제주연안 갯녹음(백화) 지역의 해수에 분포하는 세균군의 분자생물학적 분석

본 연구에서는 갯녹음(백화)현상이 세균생태계와 어떤 관련이 있는지에 대한 기초자료 및 정보를 얻기 위하여 갯녹음 현상이 일어난 제주도 성산과 강정지역 연안의 해수에 존재하는 세균군을 16S rRNA 증폭기법을 이용하여 조사하였다. 강정지역에서는 Alcanivorax, Paracoccus, Damselae, Pseudomonas, Rhodovulum, Silicibacter, Sulfitobacter, Roseobacter 등 다양한 종류의 세균이 분포되어 있었으며, Alcanivorax가 20%의 빈도로 가장 많이 나타났다. 반면, 성산 지역에서는 Pseudomonas속 균주가 우점종으로서 존재하였으며, Pseudomonas tolaasii(혹은 Pseudomonas corrugata)와 유연관계가 가까운 세균은 44%, Pseudomonas mandeli와 가까운 세균이 24%, Verrucomicrobiales와 가까운 세균은 4%, 기타 동정되지 않은 세균은 세 group으로 구분되었으며, 각각 8%, 8%, 12%를 차지하여, 두 곳에 분포되어 있는 세균군상이 상당한 차이점이 있음을 확인하였다. 갯녹음 지역인 강정과 성산 해수의 8월(표품 채집시기)의 수온은 27℃-27.5℃, 염분의 농도는 30.24-30.60%, pH는 8.23-8.36, 용존산소량(DO)은 각각 7.20-7.28로, 두 지역에서 매우 비슷한 것으로 조사된 바 있는데, 이는 수온이나 염분의 농도, 또는 pH보다는 다른 원인에 의하여 두 갯녹음 지역에 분포하는 세균군의 차이를 가져왔음을 제시해 주었다. In this study, the bacterial communities distributed in sea water of the whitening areas of Gangjeong and Seongsan, Jeju-do have been analyzed using the PCR amplification of 16S rRNA to obtain fundamental data and information on relationship of the whitening phenomenon and microbial ecosystem. In Gangjeong, diverse bacteria such as Alcanivorax, Paracoccus, Damselae, Pseudomonas, Rhodovulum, Silicibacier, Sulfitobacter, and Roseobacier have been found, and Alcanivorax was the most abundant clone. The most abundant clone from Seongsan was Pseudoinonas, of which Pseudomonastolaasii and Pseudonionas mandeli were most abundantly occurred in the frequency of approx 44% and 24%, respectively. Approx 4% of the bacterial clones closest to firruconiicrobiales and other unidentified clones were also found in Seongsan, suggesting there is a great discrepancy between bacterial communities from the whitening areas of Seongsan and Gangjeong. The mean tem- perature, chlorine concentration, pH, and dissolved oxygen (DO) of the sea water of Gangjeong and Seongsan in August of 2001 (sampling period) was 27℃-27.5℃, 30.24-30.60%, pH 8.23-8.36,7.20-7.28 ㎎/ℓ, suggesting other environmental factors except for the factors mentioned above might result in difference of bacterial communities distributed in both areas.

Hsp90 is one of the most conserved molecular chaperones ubiquitously expressed in normal cells and over-expressed in cancer cells. A pool of Hsp90 was found in cancer mitochondria and the expression of the mitochondrial Hsp90 homolog, TRAP1, was also elevated in many cancers. The mitochondrial pool of chaperones plays important roles in regulating mitochondrial integrity, protecting against oxidative stress, and inhibiting cell death. Pharmacological inactivation of the chaperones induced mitochondrial dysfunction and concomitant cell death selectively in cancer cells, suggesting they can be target proteins for the development of cancer therapeutics. Several drug candidates targeting TRAP1 and Hsp90 in the mitochondria have been developed and have shown strong cytotoxic activity in many cancers, but not in normal cells in vitro and in vivo. In this review, recent developments in the study of mitochondrial chaperones and the mitochondria-targeted chaperone inhibitors are discussed [BMB reports 2012; 45(1): 1-6]

The Rehabilitation Facilities for disabled people keeps going to grow with social changes that disabled people are dramatically increasing. Despite its growth health center newly established is organized by means of following the program and making up space being operated at current center rather than considering the relation between program and space being operated. However, future health center has to be made up not only for disabled people but also for maximum thinking of harmonization with a local resident. Now, this study suggests that we should have a thorough grasp and analyzation about program and making up space at current Rehabilitation Facilities and it is available to be rational for the future health center.

Purpose: The number of the transportation poor has been steadily increased, and also the social perspective toward the disabled has been changed. Thus, Universal Design has been needed. In particular, the unknown majority of the transportation poor have frequently used the urban railway facilities which should reflect the ideals and principles of Universal Design. However, previous research has not focused on the study on critical factors of Universal Design. Based on this observation, the present study aims to find out Universal Design critical factors of urban railway station. Methods: This study explores the criteria of planning Universal Design in the urban railway station by analyzing and comparing the similar criteria used in both domestic areas and foreign countries. Result: Findings from this study are as follows. First, we found out the critical factors of Universal Design in the urban railway station by comparing and analyzing the similar criteria previously used. Second, this study also suggests the better directions toward the domestic criteria relating to the urban railway system. Third, we found the need of the universal standard as well as individually corresponding strategy. Moreover, we found the need of preparing the critical factors for the physical environments as well as the human service. Implications: The further research on findings of the planning criteria will have been continued on the basis of findings of the present study.

