This study was designed to investigate the appearence and the characteristics of the apoptotic cells and the process of the joint cavity formation in mouse knee joint. Fetal mouse knee joints from 15 to 19 days of gestation were used. Paraffin-embedded serial sections, stained with H & E for light microscopic observation, Epon 812 embedded thin sections for electron microscopic observation and Lowicryl HM 20 embedded thin sections for immune-electron microscopic observation were prepared. Monoclonal antibodies to $\beta-tubulin$ and polyclonal antibodies to tissue transglutaminase were used for immune-electron microscopic study. The results obtained were as follows. 1. At 15 days of gestation, blood vessels, which have invaded in the mesenchymal cells, were present in the synovium, to form the joint cavity in the future. 2. At 16 days of gestation, the joint cleft was first appeared and several RBCs were present in the joint cleft. The invasion of blood vessels into the joint cleft was continuing, and apoptotic cells were present in the inner cell layer, adjacent to the joint cleft. Necrotic cells were also present in the outer cell layer; they were present 18 days of gestation, but apoptotic cells did not appear after 17 days of gestation. 3. In the apoptotic cells, transglutaminase were localized around vacuoles and the marginal site of the cytoplasm. 4. In the apoptotic cells, tubulin was around the endoplasmic reticulum and the marginal site of the cytoplasm. In the late stage of apoptotic cells, tubulin was localized diffusely in the cytoplasm. Tubulin was also strongly labeled around in the cytoplasm of the neighboring cell at which the apoptotic body was phagocytosed. Tubulin labeled particles were apparently increased in the seperated apoptotic bodies. On the basis of the above findings, it is proposed that during the development of the mouse knee joint, blood vessel invasion first occurs and then apoptosis and cell necrosis follow it. In the apoptotic cell, present in the synovium of the developing knee joint of the mouse. it is suggested that the redistribution of tubulin is associated with apoptotic process. And transglutaminase overexpressed in the apoptotic cell.
소의 분변과 도체에서 shiga toxin-producing Escherichia coli의 분리와 특성

Shiga toxin (Stx)-producing Escherichia coli (STEC) strains can cause broad spectrum of human disease, including diarrhea, hemorrhagic colitis, and the life-threatening hemolytic uremic colitis (HUS). We examined 868 samples was taken from bovine feces and carcass from January to December 2008 in Seoul. Twenty two (9.5%) shiga toxin -producing Escherichia coli were isolated from the 230 of bovine feces, and two (0.31%) were isolated from the 638 of carcasses. Serotype of E. coli isolates were O157 (10, 41.6%), O26 (10, 41.6%), O111 (1, 4.2%) and UT (3, 12.6%). In PCR, the isolates displayed three different stx gene combination (stx1〔2, 8.4%〕], stx2〔3, 12.6%〕and stx1 and stx2〔19, 87.5%〕). The eaeA and hlyA gene were found in 11 (45.8%) of the 24 strains. Saa gene was present only one strains (4.2%). Toxin typing using reverse passive latex agglutination test showed the same result in VT 1. But it showed different result in VT 2. In antimicrobial susceptibility test, all isolates were sensitive to amikacin, amoxicillin/clavulanic acid, ciprofloxacin and colistin. Eighteen strains (75.0%) of 24 isolates showed the multi-resistant patterns with over 3 drugs. PFGE was performed after the genomic DNA of twenty four isolates was digested with Xba I. the 24 isolates showed 7 (A~G) PFGE type.

Botulism in wild waterfowl has been reported in many areas of the world. In 18 October 2008 to 22, 101 dead wild ducks were found in Anyangcheon. 24 of dead birds were spotbill (Anas poeilorhyncha), 56 were Mallard (Anas platyrhynchos), 18 were green- winged teal (Anas carolinensis) and 3 were others. Clinical sign of the sick birds were flaccid paralysis of the neck. At autopsy, most birds had no specific lesions. Stomach is empty. In the mouse bioassay, Samples of serum and intestinal contents taken from 9 dead birds killed all the injected mice, and their toxicity was neutralized by the antitoxin against C. botulinum type C toxin.

In this study, a total of 2,119 samples was taken from bovine feces and carcass from March 2002 to December 2003. And those were examined for the presence of enterohemorrhagic E coli O157:H7. The properties of the isolates were characterized for biochemical features, serotypes, virulence genes and antimicrobial susceptibility. Forty five strains(3.7%) of E coli O157:H7 were isolated from 1,208 fecal samples and were not detected in carcass using immunomagnetic separation technique and selective media. In multiplex PCR using stx1, stx2, eaeA and hlyA primers, the amplified bands at 180bp, 255bp, 384bp and 534bp were observed, respectively. In antimicrobial susceptibility test, all isolates were susceptible to amoxicillin/clavulanic acid and cefazolin. The isolates were most resistant to sulfisoxazole(24.4%), followed by streptomycin(22.2%), tetracycline(20.0%). Eight strains(17.8%) of 45 isolates showed the multi-resistant patterns with over 3 drugs.

