      • 쥐의 간 시토졸 Aldehyde dehydrogenase의 정제 및 특성에 관한 연구

        쥐의 간 시토졸 Aldehyde dehydrogenase(ALDH)를 황산 암모늄 처리, DEAE-Sephacel, CM-cellulose, Sephacryl S-300, 5'-AMP Sepharose 4B chromatography 순으로 정제 하고 TSK G3000SW column을 이용하여 HPLC로 분석한 결과 순수한 단일 peak를 얻었고, Sephacryl S-300 chromatography 방법으로 측정된 분자량은 217,000 dalton이었으며 SDS-PAGE를 행한 결과 분자량이 53,000인 subunit로 구성된 homotetramer임이 밝혀졌다. 이 효소의 pI값은 6.75, 최적 pH는 9.5이며 dehydrogenase활성 뿐 아니라 esterase활성도 나타내었다. Acetaldehyde에 대한 $K_m$값은 $1.06{\times}10^{-3}M$이지만 indole 3-acetaldehyde에 대한 $K_m$, 값은 $10^{-6}M$이하, benzaldehyde에 대한 $K_m$값이 $(1.59{\times}10^{-5}M)$로 낮은 값을 나타내는 것으로 보아 시토졸 ALDH는 주로 biogenic aldehyde나 aromatic aldehyde 대사에 관여하는 것으로 생각된다. Cytosolic ALDH of rat liver was purified by ammonium sulfate precipitation, DEAE-Sephacel, CM-cellulose ion exchange chromatography, Sephacryl S-300 gel filtration, and 5'-AMP Sepharose 4B affinity chromatography. The molecular weight was determined by Sephacryl S-300 gel filtration and found to be 217,000 dalton. SDS-PAGE showed that it was homotetramer consisting of four identical subunits whose molecular weight was 53,000 dalton. Optimal pH of this enzyme was 9.5 and pI value was 6.75. $K_m$ value of rat liver cytosolic ALDH for acetaldehyde was $1.06{\times}10^{-3}M$, but those of indole-3-acetaldeghyde(<$10^{-6}M$) and benzaldehyde $(1.59{\times}10^{-5}M)$ were extremely low. These results suggested that rat liver cytosolic ALDH might be primarily involved in the oxidation of aromatic aldehyde including biogenic aldehyde.

        정제된 쥐의 간 시토졸 aldehyde dehydrogenase(ALDH)의 활성부위를 화학변형 시약을 이용하여 검색한 결과 cysteine, serine 및 glutamate 잔기가 활성에 중요한 역할을 할 것으로 생각된다. 또한 ALDH의 기절 활성화 특성과 chloral hydrate에 의한 억제양상 및 disulfiram에 의한 억제양상으로부터 ALDH의 반응 메카니즘을 추정하였다. Chemical modification experiments suggested that cysteine, serine, glutamate residues might participate in the catalytic role and it was suggested that a probable reaction mechanism of ALDH through substrate activation, chloral hydrate inhibition and disulfiram inhibition behaviors towards ALDH activity.

        쥐의 간 미토콘드리아 Aldehyde Dehydrogenase 의 분포와 특성에 관한 연구

        Separation of inner membrane, outer membrane and matrix were accomplished by treating rat liver mitochondria preparation with digitonin followed by differential centrifugation. Mitochondrial ALDH isozymes were founded to be located in outer membrane as well as matrix fraction. Soluble low K_m ALDH in matrix had a very low K_m ($lt;10^(-6)M) for acetaldehyde, but the K_m of soluble high K_m ALDH in the matrix was 9.8 × 10^(-4) M, outer membrane bound ALDH showed a relatively high K_m, 1.4 × 10^(-2)M. The outer membrane bound ALDH was purified and characterized. Optimal pH was 9.5 and pI was 6.1. Subunit molecular weight was determined by SDS-PAGE and found to be 55,000. These results suggest that the soluble low K_m ALDH might be primarily response for the oxidation of acetaldehyde in rat liver, but the outer membrane bound ALDH will serve as barrier when the concentration of acetaldehyde in cytosol is high.

        바이오필릭 디자인 기반 교실 디자인 설계

        This study aims to suggest a Biophilic classroom design of high school to obtain attention restoration for students. The learning space for youth in the modern society is composed of dry artificial structures. This space is considered to be a space that can not relieve stress caused by learning. "The Attention Restoration Theory" is divided into "Directed Attention" of humans, which is the cause of fatigue and stress, and "Involuntary Attention" as a solution to it. "Involuntary Attention" takes place in a rest state and helps the brain recover when exposed to nature. And the core of "Biophilic-Design Theory" is that humans can recover physical and mental conditions when exposed to nature. The purpose of this study is to apply "The Biophilic-Design Theory" that emphasizes the importance of exposure to nature to the educational space and plan the space where the 'Attention Restoration' can be achieved. The research method is as follows. First, we review previous studies related to "The Biophilic-Design Theory" and "The Attention Restoration Theory". Second, we analyze the application examples of "The Biophilic-Design Theory" and "The Attention Restoration Theory" in domestic and foreign educational spaces. Third, the concept of educational space is set up based on the elements derived from previous studies. Finally, we propose the planning direction of classroom design based on Biophilic-Design. The following conclusions were drawn. First, The creation of the education space to restore the learner's attention requires a visual space plan that utilizes natural elements such as natural light, artificial light, plants, and natural materials that can directly experience nature. Second, the direction in which students in the classroom can be "The Attention Restoration Theory" should consider the use of indirect natural elements that bring the surrounding natural landscape into the interior. This study will be used as the baseline data for the spatial design and planning of education facilities based on Biophilic-Design.

