo study

        Upregulation of Sirtuin Type 1 (SIRT1), a nicotinamide adeninedinucleotide (NAD+)-dependentdeacetylase, has been proved to protect against ample ocular diseases, while its effect onretinitis pigmentosa (RP) has not been illustrated. The study was aimed to explore the impactsof resveratrol (RSV), a SIRT1 activator, on the photoreceptor degeneration in a rat model of RPinduced by N-methyl-N-nitrosourea (MNU), an alkylation. The rats were induced RP phenotypesvia the intraperitoneal injection of MNU. The electroretinogram was conducted and revealedthat RSV could not prevent the decline of retinal function in the RP rats. The optical coherencetomography (OCT) and the retinal histological examination were performed and showed thatthe reduced thickness of the outer nuclear layer (ONL) was not preserved by RSV intervention. The immunostaining technique was applied. Afther the MNU administration, the number of theapoptotic photoreceptors in the ONL throughout the retinasand the number of microglia cellspresent among the outer part throughout the retinas were not significantly reduced by RSV. Western blotting was also performed. The data showed that the level of SIRT1 protein wasdecreased after MNU administration, while RSV was not able to obviously alleviate thedownregulation. Our data together indicated that RSV was not able to rescue thephotoreceptor degeneration in the MNU-induced RP rats, which might be due to the MNUinducedconsumption of the NAD+.

        Enhancement of CO2 desorption using ultrasound and vacuum in water scrubbing biogas upgrading system

        Ultrasound and vacuum were respectively employed to enhance CO2 desorption in a water scrubbing biogas upgrading system. Results showed that incomplete CO2 desorption could cause a high CO2 content in the water and seriously affect the purity of the product gas. Vacuum had a strong enhancement effect on CO2 desorption. When a vacuum of 0.04MPa was used to enhance CO2 desorption, the amount of the stripping air could be reduced to 1/16- th of that without enhancement, indicating that vacuum could greatly enhance CO2 desorption and significantly decrease the amount of the stripping air, which was expected to reduce a large amount of energy consumption. In contrast, the enhancement effect of ultrasound was not so obvious for CO2 desorption in the desorption column with air stripping, since the solution could be well desorbed by gas stripping, though ultrasound could strongly affect the static CO2 desorption.

      • Study on the pharmacological effects of Asini Corii Colla

        In this paper, 1H-NMR metabolomics technique was used to hydrolyse Asini Corii Colla acid from five different manufacturers, such as Asini Corii Colla, and the obtained 1H-NMR spectra were identified by chemical composition, and combined with relevant software for multivariate statistical analysis. Find the difference in chemical composition. It was found that 17 chemical components were identified from the Asini Corii Colla 1H-NMR spectrum; a comparison was made between Asini Corii Colla produced by A, B, C, and D manufacturers and Asini Corii Colla produced by Dong'e Ejiao manufacturer. The corresponding differential components mainly include isoleucine, hydroxyproline, arginine, leucine, phenylalanine, valine, lysine and clavulanic acid. The results showed that the nutritional composition of Asini Corii Colla products was higher, and 1H-NMR method has a good application prospect for the composition pharmacology and quality analysis of Asini Corii Colla.

        Identification of microRNAs involved in drought stress responses in early-maturing cotton by high-throughput sequencing

        Drought stress is one of the most important abiotic stresses. Cotton is classified as drought tolerant crop but the regulatory mechanism is unknown. MicroRNAs (miRNAs) have been implicated important roles in stress responses in many plants. However, the study of miRNAs in cotton responsive to drought stress is limited, especially in early-maturing cotton. In this study, we performed deep sequencing of small RNAs to identify known and novel miRNAs involved in the regulation of drought stress and understand the expression profile of miRNAs in early-maturing cotton. Three cotton small RNA libraries: non-stressed Shizao1 (early-maturing cotton variety) library (NSS), drought-stressed Shizao1 library (DSS) and non-stressed Jimian958 (medium-maturing cotton variety) library (NSJ) were constructed for deep sequencing. As a result, we identified a total of 64 known and 67 novel miRNAs in the 3 libraries and 88 of them were dramatically differentially expressed (greater than twofold) during drought stress. In addition, we found the expression of 41 miRNAs increased or reduced more than twofold in early-maturing cotton variety compared with that in medium-maturing cotton variety. Our results significantly increased the number of miRNAs in cotton and revealed for the first time the expression profile of miRNAs for early-maturing cotton.

