      • HCV, Acute, LT : A Disparate Subset of Double Negative T cells Contributes to the Outcome of Murine Fulminant Viral Hepatitis via Effector Molecule Fibrinogen-like Protein 2

        Aims: The underlying pathogenesis of fulminant viral hepatitis (FVH) has not been fully elucidated. As a subset of regulatory T cells, CD3+CD4-CD8- double negative (DN) T cells can suppress T cell responses. In this study, we present new insights into the immune mediated mechanisms involved in FVH caused by murine hepatitis virus strain 3 (MHV-3). Methods: The phenotype and cytokines of DN T cells were detected by flow cytometric analysis. The levels of mfgl2 were measured by real-time PCR and western-blot. The function of mfgl2 was measured by PCA. Results: After MHV-3 infection, the proportions of DN T cells increased significantly in BALB/cJ mice, and splenic DN T cells expressing high levels of CD69 were recruited by MHV-3 infected hepatocytes to the liver. Serum levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST) and total bilirubin (TBil) increased, accompanied by massive hepatocyte necrosis. These DN T cells were predominantly consisted of a TCRαβ+ subset expressing high levels of CD44, and did not produce cytokine except IL-2. Adoptive transfer of this subset of DN T cells to the MHV-3 infected mice resulted in an increase of murine fibrinogen-like protein 2 (mfgl2) expression in association with massive fibrin deposition in the liver. Following MHV-3 infection, membrane mfgl2 expression and functional procoagulant activity (PCA) increased remarkably in the DN T cells. Introduction of a recombinant adenovirus which encoded a microRNA specifically targeting mfgl2 gene (Ad-mfgl2-miRNA) in vivo significantly inhibited the hepatic expression of mfgl2, increased mice survival. However, under this condition, adoptive transfer of the DN T cells accelerated the disease progression and reversed the benefit from mfgl2 gene silence, led to a 100% death. Conclutions: Our results demonstrated that DN T cell-derived mfgl2 may serve as an important effector molecule contributing to the pathogenesis of MHV-3-induced FVH.

        Preliminary numerical study on hydrogen distribution characteristics in the process that flow regime transits from jet to buoyancy plume in time and space

        Hydrogen-steam gas mixture may be injected into containment with flow regime varying both spatiallyand transiently due to wall effect and pressure difference between primary loop and containment insevere accidents induced by loss of coolant accident. Preliminary CFD analysis is conducted to gain informationabout the helium flow regime transition process from jet to buoyancy plume for forthcomingexperimental study. Physical models of impinging jet and wall condensation are validated using separatedeffect experimental data, firstly. Then helium transportation is analyzed with the effect of jetmomentum, buoyancy and wall cooling discussed. Result shows that helium distribution is totallydominated by impinging jet in the beginning, high concentration appears near gas source and wallwhere jet momentum is strong. With the jet weakening, stable light gas layer without recirculating eddyis established by buoyancy. Transient reversed helium distribution appears due to natural convectionresulted from wall cooling, which delays the stratification. It is necessary to concern about hydrogenaccumulation in lower space under the containment external cooling strategy. From the perspective ofexperiment design, measurement point should be set at the height of connecting pipe and near the wallfor stratification stability criterion and impinging jet modelling validation.

        Discrimination of Camellia seed oils extracted by supercritical CO2 using electronic tongue technology

        Analytical method which combines electronictongue technique and chemometrics analysis is developedto discriminate oil types and predict oil quality. All thestudied Camellia oil samples from pressing, n-hexaneextraction and supercritical CO2 extraction (SCCE), weresuccessfully identified by principal component analysis(PCA) and hierarchical cluster analysis (HCA). Furthermore,multi factor linear regression model (MLRM) wasestablished to predict oil quality, which are indicated byacid value (AV) and peroxide value (POV). The practicalpotential of e-tongue for the discrimination and assessmentof Camellia oils has shown promising application in thecharacterization of Camellia oils in the oil qualityevaluation.

        Gap junction blockage promotes cadmium-induced apoptosis in BRL 3A derived from Buffalo rat liver cells

        Gap junctions mediate direct communication between cells; however, toxicological cascade triggered by nonessential metals can abrogate cellular signaling mediated by gap junctions. Although cadmium (Cd) is known to induce apoptosis in organs and tissues, the mechanisms that underlie gap junction activity in Cd-induced apoptosis in BRL 3A rat liver cells has yet to be established. In the present study, we showed that Cd treatment decreased the cell index (a measure of cellular electrical impedance) in BRL 3A cells. Mechanistically, we found that Cd exposure decreased expression of connexin 43 (Cx43), increased expression of p-Cx43 and elevated intracellular free Ca2+ concentration, corresponding to a decrease in gap junctional intercellular communication. Gap junction blockage pretreatment with 18b-glycyrrhizic acid (GA) promoted Cd-induced apoptosis, involving changes in expression of Bax, Bcl-2, caspase-3 and the mitochondrial transmembrane electrical potential (Δym). Additionally, GA was found to enhance ERK and p38 activation during Cd-induced activation of mitogen-activated protein kinases, but had no significant effect on JNK activation. Our results indicated the apoptosis-related proteins and the ERK and p38 signaling pathways may participate in gap junction blockage promoting Cd-induced apoptosis in BRL 3A cells.