The photosynthetic bacterium, Rhodospirillum rubrum S1, when grown under anaerobic conditions, generated three different types of catalases. In this study, we purified and characterized the highest molecular weight catalase from the three catalases. The total specific catalase activity of the crude cell extracts was 88 U/mg. After the completion of the final purification step, the specific activity of the purified catalase was 1,256 U/mg. The purified catalase evidenced an estimated molecular mass of 318 kDa, consisting of four identical subunits, each of 79 kDa. The purified enzyme exhibited an apparent Km value of 30.4 mM and a Vmax of 2,564 U against hydrogen peroxide. The enzyme also exhibited a broad optimal pH $(5.0{\sim}9.0)$, and remained stable over a broad temperature range $(20^{\circ}C{\sim}60^{\circ}C)$. It maintained 90% activity against organic solvents (ethanol/chloroform) known hydroperoxidase inhibitors, and exhibited no detectable peroxidase activity. The catalase activity of the purified enzyme was reduced to 19 % of full activity as the result of the administration of 10 mM 3-amino-1,2,4-triazole, a heme-containing catalase inhibitor. Sodium cyanide, sodium azide, and hydroxylamine, all of which are known heme protein inhibitors, inhibited catalase activity by 50 % at concentrations of $11.5{\mu}M,\;0.52{\mu}M,\;and\;0.11{\mu}M$, respectively. In accordance with these findings, the enzyme was identified as a type of monofunctional catalase.
Proteomics for monitoring of chicken responses to immune system in Korean Native Chickens

본 연구는 proteomics의 방법을 이용하여 가금의 면역시스템에 관련된 단백질을 찾고자 수행하였다. 면역활성 물질을 처리한 후 면역활성화시 발현되는 단백질을 찾아보았다. 이 결과는 가금의 면역과 관련된 단백질 마커의 이용뿐 아니라 현재까지 밝혀지지 않은 가금의 2D map을 만드는데 역시 중요하게 이용이 될 것이다. Specific biomarkers to detect significant immunological or physiological responses would be extremely valuable on the development of feeding technique. Proteomics, the study of proteins within a cell or biological samples, may offer a novel approach to immunological or physiological monitoring of chicken responses to immune system. By studying the protein content of cells responding to a vaccine or growth factor, it may be possible to develop a metric for quantitating the magnitude of immunological or physiological responses. Proteomics could also provide a tool for obtaining valuable information regarding the underlying regulatory mechanisms and pathways in Korean Native Chicken by comparing with subspecies of KNC and other species, like Cornish and White Leghorns.
금식유발 다뇨 및 요농축력 감소 병인으로 신장 수분통로 및 나트륨 운반체 조절 변화

Purpose: Starvation causes impairment in the urinary concentration ability. However, the molecular basis for the impaired urinary concentration and polyuria remains undefined. We examined the effects of food deprivation on the water handling by the kidney and it`s regulatory mechanism. Methods: Sprague-Dawley rats were used. They were placed in metabolic cages and deprived of food but had free access to water for 24 hours. Control rats had free access to both water and food. Protein expression of aquaporin-2 (AQP2) and Na(+)-K(+)-2Cl(-) cotransporter (NKCC2) was determined in the kidney by Western blot analysis. Protein expression of type VI adenylyl cyclase and prostaglandin E2 synthase (PGES) was determined. Urinary PGE2 excretion was also determined by radioimmunoassay. Results: Food deprivation (FD) resulted in impaired urinary concentration associated with decreased tubular water reabsorption and increased urine output. The expression of AQP2 proteins was significantly decreased in the inner stripe of the outer medulla (ISOM). The expression of NKCC2 was not affected in ISOM. The adenylyl cyclase VI expression was increased in ISOM in FD rats. The protein expression of PGES was decreased in the cortex/OSOM and ISOM. The 24 hr urinary excretion of PGE2 was significantly decreased in FD rats compared with controls. Conclusion: These findings indicate that FD-induced urinary concentration defect may related to a reduced abundance of AQP2 in the kidney. It is also suggested that the primary impairment in the pathway to the activation of AQP2 in food deprivation is independent of vasopressin/cAMP or prostaglandin activity.

Weight reduction for an automobile component has been sought to achieve fuel efficiency and energy conservation. In response to this trend, a new disc-brake called the circumferential friction disc-brake is suggested. This paper compares the mechanical performances of a conventional disc-brake and a suggested disc-brake, under the dynamic braking condition. The thermoelastic instability is considered to simulate the test condition. Furthermore, the metamodel using kriging interpolation method is introduced to obtain the optimum design of a suggested circumferential friction disc-brake. The design results obtained by the kriging method are compared with those obtained by the ANSYS.

The purpose of this study is to suggest suitable improvement on a physical environment and a business environment of the work facility for the worker and the user. And we can get the following important points as conclusion; ① We suggest suitable improvement on the architectural module and a core form of the work facility which considers on the disabled and the elderly. ② Convenient facilities of the work facility have to be focused a availability better than installation rate. ③ A business environment of the work facility has to be considered characteristics and a human size of the disabled and the elderly.
