        NOS 만성억제로 인한 고혈압에서 아드레날린성 활성증가기전

        Nitric oxide is a tonically produced vasodilator that maintains blood pressure in the normal animal. The chronic inhibition of nitric synthase (NOS) elicits the hypertension in rats. However, the mechanism of hypertension indused by chronic inhibition of NOS is not clear. Thus, to clarify the mechanism of the occurance of hypretension, the changes in alpha-adrenergic systems in rats treated with NOS inhibitors for 21 days were examined. Chronic administration of L-NAME significantly increased in the basal blood pressure, but chrinic administration of 7-nitroindazole did not. Phenylephrine and G-protein stimulator elicited the more potent contraction in the aorta of the L-NAME-induced hypertensive rats. However when the contractile responses by phenylephrine and G-protein stimulator were calculated the the proportion to the contraction by 24 nM KCL, there was no difference between the vehicle-treated rats and the L-NAME-treated rats. The results suggest that hypertension induced by chronic inhibition of NOS is due to the inhibition of eNOS and the increased reponses to the adrenergic drugs are due to the changes of the intracellular contactile mechanism of aoric tissue rather than the changes of receptor density.

        리우올림픽 태권도경기의 한국선수와 외국선수의 경기운영 분석을 통한 경기력 향상 방안

        The purpose of this study is to identify the ways to improve competitiveness through analyzing the competitions of Korean and foreign players in the Liu Olympic Taekwondo competition. In 2016, we analyzed the frequency of technology in the competition from the 32th round of the Olympic Games to the finals of 8 Olympic men and women, and confirmed the difference between Korean and foreign athletes. Based on this, we propose concrete game performance improvement strategy and game operation strategy as follows. First, the kicking technique, which is mainly expressed in the Olympic competitions, shows that the technique of straight forward direction is used frequently with round house-kick and pushing-kick, and a variety of kicking due to the differential score system is expressed The frequency of presentation by technology was found to be very different.Second, there was a significant difference between the foreign player group and the Korean player group in round house-kick, turning hook-kick, and punch kick technique.Third, there is a significant difference between the foreign player group and the Korean player group in the front hook-kick and turning hook-kick technique.

        태권도장 지도자의 윤리적 리더십

        In order to search for the ethical leadership of taekwondo leaders, this research is examined with the separation of respect, service, equity and justice, probity, and seeking common profit. The conclusion for the virtues of ethical leadership that taekwondo leaders should consist are as follow. First of all, leaders should respect taekwondo itself, and the virtue of respect can be enacted with the attitude of consideration in a horizontal relationship. Second of all, as taekwondo leaders are both educators and businessmen, they should have the attitude for education by equipping with various facilities and should wholeheartedly consult with the parents regarding trainees’ development and any notifications. Thirdly, within the relationship between taekwondo leaders and the instructors, the employers should secure equity and justice regarding their labor. Proper reward and punishment should also be applied. Fourthly, leaders should prepare theory and practical ability to not degrade the original value of taekwondo, and through correct attitude, the virtue of probity could be fulfilled. Fifthly, through open judgement, outdoor demonstration and volunteer work, the virtue of seeking common profit could be fulfilled.

        트레드밀 지구성 운동이 streptozotocin으로 유발된 당뇨 흰쥐의 뇌에서 PGC-1α, GLUT-1, Tfam 단백질 및 항산화 효소(Cu, Zn-SOD, Mn-SOD)의 발현량에 미치는 영향

