This study was conducted to obtain the transformed tobacco (Nicotiana tubacum) plants with cytosolic ascorbate peroxidase gene(ApxSC7) using Agrobacterium tumefaciens LBA4404. A cDNA encoding the cytosolic ascorbate peroxidase of strawberry, ApxSC7, was introduced into tobacco plants via Agrobacterium-mediated gene transfer system. The expression vector, pIG-AP8, harboring ApxSC7 gene was used for production of transgenic tobacco plants. A large number of transgenic plants were regenerated on a medium containing hygromycin. Integration of ApxSC7 gene was confirmed by PCR and Southern blot analyses with genomic DNA. Northern blot analyses revealed that the pIGap8 gene was constitutively expressed.

Comparison results of callus formation ratio from seed explants, callus sizes, regeneration ratios from callus and regeneration efficiency [calculated by following formular; callus formation ratio(%) × regeneration ratio(%)/100]for 27 ochardgrass (Dactylis glomerata L.)varieties imported and Hapsung 2 developed in Korea are as follows; 1. Among ochardgrass varieties showing more than 50% callus formation ratios, the descending order of callus formation ratio after bedding the seed explants for 4 weeks was 93>Sparta>Pizza>Condor>Lidaglo>Glorus>Hapsung2>Frode. 2. The callus sizes after bedding for 4 weeks were in the range of φ 0.43cm~4.2cm in which there was 10 times size difference between the largest one and the smallest one but most of them were between φ2.5cm~4cm. 3. The regeneration ratio from callus among varieties were in the range of 0~36% and descending order of the upper 6 varieties was Plano>Akimidori>Justus>Lidacta>Currie>Hall mark. 4. The regeneration efficiency which is calculated by the ratios of regeneration from seed explant numbers was between 0 to 17.4% among which Justus showed the highest value in the 4-week treatment. 5. The correlation between callus formation ratios and the callus sizes, callus formation ratios and regeneration efficiency, and callus sizes and regeneration efficiency were r=0.5765, r=0.6365 and r=0.6246, respectively in 4-week callus and all the correlations were significant on the 1% level. 6. In 6-week callus, the descending order callus formation ratios from seed sxplants for the best 6 varieties was Condor>Sparta>93M>Justus>Potomac>Lidaglo>Frode. 7. The callus sizes formed were between φ1.5~5.7cm in which Sparta, the largest one of φ5.7cm was five times larger than the smallest one. The callus size of the control variety. Hapsung 2 was φ3.8cm, which belonged to larger size. 8. Regeneration ratio showed a great deviation among varieties from 6-week old calli by showing from 0% to 100% in which all the calli were regenerated in Plano while no callus was regenerated in Juno, 9. The range of regeneration effciency was between 0~28% among varieties in which the values from 6-week callus treatment were larger than those from 4-week callus treatment. Especially, the value of Potomac in 6-week was 3 times larger than that in 4-week. 10. The correlation between callus formation ratios and the callus sizes, callus formation ratios and regeneration efficiency, and callus sizes and regeneration effciency were r=0.8369, r=0.6683 and r=0.5937, respectively in 6-week callus, and all the correlations were significant on the 1% level.

Comparisons of callus formation ratios from seed explants, callus sizes, regeneration ratios from callus and regeneration efficiency for 4 orchardgrass (Dactylis glomerata L.) varieties (Three were developed in Korea and one was imported from foreign nation) are as follows; Jangbeol 102 (67.0%) has the highest callus formation ratio in 4 weeks incubated callus after bedding the seed explants, but Potomac (68.4%) has the highest ratio in 6-seek callus. Potomac (3.93cm) has the highest callus size in 4-week callus, but Jangbeol 101 (4.32cm) has the highest size in 6-week callus. Jangbeol 101 (17.7%) has the highest plant regeneration ratio in 4-week callus, but Potomac (37.4%) has the highest raion in 6-week callus. Jangbeol 102 (11.5%) has the highest plant regeneration efficiency in 4-week callus, but Potomac (25.6%) has the highest efficiency in 6-week callus.
Antioxidant Effect and Functional Properties of Hydrolysates Derived from Egg-White Protein

This study utilized commercially available proteolytic enzymes to prepare egg-white protein hydrolysates (EPHs) with different degrees of hydrolysis. The antioxidant effect and functionalities of the resultant products were then investigated. Treatment with Neutrase yielded the most α-amino groups (6.52 mg/mL). Alcalase, Flavourzyme, Protamex, and Ficin showed similar degrees of α-amino group liberation (3.19-3.62 mg/mL). Neutrase treatment also resulted in the highest degree of hydrolysis (23.4%). Alcalase and Ficin treatment resulted in similar degrees of hydrolysis. All hydrolysates, except for the Flavourzyme hydrolysate, had greater radical scavenging activity than the control. The Neutrase hydrolysate showed the highest 2,2-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging activity (IC50=3.6 mg/mL). Therefore, Neutrase was identified as the optimal enzyme for hydrolyzing egg-white protein to yield antioxidant peptides. During Neutrase hydrolysis, the reaction rate was rapid over the first 4 h, and then subsequently declined. The IC50 value was lowest after the first hour (2.99 mg/mL). The emulsifying activity index (EAI) of EPH treated with Neutrase decreased, as the pH decreased. The EPH foaming capacity was maximal at pH 3.6, and decreased at an alkaline pH. Digestion resulted in significantly higher 1,1-diphenyl-2-picrylhydrazyl (DPPH) and ABTS radical scavenging activity. The active peptides released from egg-white protein showed antioxidative activities on ABTS and DHHP radical. Thus, this approach may be useful for the preparation of potent antioxidant products.
경남 지역내 소 Neospora caninum에 대한 감염률 조사

