        Development of a Novel, Anti-idiotypic Monoclonal Anti-prolactin Antibody That Mimics the Physiological Functions of Prolactin

        In this work, we prepared a panel of monoclonal anti-idiotypic antibodies to ovine prolactin (oPRL) by the hybridoma technique. Among these antibodies, one anti-idotypic antibody (designated B7) was chosen for further characterization by a series of experiments. We first demonstrated that B7 behaved as a typical $Ab2{\beta}$ based on a series of enzyme-linked immunosorbent assays. Subsequently, the results of a competitive receptor-binding assay confirmed that B7 could specifically bind to the prolactin receptor (PRLR) expressed on target cells. Finally, we examined its biological activities in CHO-PRLR and Nb2 cells and observed that B7 could activate Janus kinase 2-signal transducer and activator of transcription signalling in CHO-PRLR and Nb2 cells and induce BaF3 proliferation. The present study suggests that i) B7 can serve as a PRLR agonist or PRL mimic and has potential applications in regulating mammary gland development, milk production and maintenance of lactation in domestic animals and ii) B7 may be a biological reagent that can be used to explore the mechanism of PRLR-mediated intracellular signalling.

        RING Finger Proteins Are Involved in the Progression of Barrett Esophagus to Esophageal Adenocarcinoma: A Preliminary Study

        Background/Aims: To investigate the differential expression of RING finger (RNF) proteins in Barrett esophagus (BE) and esophageal adenocarcinoma (EAC). Methods: The differential expression of RNFs in normal esophagus (NE), BE, and EAC was screened using microarray assay. Real-time quantitative polymerase chain reaction (PCR), tissue microarray assay, and Western blot analysis were independently performed to detect the mRNA and protein expression of screened RNFs. Results: The expression of nine RNFs in the BE or EAC was 2-fold higher than those in NE. Among these proteins, the RNF32 and RNF121 expression in BE was 20.3-fold and 16.4-fold higher, respectively, than that in NE, and the expression of RNF24, RNF130, RNF141, RNF139, RNF11, RNF14, and RNF159 was upregulated more than 2-fold compared with NE. The expression of nine RNFs was not only upregulated in the EAC but was also positively related to the RNF expression in BE. The PCR results also indicated increased expression of these RNFs in BE and EAC compared to NE. Furthermore, the mRNA expression of all RNFs, except for RNF141 in EAC, was dramatically higher than those in the BE. Similar results were also obtained from the Western blot analysis. Conclusions: A total of nine RNFs play critical roles in the progression of BE to EAC. (Gut Liver 2014;8:487-494)

        Involvement of Alternative Oxidase in the Regulation of Sensitivity of Sclerotinia sclerotiorum to the Fungicides Azoxystrobin and Procymidone

        Sclerotinia sclerotiorum is a filamentous fungal pathogen that can infect many economically important crops and vegetables. Alternative oxidase is the terminal oxidase of the alternative respiratory pathway in fungal mitochondria. The function of alternative oxidase was investigated in the regulation of sensitivity of S. sclerotiorum to two commercial fungicides, azoxystrobin and procymidone which have different fungitoxic mechanisms. Two isolates of S. sclerotiorum were sensitive to both fungicides. Application of salicylhydroxamic acid, a specific inhibitor of alternative oxidase, significantly increased the values of effective concentration causing 50% mycelial growth inhibition (EC50) of azoxystrobin to both S. sclerotiorum isolates, whereas notably decreased the EC50 values of procymidone. In mycelial respiration assay azoxystrobin displayed immediate inhibitory effect on cytochrome pathway capacity, but had no immediate effect on alternative pathway capacity. In contrast, procymidone showed no immediate impact on capacities of both cytochrome and alternative pathways in the mycelia. However, alternative oxidase encoding gene (aox) transcript and protein levels, alternative respiration pathway capacity of the mycelia were obviously increased by pre-treatment for 24 h with both azoxystrobin and procymidone. These results indicate that alternative oxidase was involved in the regulation of sensitivity of S. sclerotiorum to the fungicides azoxystrobin and procymidone, and that both fungicides could affect aox gene expression and the alternative respiration pathway capacity development in mycelia of this fungal pathogen.

        Log-Average-SNR Ratio and Cooperative Spectrum Sensing

        In this paper, we analyze the spectrum-sensing performanceof a cooperative cognitive radio (CR) network consisting ofa number of CR nodes and a fusion center (FC). We introduce the“log-average-SNR ratio” that relates the average SNR of the CRnode-FC link and that of the primary-user-CR-node link. Assumingthat the FC utilizes theK-out-of-N rule as its decision rule, wederive exact expressions for the sensing gain and the coding gain— parameters used to characterize the CR network performanceat the high SNR region. Based on these results, we determine waysto optimize the performance of the CR network.

      • Activation of Neuronal Nitric Oxide Synthase by M2 Muscarinic Receptors Associated with a Small Increase in Intracellular Calcium

        We investigated the coupling of the M2 muscarinic acetylcholine receptors expressed in Chinese hamster ovary cells to activation of neuronal nitric oxide(NO) synthase. Stimulation of guanylate cyclase activity in detector neuroblastoma cells was used as an indirect measure of the generation of NO in Chinese hamster ovary cells. The muscarinic agonist carbachol induced marked time-and concentration-dependent enhancement of the activity of NO synthase. Activation of neuronal NO synthase by M2 muscarinic receptors was associated with a small increase in the concentration of intracellular Ca^2+. These data suggest the presence of alternate mechanisms of activation of neuronal NO synthase which might be operative in the absence of large changes in the concentration of cellular Ca^2+. These findings help to understand the mechanisms of activation of NO synthase.

