세포외 기질 물질에 따른 심근세포(HL-1)의 성장 연구

HL-1 세포(심근세포주)는 AT-1 쥐의 심방암세포에서 추출한 심근세포로써, 특별한 약물의 주입 없이도 맥동현상을 보이며, 여러 번의 계대 배양 후에도 그 수축성을 잃지 않는 특성이 있어 바이오센서에 적용하기 용이하다. 본 연구에서는 세포 외 기질물질(ECM: extra cellular matrix)의 영향에 따른 HL-1 의 성장과 형질변화를 분석하였다. HL-1 세포를 ECM 이 처리되지 않은 표면과 서로 다른 3 가지 ECM 인 gelatin 수용액, fibronectin 수용액 및 gelatin 과 fibronectin 의 혼합 수용액으로 처리된 표면에 배양하여 세포와 ECM의 상호작용 및 세포의 성장을 관찰하였다. 세포 배양 직후부터 배양후 4 일까지의 세포의 변화를 형광 면역 염색법 (immunostaining)을 이용하여 분석하였다. 세포 증식은 Hoechst 와 EthD-1 을 통해 관찰하였고, DAPI 염색으로 세포핵을 phalloidin 염색을 통해 F-actin 을 관찰하였다. fibronectin 수용액을 ECM으로 사용하였을 때, 세포 성장 및 형질유지에가장 효과적인 것으로 나타났다. 본 연구의 결과는 HL-1 세포와 ECM 의 상호 작용 및 세포의 성장을 이해하는 데 기초자료로 활용될 수 있으리라 기대된다. We present here the effect of extracellular matrix (ECM) on the proliferation and physiology of HL-1 cardiac cells. HL-1 cell is from AT-1 mouse atrial cardiomyocyte tumor lineage. HL-1 cell can be serially passaged, yet they maintain the ability to contract which is a promising character of HL-1 cell for the cell based biosensors. HL-1 cells grow up on the ECM which can affect on the attachment and growth of HL-1. In this paper, we discuss HL-1 cell-ECM interactions with three different ECMs and non-treated surface. HL-1 cells are grown for 4 days after seeding then observed their attachment. Also they were immunostained by hoechst and EthD-1 for proliferation, phalloidin for Factin, and DAPI for nuclei. Fibronectin was revealed as the proper ECM material for HL-1 cell culture. This study can provide basic information for understanding the cell-ECM interactions and growth of HL-1 cells.
실내 어린이 놀이공간의 색채 사용현황 및 사용자 선호도에 관한 연구

Commercial facilities have set up play space inside the facilities for consumers accompanied by their children as part of service. However, if examining the color design of the play space installed inside the facilities, it has been found that colors are composed without any plan in advance depending on children’s superficial propensity to prefer primary colors to result in confusion. Accordingly, this research analyzed the current status of color use collecting data of photographs visiting play spaces in person which spoken their willingness to cooperate out of 7 play spaces installed in department stores and large-scale retailers in Seoul region, and investigating the distribution chart of colors and the color tone. In addition, so as to investigate color image preferences of 5~10 year old children who are users of play space, they were surveyed depending on children’s sex and age with 8 representative images obtained by the process to extract sensitive language. As it is the survey results of the current status and preferences by the number of limited samples, it has limitations to do a broad interpretation targeting the whole, but by making a diagnosis of the current conditions, it will be of help to improve and reform color environment of indoor play space which will be designed in the future, which can be said to be the meaning and significance of this study.
세포외 기질 물질의 물리·화학적 영향에 따른 심근세포(HL-1)의 성장 연구

본 연구에서는 세포의 성장속도, 배양표면 부착, 체내 특이성을 유지에 중요한 요소인 세포외 기질 물질에 따른 심근세포주(HL-1 cell)의 성장, 생존률 및 표현형 등을 분석했다. 서로 다른 5 가지의 세포외 기질 환경을 조성하여 세포 성장에 미치는 영향에 대하여 분석했다. 세포외 기질 물질이 처리된 배양 표면의 물리적인 형태는 원자현미경을 통하여 분석했으며, 세포의 표면부착, 성장 및 증식과 생존률 및 표현형은 역상형광현미경 및 면역염색법을 통해 분석했다. 본 연구를 통하여 서로 다른 세포외 기질 물질이 세포의 부착 및 성장 속도에 영향을 미치며, 심근세포의 특이성을 나타내는 단백질의 형성에도 차이를 보이는 것을 확인했다. The extracellular matrix (ECM) is a key factor affecting cell growth and adhesion to the culture surface, and it is also important for maintaining the innate characteristics of cells. Here, we describe the effects of the ECM on cardiomyocyte (HL-1 cell line) growth, viability, phenotype, and contractile ability. Five different ECM materials were investigated to analyze their effects on the cell growth. The physical morphology of the ECM-coated surfaces was scanned with an atomic force microscope (AFM), and the attachment, growth, proliferation, viability, and phenotype of the cells were analyzed using fluorescence immunostaining and an inverted phase contrast microscope.

Cell growth in in-vivo or in-vitro is affected by multiple stimuli: cell-cell interactions, physical and chemical cues from cell-surface interaction, and supplied nutrients. Maintaining the natural characteristics of cells by mimicking in-vivo environments is important for the in vitro cell culture system. Here, we present the effects of extracellular matrix on myocardial cell (HL-1 cell line) growth, viability, phenotype and its contractile ability. Five different extracellular matrix materials were investigated to figure out the effects on the cell growth. The physical morphology of the ECM coated surfaces was scanned with atomic force microscope (AFM) and the attachment and growth of cells were analyzed with inverted phase contrast microscope. Cardiac cell proliferation, viability, phenotype were also analyzed with fluorescence immunostaining.
