The well-characterized gram-positive bacterium Bacillus subtilis is an outstanding industrial candidate for protein expression owing to its single membrane and high capacity of secretion,simplifying the downstream processing of secretory proteins. During the last few years, there has been continuous progress in the illustration of secretion mechanisms and application of this robust host in various fields of life science, such as enzyme production, feed additives,and food and pharmaceutical industries. Here, we review the developments of Bacillus subtilis as a highly promising expression system illuminating strong chemical- and temperatureinducible and other types of promoters, strategies for ribosome-binding-site utilization, and the novel approach of signal peptide selection. Furthermore, we outline the main steps of the Sec pathway and the relevant elements as well as their interactions. In addition, we introduce the latest discoveries of Tat-related complex structures and functions and the countless applications of this full-folded protein secretion pathway. This review also lists some of the current understandings of ATP-binding cassette transporters. According to the extensive knowledge on the genetic modification strategies and molecular biology of Bacillus subtilis, we propose some suggestions and strategies for improving the yield of intended productions. We expect this to promote striking future developments in the optimization and application of this bacterium.

To better understand users and create more personalised search experiences, a number of user models have been developed, usually based on different theories or empirical data study. After developing the user models, it is important to effectively utilise them in the design, development and evaluation of search systems to improve users’ overall search experiences. However there is a lack of research has been done on the utilisation of the user models especially theory-based models, because of the challenges on the utilization methodologies when applying the model to different search systems. This paper explores and states how to apply an Information Foraging Theory (IFT) based user classification model called ISE to effectively identify user’s search characteristics and create user groups, based on an empirically-driven methodology for content-based image retrieval (CBIR) systems and how the preferences of different user types inform the personalized design of the CBIR systems.

In December 2011 and July 2012, two sets of 840MVA hydro-generator of Three Gorges on Yangtze River with Close-loop-self-circulating evaporative cooling (CLSCEC) system were put into commercial operation. In this paper, we make engineering summary of these two generators with CLSCEC system. We also make a comparison between the internal water cooling (IWC) hydro-generator and the CLSCEC hydro-generator used in Three Gorges power plant in fields of their operating characteristics, working performances, technical features, working safety and reliability. In addition, engineering structures, type tests' results and systematic emulating calculation of CLSCEC schemes are analyzed.

Coronavirus disease 2019 (COVID-19) outbreak started in December 2019 that caused difficulties for clinical work. Practical work experience in our spinal outpatient and emergency department during the COVID-19 pandemic is summarized in this article, with combined evidence-based medical evidence to explore a standardized process of diagnosis and treatment for spinal diseases. Outpatient reservation, continuous screening, triage, and isolation, first consultation accountability system, pandemic reporting system, and online revisit were strictly followed. We hope that our experience in prevention and control of COVID-19 can help spine surgeons globally in stopping the spread of COVID-19. Spine surgeons should collaborate with infection control specialists to avoid cross-infection in hospitals and optimize treatment.

In this study, in the respect of rafting behavior of Ni-based single crystal (SC) superalloy under creep and thermal mechanicalfatigue (TMF), an image processing program is developed to investigate the probability characteristic of γ channel width, anda channel width evolution model considering non-quasi-static modification is proposed. Firstly, the fractured and interruptedtests are conducted on SC superalloy. The channel width evolution behavior under different load conditions is observed viascanning electron microscope. Then, an image processing program based on image binarization is developed to conductstatistics on channel width. The statistical results show that the stress and temperature have significant effects on raftingrate, while the phase difference has a small effect. Meanwhile, the channel width obeys lognormal distribution, and the meanvalues follow linear relationship with standard deviations. In the modeling part, a static model is established to predict themean value evolution of channel width under creep condition. To be generalized to TMF condition, the model is modifiedby considering the non-quasi-static effect. After that, the channel width evolution model is further derived to describe theprobability density distribution of channel width. The predicted channel width evolutions are in good agreement with theexperimental results. The statistical results and models can provide the basis for multiscale modeling of SC superalloy.

Three crystal phases of BiPO4 (HBIP, nMBIP and mMBIP), corresponding photocatalysts (Er3+:Y3Al5O12/HBIP, Er3+:Y3Al5O12/nMBIP and Er3+:Y3Al5O12/mMBIP) and their nanocomposite photocatalyst (Er3+:Y3Al5O12/(H-nM-mM)BIP) were prepared by hydrothermal, ultrasonic dispersion and liquid boilingmethods. The prepared photocatalysts were characterized by X-ray diffractometer (XRD), energydispersive X-ray spectroscopy (EDX), scanning electron microscopy (SEM) and UV–vis diffuse reflectancespectra (DRS). The catalytic activity of prepared photocatalysts was evaluated via photocatalyticconversion rate of nitrite under ultraviolet-light, visible-light and simulated solar-light irradiations. These three photocatalysts all exhibited excellent performance under simulated solar-light irradiationand reached 85.36%, 84.42% and 78.53% conversion rates, respectively, for Er3+:Y3Al5O12/mMBIP, Er3+:Y3Al5O12/nMBIP and Er3+:Y3Al5O12/HBIP. Particularly, Er3+:Y3Al5O12/(H-nM-mM)BIP was also found toplay a high catalytic activity, resulting in 78.68%, 70.04% and 82.57% conversion rates, respectively, underultraviolet-light, visible-light and simulated solar-light irradiation. In addition, the study of used timesshowed that the prepared photocatalysts can be effectively recycled without an apparent inactivation onthe photocatalytic activity. This research may provide a potential way for converting nitrite and othercontaminants by utilizing solar energy efficiently.
