Precipitation and purification of amylase secreted by Streptomyces aureofaciens 77 in liquid inorganic salts-starch medium under the optimum conditions were carried out. Ammonium sulphate fractionation was used to precipitate amylase in cell free culture filtrate. (NH/sub 4/)/sub 2/ SO/sub 4/ at a concentration of 50-70% saturation gave the highest enzyme yield. The obtained precipitates were redissolved in phosphate buffer (pH 7.0) and subjected to dialysis. The dialyzed enzyme preparation was applied to DEAE-cellulose column chromatography which resulted in an increase of purification up to 59.48 fold. A further step of purification was done by applying the obtained purified sample to Sephadex-G200 column chromatography which resulted in ann increase of purification up to 73. 92 fold. The results clearly indicated that the isolated amylase from S. aureofaciens 77 was only on type.

The effects of pH values, temperature and some elements on the amylolytic activity and stability of the purified S. aureofacienc 77 amylase were studied in this investigation. The purified enzyme showed its maximum activity at pH 6 within 8 min incubation at $40^{\circ}C$. None of the tested 6 metals showed on stimulatory effect on the enzymatic activity, $Fe^{+++}$, $Cu^{++}$ and $Hg^{++}$ at high dose inhibited the enzyme activity to great extent as compared with $Zn^{++}$, $Mn^{++}$ and $Fe^{++}$ whih gave less effect in this respect. The enzyme liquor was found to be thermolabile, since it lost completely its activity after 4 days incubation under room temperature and showed maximum activity during this period as a result of additions of $Ca^{++}$and NaCl, Gradual reduction was however recorded until activity reached 30% after 60 days of incubation.
Comparative Study of Itraconazole-Cyclodextrin Inclusion Complex and Its Commercial Product

Itraconazole (ITZ) solid complex using hydroxypropyl-β-cyclodextrin (ITZ-HP-β-CD) with 20% polyvinylpyrrolidone was prepared by a co-evaporation method. The complex improved antifungal activity against C. parapasilosis and C. albicans. The complex demonstrated good flow and compressibility characteristics. The complex was formulated as a capsule dosage form and drug release was evaluated. Capsules containing ITZ-HP-β-CD at a molar ratio of 1:3 with 20% polyvinylpyrrolidone have a faster dissolution rate than commercial capsules (Sporanox ®). About 88% of ITZ was released in less than 30 min and the initial dissolution rate exhibited a 3.5-fold increase compared to the commercial product. UV spectrophotometeric, HPLC, and antimicrobial methods were used to determine ITZ concentration in the release medium and the results obtained by these methods are reported. It was found that HPLC analysis is a suitable and reliable method for determination of the drug concentration with a coefficient of variation less than 10%. The intraday precision showed a coefficient of variation less than 3.96%, and that for interday was less than 4.99%. The HPLC method was more accurate and precise than the antimicrobial and UV-spectrophotometric methods for determination of ITZ concentration present in the release medium.

Two novel nanocomposites based on polyaniline functionalized ZnO and ZnO-SiO2 nanoparticles(ZnO@PANi and ZnSiO@PANi) were synthesized, characterized, and evaluated as protective films for steelpipelines in the pickling process at 25–55 C. The prepared nanocomposites were described using differentspectroscopic characterization methods including UV–vis, FTIR, DLS, XPS methods, and other physicochemicaltechniques including XRD, FESEM, and HR-TEM. The novelty of these films is in the fact thatthe ZnO and ZnO-SiO2 nanoparticles are functionalized by polyaniline which is electrochemically stablein acidic solution and has high conductivity. Electrochemical systems such as open circuit potentials(OCP), electrochemical impedance spectroscopy (EIS), and potentiodynamic polarization (PDP) were utilizedto attain the kinetics and mechanistic findings of the corrosion protection route. The outcomes indicatethat the two fabricated ZnO@PANi and ZnSiO@PANi are efficient acidic corrosion inhibitors. Theprotection performance of ZnSiO@PANi (98.6%) was more pronounced at a dose (150 mg/L) thanZnO@PANi (92.3%) and individual PANi (83.4%) as achieved from PDP findings. The effect of temperatureand flowing conditions reinforces further the performance of both nanocomposites. Surface characterizationusing FESEM/EDS delivered more indication for the steel surface protection with the ZnO@PANi andZnSiO@PANi nanocomposites. Molecular modeling using DFT calculations and MC simulations supportedthe experimental findings. The binding energies of the compounds and Fe interface follow the order ofZnO@PANi > ZnSiO@PANi, which is in good agreement with the empirical results. An important suggestionof these results is the probability of substituting traditional toxic small molecules with cost-effectiveand non-toxic polymeric nanocomposites as protective films for use in the oil and gas industry.
Nonlinear implicit fractional differential equation involving -Caputo fractional derivative

In this paper, we consider a nonlinear implicit fractional dierential equation with nonlocal condition and involving the Caputo fractional derivative with respect to another function. We investigate the existence, uniqueness of solution on subinterval of the original interval. Hence we give an estimation for this solution. Further, we discuss the continuous dependence of solution involved in the problem. The results obtained by means of a variety of tools fractional calculus including Banach contraction mapping principle. Illustrative examples are also given.

Background: For children with cleft palates, surgeries at a young age are necessary to reduce feeding or phonation difficulties and reduce complications, especially respiratory tract infections and frequent sinusitis. We hypothesized that dexmedetomidine might prolong the postoperative analgesic duration when added to bupivacaine during nerve blocks. Methods: Eighty patients of 1-5 years old were arbitrarily assigned to two equal groups (forty patients each) to receive bilateral suprazygomatic maxillary nerve blocks. Group A received bilateral 0.2 mL/kg bupivacaine (0.125%; maximum volume 4 mL/side). Group B received bilateral 0.2 mL/kg bupivacaine (0.125%) + 0.5 µg/kg dexmedetomidine (maximum volume 4 mL/side). Results: The modified children’s hospital of Eastern Ontario pain scale score was significantly lower in group B children after 8 hours of follow-up postoperatively (P < 0.001). Mean values of heart rate and blood pressure were significantly different between the groups, with lower mean values in group B (P < 0.001). Median time to the first analgesic demand in group A children was 10 hours (range 8-12 hr), and no patients needed analgesia in group B. The sedation score assessment was higher in children given dexmedetomidine (P = 0.03) during the first postoperative 30 minutes. Better parent satisfaction scores (5-point Likert scale) were recorded in group B and without serious adverse effects. Conclusions: Addition of dexmedetomidine 0.5 μg/kg to bupivacaine 0.125% has accentuated the analgesic efficacy of bilateral suprazygomatic maxillary nerve block in children undergoing primary cleft palate repair with less postoperative supplemental analgesia or untoward effects.
