An Optimal Layout Design Scheme of Yacht Using Digital Human Modeling

Objective: In this study, working posture and traffic line of crews were analyzed using Task analysis toolkit of Digital Human Modeling program. These results would suggest tips of ergonomic design. Background: Leisure activities alter overland tourism into marine tourism according to a rise in national income. It is expected that the leisure boat with period of introduction in marine tourism will be developed rapidly. Method: (1) Leisure boat design by Rhino program (2) Conversion of design file from Rhino program into Jack program (3) To build the Human model and to apply the anthropometric data (4) Modeling the posture of crews (5) Using the Task analysis toolkit (ex. OWAS, RULA). Results: We found the space problems and load of postures. Results: We found the space problems and load of postures. Conclusion: (1) Unnatural posture of crews were sustained. (2) Workload that occurs in the human body were overestimated. (3) Crew’s work space was very narrow. Application: This study will be applied the new design of yacht.
Murine Nkx-1.2 와 상호작용하는 유전자의 클로닝

호메오 단백질은 DNA에 결합하여 전사를 조절함으로써 생명체의 형태형성에 있어 마스터의 역할을 한다. 본 연구에서 생쥐 호메오 단백질의 전사조절기작을 연구하기 위하여, 초파리의 형태형성에 중요한 역할을 하는 NK-1 유전자에 대한 생쥐의 상동유전자인 Nkx-1.2 유전자를 분리하여 그 염기서열을 밝혔다. Nkx-1.2 유전자는 Sax-1으로도 알려져 있으며 중추신경계의 형성에 중요한 역할을 하는 것으로 밝혀졌다. 이러한 Nkx-1.2와 상호작용하는 단백질을 암호화하는 유전자를 클로닝하기 위하여 Yeast two-hybrid 실험법을 이용하여 생쥐의 cDNA library로부터 5개의 서로 다른 유전자를 클로닝하여 #4, #7, #30, #41, #45 라 명명하였다. 이들 유전자의 염기서열을 분석한 결과 기존에 밝혀지지 않은 새로운 유전자임을 확인하였다. Nkx-1.2 의 카르복시말단을 부분적으로 제거한 재조합체와 클로닝된 유전자의 상호작용을 β-galactosidase 활성도로 조사한 결과, Nkx-1.2 전체 유전자를 bait로 사용한 실험군이 가장 높은 활성을 나타내었다. 또한 #45 클론을 탐침으로 사용하여 Northern blot 을 수행한 결과, #45 유전자는 대부분의 조직에서 발현하였으며, 그 크기가 약 4.5kb임을 확인하였다. 이러한 결과는 Nkx-1.2 가 여러 단백질과 상호작용하는 전사조절자로서의 역할을 하는 것으로 사료된다. Homeodomain transcription factors behave as master regulators of morphogenesis by controlling the expression of target genes. A fundamental problem is how homeodomain proteins achieve functional sepcificity in vivo in light of the facts that homeodomains bind in vitro to DNA sequences that ar remarkably similar, and that homeodomain transcription factors can act either as transcriptional activators or repressors. In order to study this problem we initiated experiments using the yeast two-hybrid system to identify genes encoding proteins that interact with the NK-homeodomain proteins and to evaluate their contribution to the functional specificity of the NK-homeodomain proteins. From the screening of mouse embryonic match-marker cDNA libraries using Nkx-1.2 as bait we discovered novel gene clones, numbered as 4, 7, 30, 41, and 45. Nucleotide sequence analysis revealed that these clones originated from different unknown genes. In order to see which region of the Nkx-1.2 protein could interact with these gene product, various GAL4 : Nkx-1.2 bait constructs containing different parts of the coding region of the Nkx-1.2 homeobox gene were generated. Domain analysis showed that full Nkx-1.2 sequence had the strongest β-gal activity. Multiple-tissue northern blot analysis revealed that #45 gene clone expressed ubiquitously and its mRNA size was 4.5Kb. These results suggest that the Nkx-1.2 functions as a protein-binding interface and thereby regulate the DNA-binding specificity of homeodomain transcription factors.