      • THE INFLUENCE OF THE SHARING PLATFORM SOCIALIZATION ON THE CONSUMERS’ INTENTION TO PARTICIPATE - TRUST AS A MEDIATOR

        Introduction In order to solve the information asymmetry and make full use of resources (unused goods, spare capacity and so on), the sharing economy, which based on the application of mobile internet technology and focused on the consumption of the right to use, is developing rapidly. Lack of trust is considered to be one of the important problems hindering the development of sharing economy. M?hlmann (2015) said that trust is a fundamental determinant of sharing economy development and ensuring shared satisfaction. At present, sharing economy enterprises have established product evaluation system and docked a third party credit rating system (i.e. Zhima Credit) to promote consumers’ trust. However, with most enterprises are becoming more and more consistent in evaluation and credit rating system, there is a new problem emerged: how to further deepen consumers’ trust in order to promote consumers’ intention to participate continuously (Zucker, 1986; Lewis & Weigert, 1985). Granovetter (1985) mentioned that trust is the product of social relationships. Botsman and Rogers (2011) made it clear that social networks play an important role in building trust in sharing economy. And people are always more willing to accept the opinions of people who have social relations with them (Lu, Zhao, & Wang, 2010). Hence, with the feature of participant of everyone in sharing economy, it is necessary to further strengthen consumers’ trust through the social interaction among consumers and between consumers and sharing platforms. Theoretical Development Sharing economy is based on LBS and other new technologies to realize the matching of supply and demand information across time and space. And to a certain extent, it solves the problem of information asymmetry in the era of e-commerce. Yet consumer have new requirements for product information. For example, consumers are more concerned about whether the utility of products and services can meet their needs (i.e. some consumers are more concerned about when to reach their destination than the performance of the car). But it is difficult for company to understand each everyone of consumers’ utility preference and publish product utility information based on these preferences (Xie & Gerstner, 2007). Because this kind of utility preference has the characteristic of timeliness and personalization. Therefore, consumers’ perceived uncertainty about the utility of the product will also affect the consumers’ trust. In summary, we then address these research questions:(1) can the promotion of trust between consumers and sharing platforms be achieved through the socializing of sharing platforms and then affect consumers’ intention to participate continuously; (2) if so, which type of privacy protection method and social information can promote consumer trust; and (3) does the impact of sharing platform socialization on consumer trust vary according to consumers’ concerns about the perceived utility uncertainty of the product. Research Design We then address these issues using experiment data. The first experiment was a 2 (social media: have, not have) x 2 (individual trust propensity: trust, not trust) between-subject design on the intention to trust sharing platform and participant continuously. Perceived utility uncertainty was added to this design as a moderator. Moreover, the second experiment was a 2 (social information type: hedonic, utilitarian) x 2 (degree of privacy protection: week, strong) between-subject design on the intention to trust sharing platform. Our scales come from existing scales and optimize it according to the characteristics of sharing economy (Schneider, 1999; Dhar & Wertenbroch, 2000; McKnight, Choudhury, & Kacmar, 2002). Results and Conclusion The results of the first experiment show that whether or not the social platform has no significant impact on the consumer trust if the individual tends to trust others. And for individuals who tends to do not trust others, they are more likely to trust socialized sharing platforms. The higher the consumer perceived utility uncertainty is, the higher (lower) trust of the consumer platform in the socialized (not socialized) sharing platform. For the second experiment results, in the context of sharing economy, the utilitarian information has a greater positive impact on the consumer trust than the hedonic information. In addition, the stronger the privacy protection of sharing platforms, the higher the trust of consumes. This study makes several academic contributions. First, we extend the topic of socialization to sharing economy context. Second, we add the perceived utility uncertainty as a moderator, and supplement the role of perceived uncertainty. This research also provides several practical implications. First, sharing platform can promote their consumers’ trust by establishing their own social platform. This platform can be used as a differentiated competitive strategy for sharing platform. Second, sharing platform should guide consumers to share utilitarian information (i.e. weather for specific place or real time traffic) to improve consumer satisfaction. Third, sharing platform can access consumer preference information through this social platform for further product development and classification.

        A bibliometric review on latent topics and trends of the empirical MOOC literature (2008–2019)

        Massive Open Online Courses (MOOCs) have become a popular learning mode in recent years, especially since the outbreak of COVID-19 in late 2019, which had resulted in a signifcant increase in associated research. This paper presents a bibliometric review of 1078 peer-reviewed MOOC studies between 2008 and 2019. These papers are extracted from three infuential databases, the Web of Science (WOS), Scopus, and the Education Resources Information Center (ERIC). The MOOC literature analysis with a bibliometric approach identifed the research trends, journals, countries/regions, and institutions with high H-index, scientifc collaborations, research topics, topic distributions of the prolifc countries/regions and institutions, and annual topic distributions, after which the representative research and research implications were discussed. This review gives researchers a deep and comprehensive understanding of current MOOC research and identifes potential research topics and collaborative partners, which supports MOOC-related future research.

        Temperature Dependence of the Optical Transmission of a Magnetic Fluid with an Applied Magnetic Field

        (Received 25 August 2008) The physical origin of the temperature dependence of the optical transmission of a magnetic fluid under an applied magnetic field is studied in this paper When a magnetic field is applied parallel to the plane of a magnetic fluid thin film, magnetic chains form in the same direction as the magnetic field, which suppresses the optical transmission. We observed that the optical transmission could be tuned by varying the ambient temperature of the magnetic fluid thin film. The design and the experimental results are also analyzed.