        이 연구는 지구성 운동이 streptozotocin (STZ)으로 유발된 제1형 당뇨 특징을 가진 쥐 뇌의 글루코스 운반, 미토콘드리아 기능 및 항산화효소 단백질 발현에 미치는 영향을 규명하는데 목적이 있다. 제1형 당뇨 모델 쥐는 50 ㎎/㎏의 streptozotocin을 수컷 Sprague-Dawley (SD) 흰쥐의 복강에 1회 주입하여 생산하였으며 본 실험 시집단은 NON-STZ 집단(n=8), STZ-CON 집단(n=8) 및 STZ-EXE 집단(n=8) 등 3집단으로 구분하여 실시하였다. 트레드밀 지구성 운동은 총 6주, 주 5일, 2주 간격으로 속도를 약 3∼4 m/min으로 점증적으로 증가시켰으며 운동시간은 1주와 3주차에 10분씩 증가시켰다. 분석 결과 혈청 글루코스 수준은 STZ-EXE 집단은 STZ-CON 집단에 비해 현저하게 감소(p<0.05)하였으며 PGC-1α (p<0.001), mtPGC-1α (p<0.001), GLUT-1 (p<0.001), Tfam(p<0.001), Cu,Zn- SOD (p<0.001), Mn-SOD (p<0.01) 경우도 STZ-EXE 집단이 STZ-CON 집단에 비해 현저하게 증가하였다. 이러한 결과는 장기간 지구성 운동이 뇌의 글루코스 이용능력과 관련된 단백질인 GLUT-1과 미토콘드리아 기능 향상과 관련된 단백질인 PGC-1α과 Tfam을 증가시키고 산화적 스트레스의 방어 기전으로서 역할을 수행하는 항산화 효소인 Cu,Zn-SOD와 Mn-SOD를 활성화시키는데 긍정적인 역할을 수행한 것으로 나타났다. The purpose of this study is to identify the effects of exercise training [ET, 10~18 m/min (speed), 20~30 min (exercise duration)/a day for 5 day/wk, 6 wk) on PGC-1α, GLUT-1, Tfam, Cu,Zn-SOD and Mn-SOD proteins in brain of STZ-induced diabetic rats. The male Sprague-Dawley (SD) rats were single-injected intraperitoneally with 50㎎/㎏ of streptozotocin (STZ) to produce STZ-induced diabetic rats. Rats were divided into 3 experimental groups with 8 rats in each group, as follows: (1) non-STZ group (n=8), (2) STZ-CON group (n=8), (3) STZ-EXE group (n=8). The results of this study suggest that i) serum glucose level was significantly reduced in STZ-EXE group compared with STZ-CON group (p<0.05), ii) PGC-1α (p<0.001), mtPGC-1α (p<0.001), GLUT-1 (p<0.001), and mtTfam (p<0.001) proteins in brain of STZ-induced diabetic rats were significantly increased in STZ-EXE group compared with STZ-CON group, iii) Cu,Zn-SOD (p<0.001) and Mn-SOD (p<0.01) proteins in the STZ-induced diabetic rats were significantly increased in STZ-EXE group compared with STZ-CON group. In conclusion, the findings of the present study reveal that treadmill exercise training increases brain GLUT-1 protein level possibly through up-regulation of PGC-1α and Tfam proteins which represent key regulatory components of stimulation of brain mitochondrial biogenesis. In addition, treadmill exercise training may prevent oxidative stress by up-regulation of Cu,Zn-SOD and Mn-SOD proteins in the STZ-induced diabetic rats.

        인삼 Total Saponin , Protopanaxadiol 및 Protopanaxatriol 의 소염작용 (消炎作用) 에 관한 연구

        In an attempt to elucidate the anti-inflammatory action of ginseng total saponin, protopanaxadiol and protopanaxatriol, the anti-inflammatory activity of three compounds was investigated under various acute and chronic inflammatory models. The blood vessel permeability was slightly inhibited by total saponin and protopanaxatriol treatments. Granuloma formation induced by 2% carrageenan was inhibited by total saponin and protopanaxatriol. The cotton-pellet granuloma formation was significantly inhibited by intraperitoneal injection of total saponin. Total saponin and protopanaxadiol inhibited leukocyte emigration and protein exudation in CMC-induced pouch but protopanaxatriol increased leukocyte emigration. The swelling of rat hind paw induced by 1 % carrageenan was significantly inhibited by total saponin, protopanaxadiol and protopanaxatriol both single and 2 weeks treatments. Total saponin, protopanaxadiol and protopanaxatriol decreased the anti-inflammatory activity in adrenalectomized rat. Our results suggest that total saponin, protopanaxadiol and protopanaxatriol have potent anti-inflammatory activity, this may be mediated in part through stimulation of adrenal glands.

      • Production and characterization of monoclonal antibodies against Clostricium botulinum type B toxin

        Clostridium botulinum type B toxin was purified by sequential steps of colunm chromatography with DEAE-Sephadex, Sephadex G-200, Sephacryl S-300, and DEAE-Sephadex. Monoclonal antibodies against type B toxin were prepared by fusion of myeloma cells SP2/O-Ag14, with spleen cells from the BALB/c mice immunized by type B toxoid. Growing hybridoma cells were screened by enzyme-linked immunosorbent assay (ELISA) and two cell lines, B2 and B3, wre cloned by limiting dilution The two monoclonal antibodies reacted with heavy chain of the toxin and B3 highly neutralized the toxin.