본 논문에서는 사장교를 지탱하는 주요 부재인 케이블의 손상 위치를 빠르게 검출할 수 있는 손상평가 기술을 개발하고, 모형 교량 손상 실험을 통하여 개발한 기술의 손상평가 성능을 검증하고자 하였다. 손상평가 기술의 개발을 위하여 통계적 패턴 인식 기술인 마할라노비스 거리 이론을 활용하였으며, 복잡한 구조체의 손상위치 판별을 위하여 계측 위치별 획득 데이터의 변동성을 손상평가 기술에 반영하였다. 개발한 기술의 손상평가 성능을 확인하기 위하여 모형 사장교를 대상으로 손상 실험을 진행하였다. 그 결과, 개발한 손상평가 기술은 무손상 상태의 응답과 손상 상태의 응답을 활용하여 사장교 케이블의 손상 위치를 검출할 수 있는 통계적 패턴을 제공하는 성능을 보이는 것을 확인하였다.
사장교의 케이블 손상 검출을 위한 변동성이 고려된 손상평가 기술 개발

본 논문에서는 사장교와 같은 장대형 구조물의 손상위치를 판단할 수 있는 손상평가 기법을 개발하고, 개발한 기법의 성능을 실험을 통하여 검증하고자 하였다. 손상평가 기법은 무손상 데이터가 확보되지 않은 상태에서 구조물의 손상평가가 가능하고, 구조물의 응답 데이터의 분석만으로 손상위치를 판단할 수 있는 데이터를 추출하는 것을 목표로 하였다. 이러한 목표를 완성하기 위하여, 손상 위치 판별을 위하여 통계적 패턴인식 기술인 개선된 마할라노비스 거리(IMD : Improved Mahalanobis Distance) 이론에 기반하여 변동성이 고려된 손상평가 기법을 개발하였다. 개발한 손상평가 기법에는 구조물의 고유한 정보에 기반한 Simulation 프로그램을 반영하여 다양한 외력에 따른 구조물의 무손상 응답을 출력하도록 하였다. 개발한 기법의 성능을 실험적으로 평가하기 위하여 모형 사장교를 대상으로 케이블 손상실험을 수행하였다. 그 결과, 변동성이 고려된 손상평가 기법은 외력에 따른 무손상 데이터를 자동으로 출력하고, 출력된 무손상 데이터와 계측된 손상 데이터의 분석을 통하여 케이블의 손상 위치를 판단할 수 있는 정보를 추출하는 성능을 보이는 것을 확인하였다. In this paper, we developed a damage evaluation technique that can determine the damage location of a long-sized structure such as a cable-stayed bridge, and verified the performance of the developed technique through experiments. The damage assessment method aims to extract data that can evaluate the damage of the structure without the undamage data and can determine the damage location only by analyzing the response data of the structure. To complete this goal, we developed a damage assessment technique that considers variability based on the IMD theory, which is a statistical pattern recognition technique, to identify the damage location. Prediction of Failure Mode in Hole Clinching of Al Alloy and Advanced High-Strength Steel

<P>Common joining methods used in automotive industry are welding, adhesive bonding, friction stir welding, mechanical fastening, self-piercing rivets, mechanical clinching and so on, for multi-material designed automotive bodies. Among different joining methods, mechanical clinching which achieves geometrical interlocking by plastic deformation has several advantages such as no need of additional joining elements and fast joining. But mechanical clinching is difficult to join a ductile material with a high-strength or low ductility material. Therefore the hole clinching as a new mechanical clinching process has been proposed to join these material combinations. In the hole clinching process, as punch force is applied to a upper sheet (a ductile material), it is indented into hole of lower sheet (low ductile material) on die and then interlocked by plastic deformation. It is very important for a successful design of hole clinching to predict the failure mode such as neck fracture and button separation and the strength of hole-clinched joint. For this an analytical approach was carried out for the hole clinching process of Al6061 and DP980. Tool geometry used in hole clinching was designed by the predicted failure mode. Preliminary finite element simulation was performed to validate the geometrical interlocking and joinability. The predicted failure mode and strength were verified by the results of cross tension test.</P>