      • A Systematic Review of Risk Factors for Brain Metastases and Value of Prophylactic Cranial Irradiation in Non-Small Cell Lung Cancer

        Background: The incidence of brain metastases (BM) varies in patients with non-small cell lung cancer (NSCLC), calls into question the value of prophylactic cranial irradiation (PCI). It is possible that clinicopathologic characteristics are associated with the development of BM, but these have yet to be identified in detail. Thus, we conducted the present meta-analysis on risk factors for BM and the value of PCI in patients with NSCLC. Methods: Eligible data were extracted and the risk factors for BM and the value of PCI in patients with NSCLC were analyzed by calculating the pooled odds ratio (OR). Heterogeneity was detected using Q and I-squared statistics, and publication bias was tested by funnel plots and Egger's test. Results: Six randomized controlled trials with a focus on the value of PCI and 13 eligible studies with a focus on risk factors for BM were included. PCI significantly reduced the incidence of BM in patients with NSCLC (p=0.000, pooled OR=0.34, 95% confidence interval = 0.37-0.59). Compared with non-squamous cell carcinoma, squamous cell carcinoma was associated with a low incidence of BM in patients with NSCLC (p=0.000, pooled OR=0.47, 95% confidence interval =0.34-0.65). The funnel plot and Egger's test suggested that there was no publication bias in the current meta-analysis. Conclusions: This meta-analysis provides statistical evidence that compared with non-squamous cell carcinoma, squamous cell carcinoma can be used as a predictor for BM in patients with NSCLC, and PCI might reduce the incidence of BM in patients with NSCLC, but does not provide a survival benefit.

        Effects of annealing holding time on capacitance performance of RuO2–IrO2–graphene/Ti electrodes

        The thermal decomposition method was used to prepare composite electrodes of the Ruthenium oxide–Iridium oxide–Graphene (RuO2–IrO2–G). Scanning Electron Microscopy (SEM), X–ray diffraction analysis (XRD), and electrochemical tests were used to study the influence of different annealing holding time on the surface morphology, phase composition, and capacitive performance of the coatings. The results showed that more and more RuO2, IrO2 nanoparticles were observed on the surface and cracks of the coating as the annealing holding time increasing. The RuO2–IrO2–G/Ti electrode was obtained by annealing for 5 h. The coating of the electrode consists of a certain amount of amorphous phase and nano–crystalline phase, and it had good electronic conductivity and ionic conductivity. At the same time, the electrode was prepared at 5 h had the largest specific capacitance of 778.46 F/g, which increased by 430.89 F/g than the electrode was prepared at 1 h. In addition, the electrode also had superior capacitance performance, capacitance retention and power characteristics.

        중국 쯔보시의 도시 거버넌스 연구

        도시화의 지속적인 발전에 따라 현대의 도시 사회는 커다란 기회와 도전에 직면해 있다. 도시 거버넌스는 다원적인 도시개발과 관리과정으로서 도시의 경쟁력, 지속가능성 및 주민의 삶에 큰 영향을 미치고 있다. 본 연구는 성공적인 도시 거버넌스의 사례로 평가받고 있는 중국 쯔보시의 사례에 대한 분석을 통해서 현대 도시 거버넌스에서 로컬 거버넌스 이론의 핵심적인 역할을 고찰하였다. 기존의 연구들은 쯔보시의 도시 거버넌스가 도시관리 수준의 향상, 도시 브랜드의 구축, 사회참여 및 사회적 자본 형성 등에 있어서 성과를 거둔 것으로 분석하였다. 본 연구는 쯔보시의 도시 거버넌스 사례의 주요 주체를 정부, 사회단체, 기업, 시민 등으로 설정하고, 주요 주체들 간의 상호작용에 대해서 다각적으로 분석하였다. 분석을 통해서 쯔보시의 도시 거버넌스 구조는 다원화, 협력, 네트워크 등의 특징을 가지고 있으며, 정부와 다른 주체들 간에 효율적인 협력관계가 형성되어 있다는 것을 알 수 있었다. 또한 쯔보시의 도시 거버넌스 구축의 핵심 요소는 다음과 같다. 첫째, 주민과 관련 기관의 적극적인 참여. 둘째, 참여주체 간 상호신뢰를 바탕으로 한 정보의 교환. 셋째, 수평적 파트너십 구축을 통한 효과적인 협력 네트워크. 쯔보시 도시 거버넌스는 바비큐 도시 브랜드 구축 과정에서 발생한 다양한 문제 해결에 있어서 중요한 역할을 하였다. With the continued development of urbanization, modern urban society faces great opportunities and challenges. Urban governance is a pluralistic urban development and management process that has a significant impact on the competitiveness and sustainability of cities and the lives of residents. This study examined the key role of local governance theory in modern urban governance through an analysis of the case of Zibo City, China, which is evaluated as an example of successful urban governance. Existing studies analyzed that Zibo City's urban governance achieved results in improving the level of urban management, building a city brand, social participation, and forming social capital. This study set the main subjects of Zibo City's urban governance case as the government, social organizations, companies, and citizens, and analyzed the interactions between the main subjects from various angles. Through the analysis, it was found that Zibo City's urban governance structure has characteristics such as pluralism, cooperation, and networks, and that efficient cooperative relationships are formed between the government and other entities. Additionally, the key elements of building urban governance in Zibo City are as follows: First, active participation of residents and related organizations. Second, the exchange of information is based on mutual trust between participating entities. Third, an effective cooperation network through the establishment of horizontal partnerships. Zibo City's urban governance played an important role in resolving various problems that arose in the process of building the Barbecue City brand.