Purpose - To understand the growing interests in the professional temporary workforce, this study aimed to examine the different types of professional temporary workers and their different effects on creative performance. Design/methodology/approach - By sub-classifying professional temporary workers as boundaryless workers and transitional workers, this study developed propositions to provide comprehensive framework for directing meaningful topics for future research on professional temporary workers. Findings - This study suggested that two types of professional temporary workers may differ significantly in terms of their work efforts for getting approval or acceptance from the team members and for achieving creative contributions in teams. Specifically, drawing on expectance theory, social identity theory and resource-based theory, this study discussed the relationship between acceptance(task-oriented vs. relational-oriented acceptance), employment duration(short-term status vs. long-term status), contract security(volition being temporary worker) and creative performance for boundaryless workers versus transitional workers. Research implications or Originality - This study have implications for increasing our understanding of professional temporary workers by showing that creative performance may vary by employment types.

Yersinia enterocolitica is a zoonotic agent, and to cause food poisoning. This study was carried out to get some basic information for the control of Yersinia infection. A total of 1,680 samples were collected from beef and pork carcasses from January 2006 to December 2007 in Seoul. The isolation rate was higher in pork carcass than in beef carcass. Five(0.59%) Yersinia enterocolitica were isolated from the 840 of beef carcasses, and eighteen(2.14%) were isolated from the 840 of pork carcasses. Among 23 strains, 22 were classified into biotype 1A, and one was biotype 6. In serotyping of Y enterocolitica isolates, 21 strains were untypable (UT), and 2 were O5 and O8 respectively. In PCR, Ail gene was not detected in all of 23 strains that determined non-pathogenic. In antimicrobial susceptibility test, twelve strains(52.2%) of 23 isolates showed the multi-resistant patterns with over 3 drugs. PFGE was performed after the genomic DNA of twenty three isolates, which was digested with Xba I. the 23 isolates showed 12(A~L) PFGE type.
북한산 국립공원 주변지역 사육견의 광견병 항체 분포조사(2)

Recently, the rabies cases have been reported in Paju-and Yangju-gun, Gyeonggi province near Seoul metropolitan area. The Pukhansan national park, nearly located from the cities, is suspected to be a high risk area for incidence and spread of the rabies to metropolitan area. This study was conducted to investigate the prevalence rate for rabies antibody of dogs near the Pukhansan national park and in some other districts in Seoul metropolitan city. From march to october 2001, a total of 306 serum samples were taken from dogs for breeding(189) and pet dogs(117) in 4 districts near the Pukhansan national park and other districts of Seoul. Rabies virus antibodies in sera were detected by neutralizing peroxidase-linked assay(NPLA). Of the 306 sera of dogs tested, 74(24.2%) were positive to rabies virus antibody. The prevalence rates of rabies antibody in Pukhansan national park area and in the other districts of Seoul city were 23.7% and 25.3%, respectively. There was no significant difference in the prevalence rate between these two districts. The prevalence rates of rabies antibody in pet dogs and dogs for breeding were 40.2% and 14.3% respectively. The prevalence rates of rabies antibodies in less than 1 year, 1~<2 years, 2~<3 years, and over 3 years old dogs were 14.5%, 22.4%, 32.6%, and 27.1%, respectively, and overall 24.2% in the dog population. In addition, we found that dogs less than 1 year old had lower antibody prevalence than those over 1 year old. It was concluded that enhancement of vaccination is important in the prevention of the rabies, and that rabies vaccines should not be less supplied than the population of the dog.
Pulsed-field gel electrophoresis(PFGE)를 이용한 Listeria monocytogenes의 molecular typing

A total of 1,354 samples was collected from bovine and porcine carcass from January 2005 to December 2006 in a slaughter house. Twenty five strains(1.8%) of L monocytogenes were isolated from 1,354 samples using selective media. Ten(1.4%) L monocytogenes were isolated from the 677 of bovine carcasses, and 15(2.2%) were isolated from the 677 of porcine carcasses. Among 15 L monocytogenes from porcine, 11 siolates were serovars 1/2c, followed by 1/2b (3 strains, 20.0%) and 1/2a(1 strain) Out of 10 bovine samples, positive cases in 1/2a were 9 strains (90.0%), 1/2b were 1 strains(10.0%). PCR primers were selected to amplify a 520-base pair(bp) DNA fragment from the listeolysin O gene (hlyA) of L monocytogenes. A 520-bp product was detected in PCR with DNA from L monocytogenes, but not from the other Listeria species tested. A total of 25 L monocytogenes strains were analysed by PFGE after digestion with ApaⅠ. PFGE analysis of genomic DNA showed the 14~18 fragments ranging in size from 30 to 550 kb, resulting in 14 patterns.

From February 2000 to December 2001, A total of 1,785 samples was taken from beef and pork carcasses in Seoul. Seven(0.69%) Listeria spp. were isolated from the 1,014 of beef carcasses, and five(0.65%) were isolated from the 771 of pork carcasses. The isolates were identified L monocytogenes by API listeria, and VIDAS LMO kit, serological test and PCR assay were performed. A total 12 strains of L monocytogenes were isolated form samples tested and all of the strains were classified into serotype 1. PCR primers are selected to amplify a 520-base pair(bp) DNA fragment from the listeolysin O gene(hlyA) of Listeria monocytogenes. A 520-bp product was detected in PCR with DNA from L monocytogenes, but not from the other Listeria species tested.