This survey was carried out to investigate the seroprevalence of antibodies to Neospora caninum in Korean indigenous cattle that was raised in central area province Gyeongnam, Korea. A total of 719 sera were tested for N. caninum antibodies using ELISA (Herdcheck anti-Neospora, IDEXX Laboratories Inc., Westbrook, Maine USA). Seroprevalence of individual and farm were 29.8%(214/719) and 53.2%(50/94). Regional seropositive rates of the samples were 61%(47/77), 23.3%(7/30), 13.8%(49/355), 37.6%(77/205), 65.4%(34/52) at Changwon, Jinhae, Gimhae, Miryang, Yangsan, respectively. It showed difference at the age and on the herd size of farms. A herd of cattle above the age of 5 was more infective than under 4 years. And in seroprevalence by herd size farms having under 30 heads was top(35.7%). Seropositive 214 herds of N. caninum antibidies were tested for brucellosis by test tube. Positive rate of double infection was 16.4%(35/214).

This study was conducted to evaluate the alleviative effects of activated charcoal (AC) and Houttuynia cordata (HC) singly or in combination in broiler chickens during aflatoxicosis. Activated charcoal (1% or 0.5%) and H. cordata (1% or 0.5%) were mixed into the diets for the ability to reduced the deleterious effects of 2.4mg total aflatoxin (AFB1) kg-1 diet on growing broiler chickens from 1 to 21 days of age. A total of 160 1-day-old (Hyline Variety Brown) broiler chicks were housed in eight treatment groups [Control, AFB1, AC 1%, HC 1%, AFB1 plus AC 1% plus HC 1%, AFB1 plus AC 1% plus HC 0.5%, AFB1 plus AC 0.5% plus HC 1%, AFB1 plus AC 0.5% plus HC 0.5%] each consisting of 20 chicks. Compared to control, 2.4mg AFB1 alone treatment group significantly decreased body weight gains of chickens. The addition of mixed AC 1% and HC 1% including 6, 7 groups to the 2.4mg AFB1-containing diet moderately reduced the adverse effects of AFB1 on performances of chickens. The chickens consuming 2.4mg AFB1 plus AC 0.5% and HC 0.5%-containing diet showed very slightly reduced the adverse effects on investigated parameters compared to the AFB1 only treated group. Also, the single addition of AC or HC to the AFB1-free diet had no adverse effects in chickens. These results suggest that AC and HC mixed can reduced the aflatoxicosis in broilers and may be contribute to a solution of the aflatoxicosis problem in poultry production.

Parkinson’s disease (PD) is a neurodegenerative disability caused by a decrease of dopaminergic neurons in the substantia nigra (SN). Although the etiology of PD is not clear, oxidative stress is believed to lead to PD. Catalase is antioxidant enzyme which plays an active role in cells as a reactive oxygen species (ROS) scavenger. Thus, we investigated whether PEP-1-Catalase protects against 1-methyl-4-phenylpyridinium (MPP+) induced SH-SY5Y neuronal cell death and in a 1-methyl- 4-phenyl-1,2,3,6-trtrahydropyridine (MPTP) induced PD animal model. PEP-1-Catalase transduced into SH-SY5Y cells significantly protecting them against MPP+-induced death by decreasing ROS and regulating cellular survival signals including Akt, Bax, Bcl-2, and p38. Immunohistochemical analysis showed that transduced PEP-1-Catalase markedly protected against neuronal cell death in the SN in the PD animal model. Our results indicate that PEP-1-Catalase may have potential as a therapeutic agent for PD and other oxidative stress related diseases. [BMB Reports 2015; 48(7): 395-400]
경요도적 전립선 절제술시 혈청 소디움 및 삼투질 농도에 미치는 Furosemide 의 효과

This study was undertaken to compare the effects of furosemide on the serum concentration of sodium and osmolality after transurethral prostatic resection(TURP) using cytal solution, and to determine the adequate time of administration of frurosemide. At the end of prostatic resection, 15 patients were allocated randomly to receive furosemide (furosemide group) and were compared with 15 patients without administration of furosemide (control group). There was no difference in mean serum concentation of sodium between two groups. Serum osmolality in furosemide group was significantly increased as compared with control group one hour after operation. So cytal solution used during staged TURP and short operation within one hour do not affect serum corcentration of sodium and administration of furosemide is not associated with a change in serum concentration of sodium. But furosemide meaningfully increases the serum osmolality and it is more effective to administer it with administration at the end of prostatic reseetion.
Microwave absorption of poly‐<i>N</i>‐vinylcarbazole–polyaniline composites

<P><B>Abstract</B></P><P>Poly‐<I>N</I>‐vinylcarbazole–polyaniline (PANI) composites were synthesized using different loading concentration of aniline (0.025–0.1 M) for their microwave absorption characteristics. The obtained composites were studied by Fourier transform infrared spectroscopy, thermogravimetric analysis technique, and atomic force microscope for their chemical structure, thermal stability, and the surface modifications, respectively. The conductivity increased much with the increase of aniline concentration in the composites. The composite sheets exhibited a strong microwave absorption in the microwave range of 1–10 GHz and achieved a maximum absorption value of 33 dB. The position of absorbing peak shows a mixed trend moving from lower to higher and again to lower with an increasing the concentration of aniline in the poly‐<I>N</I>‐vinylcarbazole–PANI. The new polymer composite exhibited an appreciable electromagnetic interference shielding efficiency compared with the previously reported PANI composites. POLYM. ENG.
