Bacillus subtilis K-1에 의한 biopolymer의 생산과 levansucrase활성의 최적배양조건

In order to study on the biopolymer production and the changes of its molecular composition, optimal culture condition for biopolymer production, levansucrase activity increase using identified strain and molecular structure and conformation of produced biopolymer were investigated. The mutants which was improved production ability of biopolymer and changed levansucrase activity were derived. The optimal culture conditions for bioplymer production and levansucrase activity increase were sucrose 6%, M.S.G. 2%, NaH₂P0₄·2H₂O 0.3%, MgSO₄·7H₂O 0.05%, biotin 0.1㎍/㎖ at pH 6.0, 37℃ for 48hours. Under this conditions, biopolymer production and levansucrase activity were about 25g/ℓ, 830 unit respectively. Among the several mutants which were gaind by UV light irradition, levansucrase activity of mutant M-4 was 3 fold higher and biopolymer production of mutant M-1 was 2 fold higher than wild type strain..
A new Middle-Late Maturity rice cultivar with Excellent Grain Quality "Hopyeongbyeo"

Hopyeongbyeo is an anther culture derived Japonica rice (Oriza sativa L.) cultivar from the cross between Hitomebore and Hwajinbyeo by the rice breeding team of Honam Agricultural Research Institute (HARI), NICS, RDA, in 2003. This cultivar has a about 126 days growth duration from transplanting to harvesting. The short and round grain shape of Hopyeongbyeo is translucent with non-glutinous endosperm. Amylose content of rice grain is about 17.8% and high palatability of cooked rice compared with Nampyeongbyeo. This cultivar shows resistant to bacterial blight pathogenes of K1. The milled rice yield of Hopyeongbyeo is about 5.13MT/ha at ordinary transplanting culture in the local adaptability test. Hopyeongbyeo would be adaptable for the southern plain area of Korea.
Assessment of Water Quality Calibration Criteria for Field-Scale Water Quality Model

The objective of this study was to assess the statistics and associated criteria for field-scale model used to simulate nutrient concentrations in paddy field. CREAMS-PADDY, a modified version of the field-scale CREAMS model, simulates the hydrologic, sediment, and nutrient cycles in paddy fields was applied in this study. The model was calibrated and validated using data from study rice paddy fields in Republic of Korea. The calibration statistics include mean and the minimum-maximum range associated with a “temporal window” that spans a period of several days. Because nutrient concentrations in paddy filed are typically sampled infrequently (on a weekly basis, at best) and represent only an instant in time, it is not reasonable to expect any model to simulate a daily average concentration equal to an observed value on a particular day. The 5-day window and related calibration statistics were applied in this study and the applicability of this concept was tested for field-scale water quality model. As a result of calibration and validation, the ponded water nutrient concentration values showed only small changes except the fertilization period. Because of the small changes in ponded water concentration, the size of 5-day window was too small to capture the observed values. Further study is required to establish the 5-day window calibration method for field-scale water quality modeling.
농업소유역의 홍수유출량 추정을 위한 단위도 모형 비교연구

In this study, three different unit hydrograph methods (Snyder, SCS, Clark) in the HEC-HMS were compared to find better fit with the observed data in the small agricultural watershed. Baran watershed, having 3.85㎢ in size, was selected as a study watershed. The watershed input data for HEC-HMS were retrieved using HEC-GeoHMS which was developed to assist making GIS input data for HEC-HMS. Rainfall and water flow data were monitored since 1996 for the study watershed. Fifty five storms from 1996 to 2003 were selected for model calibration and verification. Three unit hydrograph methods were compared with the observed data in terms of simulated peak runoff, peak time and total direct runoff for the selected storms. The results showed that the coefficient of determination (R<sup>2</sup>) for the observed peak runoff was 0.8666~0.8736 and root mean square error, RMSE, was 5.25~6.37㎥/s for calibration stages. In the model verification, R<sup>2</sup> for the observed peak runoff was 0.8588~0.8638 and RMSE was 9.57~11.80㎥/s, which were slightly less accurate than the calibrated data. The simulated flood hydrographs were well agreed with the observed data. SCS unit hydrograph method showed best fit, but there was no significant difference among the three unit hydrograph methods.

