      • Lactobacillus acidophilus 88과 Lactobacillus bulgaricus IFO 13953간의 세포융합주의 특성에 관한 연구

        유산균의 균주개량방법의 일환으로 protoplast fusion 기법과 electrofusion법을 이용하여 protease 활성, lipase 활성, 내열성, 내산성 등이 우수한 L. bulgaricus 와 bacteriocin을 생산하는 L. acidophilus를 융합시켜 얻은 융합주들의 생리학적 성질을 검토하였다. 산 생성능, 내열성, 내산성, protease, lipase 활성 등은 L. bulgaricus가 L. acidophilus 보다 우수하였다. L. bulgaricus는 lactose와 sorbose를 이용하였으나 maltose와 sorbitol을 이용하지 못하는 반면, L. acidophilus는 maltose를 이용하고 lactose와 sorbose를 이용하지 못하였다. 융합주 가운데서는 3, 6, 7, 8, 10번이 모균주의 발효능 특성을 함께 지님으로서 재조합체임을 확인 할 수 있었으며, sorbitol의 경우 모균주에서는 발효능이 전혀 나타나지 않았음에도 불구하고 융합주 4, 7번이 발효능을 나타내어 융합과정에서 새로운 형질을 획득하기도 한다는 사실을 알 수 있었다. Lactase 활성은 모균주 모두 높은 β-galactosidase 활성을 보였으나, phospho-β-galactosidase 활성은 거의 없었으며 융합주들도 다소 차이는 있었지만 모균주와 유사한 효소 활성을 나타내었다. 발효에 있어서 key enzyme으로 작용하는 protease, lipase 등의 효소 활성도 모균주의 활성 보다 우수한 융합주도 존재하였다. Aninterspecific fusant was made from the protoplasts of two strains of lactobacillus genus (streptomycin and lincomycin resistant L. bulgaricus and kanamycin resistant L. acidophilus 88). The functional properties of the fusant were examined by determining bacteriocin productivity, acid producing activity, ability of carbohydrates fermentation and three important enzyme activities. The recombinant strain revealed bacteriocin productivity. Acid production and β galactosidase, phospho-β-galactosidase, lipase and protease activity of L. bulgaricus were better than those of L. acidophilus 88. Among fusants, β-galactosidase activity of two strains were better than that of the parent strains but phospho-β-galactosidase activity remarkably lower. One fusant revealed the improved proteolysis compared to that of the parent strains- Lipase activity of L. bulgaricus was better than that of L. acidophilus but another fusant exhibited the highest lipase activity.

      • Siderophore A3에 의한 Pseudomonas synxantha A3의 철(Fe) 흡수와 그 Receptor Protein에 관한 연구

        P. synxantha A3로 부터 siderophore A3의 생성이 결핍된 변이주(fluroescent negative;Flu^- )와 siderophore A3에 의한 철의 흡수가 결손된 변이주(iron uptake negative;Iu )를 얻었다. P. synxantha A3의 생육은 EDDA농도가 증가할수록 감소 하였으며, siderophore A3의 생성도 균의 생육과 함께 감소하였다. 철(Fe)이 고농도로 존재하는 상태(100μM)에서의 생육정도는 P. synxantha A3와 Flu^- 변이주의 경우 비슷한 생육도를 보였으며, 철이 저농도로 존재하는 상태(0μM)에서는 P. synxantha A3의 성장속도는 약간 느리나 Flu^- 변이주보다 약간 높은 생육도를 보였다. 여과멸균한 Fe-siderophore A3 complex를 100μM/ml 되게 최소배지에 첨가한 상태에서의 생육도는 P. synxantha A3가 가장 양호하였으며, Flu^-, Iu^-변이주의 순으로 생육정도를 나타내었다. Citrate농도가 높아질수록 균의 생육은 증가하였으나, siderophore A3 생성은 저하되었다. P. synxantha A3의 경우 복합배지에서 자란 균체보다 siderophore A3생성용 죄소배지에서 자란 균체에서 약 150,000 dalton정도의 2개의 protein band가 더 존재하였으며, Iu^- 변이주의 경우 M.W.30,000과 45,000사이에 뚜렷한 2개의 protein band가 형성되었다. In an iron-limited enviornmemt, pesudomonas synxantha A3 produces a yellow-green fluorescent siderophore called sideropable of iron uptake(Iu^-)from Fe-siderophore A3 complex and a nonfluorescent mutamt(Flu^-)unable to synthesize siderophore a# were obtained, and their outer membrane protein(OMP) extracts were analyzed on SDS-PAGE. When increase the concentration of ethylendiamine-N,N'-diacetic acid(EDDA) in medium, the growth and production of siderophore A3 of P. synxantha A3 was gradually decreased and rapidly reduced ovwe 1.0㎎/ml of EDDA. Responding to the increased concentration of Fe(Ⅲ)and Fe-siderophore A3 complex in the iron limitted siderophore A3-production medium, the growth of P. synxantha A3, Flu mutant and Iu^- mutant were increased, but the that of Iu-mutant was much lower. Depending on the increase the concentrations of citrate, the grpwth of the test strains was increased but the production of siderophore A3 was inhibited. When P. synxantha A3 was cultured in siderophore A3 production medium, two more bands around the zone of M.W. 150,000 were identified. Qnd two distinct protein bands between the zone of M.W.30,000and 45,000 were also identified in OMP from Iu^- strain compared to that of mother strain.