        FAM46B inhibits cell proliferation and cell cycle progression in prostate cancer through ubiquitination of β-catenin

        FAM46B is a member of the family with sequence similarity 46. Little is known about the expression and functional role (s) of FAM46B in prostate cancer (PC). In this study, the expression of FAM46B expression in The Cancer Genome Atlas, GSE55945, and an independent hospital database was measured by bioinformatics and real-time PCR analysis. After PC cells were transfected with siRNA or a recombinant vector in the absence or presence of a β-catenin signaling inhibitor (XAV-939), the expression levels of FAM46B, C-myc, Cyclin D1, and β-catenin were measured by western blot and realtime PCR. Cell cycle progression and cell proliferation were measured by flow cytometry and the CCK-8 assay. The effects of FAM46B on tumor growth and protein expression in nude mice with PC tumor xenografts were also measured. Our results showed that FAM46B was downregulated but that β-catenin was upregulated in patients with PC. FAM46B silencing promoted cell proliferation and cell cycle progression in PC, which were abrogated by XAV-939. Moreover, FAM46B overexpression inhibited PC cell cycle progression and cell proliferation in vitro and tumor growth in vivo. FAM46B silencing promoted β-catenin protein expression through the inhibition of β-catenin ubiquitination. Our data clearly show that FAM46B inhibits cell proliferation and cell cycle progression in PC through ubiquitination of β-catenin.

        Effect of Phosphodiesterase-5 Inhibitors on the Treatment of Male Infertility: A Systematic Review and Meta-Analysis

        Purpose: Male infertility is a worldwide problem with limitations in the treatment. Phosphodiesterase-5 inhibitors (PDE5is) is the first choice for the treatment of erectile dysfunction, more and more studies show that it has a certain effect on male infertility in recent years. But there was currently no high quality of systematic review to evaluate the effects of PDE5is on semen quality. Materials and Methods: We retrieved the electronic databases of MEDLINE, PubMed, Web of Science, EMBASE, etc. Related randomized controlled trials (RCTs) were collected and selected up to May 20, 2020. We have searched literature with terms “male infertility”, “phosphodiesterase-5 inhibitors”, “PDE5i”, “Tadalafil”, “Sildenafil”, “Vardenafil”, “Udenafil”, “Avanafil”, “semen”, and “sperm”. Mean value and its standard deviation were used to perform quantitative analysis. All statistical analyses were conducted by RevMan 5.3 and Stata software. Results: There were a total of 1,121 participants in the nine included studies. There was a statistically significant improvement treated with PDE5is compared with sham therapy, which including sperm concentration (mean difference [MD]=1.96, 95% confidence interval [CI]=1.70–2.21, p<0.001; MD=3.22, 95% CI=1.96–4.48, p<0.001), straight progressive motility (%) Grade A (MD=3.71, 95% CI=2.21–5.20, p<0.001), sperm motility (MD=8.09, 95% CI=7.83–8.36, p<0.001), morphologically normal spermatozoa (%) (MD=0.67, 95% CI=0.20–1.15, p=0.005; MD=1.27, 95% CI=0.02–2.52, p=0.05), sperm abnormalities (%) (MD=-0.64, 95% CI=-0.81–-0.47, p<0.001), and progressive motile sperm (MD=5.34, 95% CI=3.87–6.81, p<0.001). Conclusions: In this meta-analysis of nine RCTs, treatment with PDE5is could improve some indicators of male sperm.