In this study, rainfall characteristics was analyzed for a feasibility study of wastewater reuse for agriculture in Jeju island. Selected gauging stations which have long term data are Jeju(north), Seogwipo(south), Gosan(west) and Seongsan(east). While the annual rainfall of Seongsan and Seogwipo was 1,856 mm and 1,821 mm, that of Gosan and Jeju was 1,121 mm and 1,430 mm respectivly. The probability drought analysis was performed with thirty-one years rainfall data for four gauging station. The drought day of 10 year return period was 9~55 days, especially 18-42 days from September to November in Gosan.

This paper presents how effective the detention storage is to control floods at a test watershed. HEC-HMS model was applied to simulate the effects of the storages of different levels and installation methods on the flood peak reduction. The results showed that the detention storage may significant reduce the flood peaks, and the effectiveness depends on the sizes of the storage and types of installation. The simulated peak values reduce considerably for the design storm events. The results also showed that alternatives to control flood may be evaluated using the model.

A hydrologic gauging network include a real-time measuring equipment was established within the Balhan watershed. Rainfall, stream water level, flow velocities were monitored at the six gauging stations. For stream flow gauging stations, the stage-discharge relationships were developed. The flow rate of the Balan watershed was 83.60% in 2001 and 48.79% in 2002.
Treatment outcome of postoperative radiotherapy for retroperitoneal sarcoma

Purpose: To evaluate the treatment outcome and prognostic factor after postoperative radiotherapy in retroperitoneal sarcoma. Materials and Methods: Forty patients were treated with surgical resection and postoperative radiotherapy for retroperitoneal sarcoma from August 1990 to August 2008. Treatment volume was judged by the location of initial tumor and surgical field, and 45-50 Gy of radiation was basically delivered and additional dose was considered to the high-risk area. Results: The median follow-up period was 41.4 months (range, 3.9 to 140.6 months). The 5-year overall survival (OS) was 51.8% and disease free survival was 31.5%. The 5-year locoregional recurrence free survival was 61.9% and distant metastasis free survival was 50.6%. In univariate analysis, histologic type (p = 0.006) was the strongest prognostic factor for the OS and histologic grade (p = 0.044) or resection margin (p = 0.032) had also effect on the OS. Histologic type (p = 0.004) was unique significant prognostic factor for the actuarial local control. Conclusion: Retroperitoneal sarcoma still remains as a poor prognostic disease despite the combined modality treatment including surgery and postoperative radiotherapy. Selective dose-escalation of radiotherapy or combination of effective chemotherapeutic agent must be considered to improve the treatment result especially for the histopathologic type showing poor prognosis.

Purpose: To investigate the association between binocular function and vision after cataract removal and primary posterior chamber intraocular lens (PC-IOL) implantation in children with unilateral cataract and to identify visual function differences according cataract type. Methods: Clinical records of 2- to 6-year-old patients with unilateral cataract removal and primary PC-IOL implantation were reviewed retrospectively. Visual acuity and ocular alignment were measured. Sensory fusion was assessed with the Worth 4-dot test, and stereoacuity with the Titmus stereo test. Cataracts were classified according to cause, lens opacity location, age at onset, and presence of strabismus. Clinical characteristics of patients who obtained good visual function were identified. Results: Forty-seven patients were included. Among 22 (46.8%) with good vision (20/40 or better), only 6 (27.3%) achieved good binocular function (the presence of fusion and 100 seconds of arc or better of stereoacuity). Visual acuity was better in eyes with good binocular function (p=0.002). No other variables were significant for achieving good binocular function. Conclusions: The removal of unilateral cataract in a visually immature child can result in a combination of good visual acuity and binocular function. Good binocular function is closely related to good visual acuity.