        건강생성 모델기반 노인복지관 계획 요소

        The purpose of this study is to propose the clue of planning guideline for the elderly welfare center supporting the healthy life of the elderly through deriving the planning element of the elderly welfare center based on the Salutogenic Model(SM). The research method was proceeded with literature review. The meaning of Sense of Coherence(SOC) and Generalised Resistance Resources(GRR) which constitute Salutogenic Model is established. The correlation between SOC and GRR is analyzed. The conclusions of this study are as follows. First, EBD, BD, HD, UD, and BFD were studied as planning theories that could enhance the SOC of space. Second, based on the five planning theories, 43 planning elements needed for the elderly welfare center plan were derived. Third, as a result of classifying the plan elements based on the SOC, ‘Manageability’ is divided into 22 elements, ‘Meaningfulness’ is 11 and ‘Comprehensibility’ is 10 elements. Fourth, the details of the SOC items for each theory are as follows: BFD focuses on ‘Manageability’ with ‘Manageability’(74%) and ‘Comprehensibility’(26%), but ‘Meaningfulness’ does not exist. And UD regards ‘Comprehensibility’(66%) as important, and ‘Manageability’ and ‘Meaningfulness’ as 17%. BD, on the other hand, has a high percentage of ‘Meaningfulness’(70%), ‘Comprehensibility’(21%) and ‘Manageability’(9%). Next, the ‘Manageability’ of HD and EBD is 46%. And HD was ‘Meaningfulness’(34%), ‘Comprehensibility’(20%), and EBD was ‘Meaningfulness’(30%) and ‘Comprehensibility’(24%). The three items of SOC showed different distribution according to the spatial planning theory. As a result of the analysis, the spatial planning theory with the ‘Comprehensibility’ was related to Universal Design(UD), and the spatial planning theory with the ‘Manageability’ was related to Barrier-Free Design(BFD). In addition, the spatial planning theory of ‘Meaningfulness’ was related to Bio-philic Design(BD). Therefore, the plan of the elderly welfare center needs to approach the multidimensional design methodology to enhance the SOC(Sense of Coherence).

        실내공간의 주시에 나타난 정보획득률과 주시시간 분석에 관한 연구

        This study is to set up the appropriate range of observation time through contemplating the characteristics of observation time run for the information acquisition of space. The conclusions reached through this study are as the followings. First, this study could find out that even though the evaluation elements on the three types for image evaluation were the same, the information acquisitions were different as those types varied. On the other hand, the change of the average run-time by type for the information acquisition was found not to be big, in other words, even though the run-time was alike, the information acquisitions varied depending on the type. Second, he evaluation by language media showed the average value by element had the order of [shape> position>number>existence] and the range of their run-time was 94.6∼102.9 seconds. The average rate of information acquisition shown at the visual media had the order of [composition>shape>material&color] and the range of run-time was 93.1∼99.7 seconds. Third, the evaluation by language media showed that for male subjects the range of information acquisition rate was 39.1∼91.4% and that of run-time 85.1∼106.0 seconds and for female ones 46.0∼94.6% and 96.3∼112.3 seconds respectively. In case of the visual media, male subjects showed the range of information acquisition rate was 40.3-66.7% and the range of run-time 82.4∼97.9 seconds and the female ones, 42.2∼71.0% and 94.0∼115.1 seconds respectively, through which we could see that at the evaluation by language media and visual media both the female"s range of information acquisition and that of observation time were higher than the male"s.

      • 고속 자율 주행 차선유지시스템을 위한 DNN 기반 레이더를 이용한 차선 추정

        In this paper, we propose a novel Deep Neural Network(DNN)-based lane estimation method with radar for lanekeeping system(LKS) without vision sensors. The vision sensor is widely utilized for detecting the road lane. However, it is well known that the vision sensor has weaknesses about environmental effects such as weather, contaminated lane, and so on. Thus, we exploit the radar sensor, which is robust against environmental effects, to estimate the road lane model for LKS. Scaled Conjugate Gradient(SCG) method is used for optimization of the neural network. We had a comparative study between the proposed system and the camera system which is conventionally used for LKS. The proposed system is expected to improve the performance of LKS by sensor fusion.