        I have synthesized the values of music education in this thesis. In order to reach the goal, antithetical approaches have been used. The substance of my study is as follows. 1. The values of education and arts: the conceptions of value, the theories of education values, the characteristics and the values of arts. 2. The characteristics and the values of music; the characteristics of music, the values of music as an art, the values of music as a function. 3. The values of music education; 1) Aesthetic value―This value is the embodied value of music. This has been studied in absolute standpoint and referential one. 2) Functional values―These are the external values of music. These have emotional value, effective value and cultural one. ① Emotional value―This value has been studied in the viewpoint of Apollo and Dionysus. ② Effective value―Music is used in industry, advertisement, therapy, mood, realistic matter etc. The effectivenesses of music have been studied in the viewpoint of artistic intention and effective one. ③ Cultural value―This is an embodied value and simultaneously a window recognizing music. This has been studied in the viewpoint of customary concept and inherent one. If the values of music education are appropriatly modulated by the elements of region, culture, nation, society etc., the value system of music education will be desirably taken a triangular position.

      • 旋律創作學習의 構造的 接近

        It is the purpose of this thesis that know the function of scale degrees and the melodic movement of the stable and active tones. I studied as follows. 1. the nature of melody through the acoustic, aesthetic, psychological viewpoints. 2. the function of scale degrees in tonality. 3. the relation of the scale degrees and the other musical elements. 4. the melodic movement of the stable tones and the active ones. 5. the application of the stable and the active tones in melodical composition of primary school. The function of the scale degrees are different each other. In tonality, the points of repose are 1st.,3rd. and 5th. degree. The others are active. The function of the scale degrees has the influence of tone system, harmony, rhythmic pattern, meter, accent, tempo, structure(phrase etc. ), cadence etc,. Melody is figured by the rectilineal, recurrent, enriched, weakened, transfered, mixed movement of the stable and the active tones, Using the function of the scale degrees in tonality, we shall have the organic and actual guidance of melodical composition.

      • 濟州島 勞動謠의 리듬적 特性과 勞動行爲와의 관계

        The work song is the Sympathetical products of regional society. For that reason, the musical characteristics of worksongs have been influenced by the outside factors of music. The most important of the factors is the work behaviors and the common personality of performers. In this treatise, it is my object to study on the correlation of the work behaviors, the regional personality and the musical characteristics of Cheju worksongs. The nature and life environments of Cheju island are very rough. And then, the members of Cheju regional society have tried to vanquish them. Through the long time and tide, their endeavours have made the work behaviors of Cheju worksongs collective, various, open and practical. And they have had the common personality that is cooperative inside and competitive outside. The characteristics of Cheju working groups have been made by a connection with the nature and life environments and the regional personality of Cheju island. The characteristics are as follows. ① The working groups of Cheju island are primary. ② They have been made by the spontaneous paticipation of their members who work in a permitted process and a conventional role than a interpersonal role. ③ The members promote the establishment of empathy and identify the work behaviors with the collective empathy. In view of the results that are studied with the various things as mentioned above, we know that the musical characteristics of Cheju worksongs are connected with the work behaviors and the regional personslity of Cheju island. The correlation among them is as follows. ① The worksongs of formal meter have the work behaviors that are regular, simple and collective, but the nonformal have the work behaviors that are irregular, complex and personal. ② It is easy that we find the three and nonsymetric dividing and the scotched rhythms(?? etc.) in Cheju worksongs. Such the rhythm patterns are connected with the common personality of Cheju regional society. ③ The melodic phrases of formal metric worksongs have <the strong-weak> accent pattern which is connected with the working motion that have <the strong-weak> one. And the accent pattern of nonformal metric worksongs depends on the nonformal accent pattern of the working motiom. ④ The fast worksongs have a fast working motion, and the slow have a slow or irregular working motion. The meaning of Cheju worksongs has been made by the sympathetic emotion of Cheju regional society, which is a result of significant forms. The most important of the significant forms is the work behaviors. Therefore the real musical meaning of Cheju worksongs comes from a connection with the work behaviors.

      • 김치 유산균인 Lactobacillus sp. JJ-2001이 생산하는 Bacteriocin에 관한 연구

        The microorganisms producing bacteriocin from kimch were screened, and among them,a strain showing the highest bacteriocin activity and stability was isolated. Based on the taxonomic characteristics of its morphological, cultural and physiological properties, the isolatef strain was characterized as Lactobacilius sp..To identify optimal conditions for the growth and the production of bacteriocin from the isolated strain,nutrient source, initial pH, and culture temperature were evaluated. Physiochemical properties of bacteriocin was also investigated. The potimal condition for the production of bacteriocin and the cell growth was obtained at 2% glucose, 1% beef extract as organic nitrogen source,1%(NH_4)_2SO_4 as inorganic nitrogen source,and 0.1% Tween 80. The isolated strain showed the maximum growth at 25℃. The inhibitory range of its antimicrobial substance aginst gram positive bacteria was marrow and its inhibitory activity was maintained between pH3.0 and pH8.0. The antimicrobial activity of bacteriocin from the isolated strain was stable at 50℃.
