Background : Ginseng seeds are harvested in immature stages and take 3 - 4 month to be germinated following cold stratification. Improvement of germination speed can be a valuable traits to be characterized for economically important medicinal plant. Modulation of cell wall compositions is also considered as another agricultural trait when the crops can be processed for foods and forages. Here we show the functional characterization of ginseng pPLAIIIβ focused on its possible values on germination and modulation of cell wall compositions. Methods and Results : Patatin-related phospholipase AIII family genes were identified from ginseng EST clones and further confirmed by PCR. Clone showing the highest homology with pPLAIIIβ was overexpressed in model plant Arabidopsis, and it displayed dwarf plants. qPCR analysis showed that pPLAIIIβ expressed in all organs of 2-years-old ginseng. Overexpression of ginseng pPLAIIIβ also displayed apparently enlarged seeds as well as altered germination speed in early stage compared to the control. Chemical stainings and direct quantification of lignin and cellulose were performed to understand the link of cell elongation patterns and cell wall compositions. Conclusion : Overexpression of ginseng pPLAIIIβ inhibited longitudinal cell elongation in all tested organs except seeds which is enlarged in both directions than the control. Shorter root length is related with auxin responsive genes and its dwarf morphological phenotype is resulted in altered cell wall compositions.

Background: The patatin-related phospholipase AIII family (pPLAIIIs) genes alter cell elongation and cellwall composition in Arabidopsis and rice plant, suggesting diverse commercial purposes of theeconomically important medicinal ginseng plant. Herein, we show the functional characterization of aginseng pPLAIII gene for the first time and discuss its potential applications. Methods: pPLAIIIs were identified from ginseng expressed sequence tag clones and further confirmed bysearch against ginseng database and polymerase chain reaction. A clone showing the highest homologywith pPLAIIIb was shown to be overexpressed in Arabidopsis using Agrobacterium. Quantitative polymerasechain reaction was performed to analyze ginseng pPLAIIIb expression. Phenotypes were observedusing a low-vacuum scanning electron microscope. Lignin was stained using phloroglucinol and quantifiedusing acetyl bromide. Results: The PgpPLAIIIb transcripts were observed in all organs of 2-year-old ginseng. Overexpression ofginseng pPLAIIIb (PgpPLAIIIb-OE) in Arabidopsis resulted in small and stunted plants. It shortened thetrichomes and decreased trichome number, indicating defects in cell polarity. Furthermore, OE linesexhibited enlarged seeds with less number per silique. The YUCCA9 gene was downregulated in the OElines, which is reported to be associated with lignification. Accordingly, lignin was stained less in the OElines, and the expression of two transcription factors related to lignin biosynthesis was also decreasedsignificantly. Conclusion: Overexpression of pPLAIIIb retarded cell elongation in all the tested organs except seeds,which were longer and thicker than those of the controls. Shorter root length is related to auxinresponsivegenes, and its stunted phenotype showed decreased lignin content.

The phenylurea herbicides are used to control annual and perennial weeds on crop cultivating fields. The excessive usage of these agrochemicals increase many environmental problems. Thus, engineering transgenic plant for herbicide metabolism can provide efficient and eco-friendly means for enhanced phytoremediation capacity. Cytochrome P450 enzymes comprise one of the major plant enzyme families that mediate the oxidative degradation of xenobiotic chemicals, including herbicides. Considering these notions, phytoremediation properties of transgenic ginsengderived PgCYP76B93 in Arabidopsis to phenylurea herbicides were assessed. Phylogenetic tree of PgCYP76B93 clustered in between close to the herbicide metabolism-related enzyme families and terpenoid biosynthesis-related. The expression of PgCYP76B93 was considerably upregulated upon treatment with phenylurea herbicide, chlorotoluron. Simulated docking using Autodoc program predicted possible interaction with chlorotoluron. Transgenic Arabidopsis plants overexpressing PgCYP76B93 were resulted in slightly reduced plant height with relatively small leaves. The lower plant height in the PgCYP76B93-overexpressing line than in the control revealed that it was linked to the expression of gibberellin oxidases (GAox). The bioassay of transgenic plants growing on herbicide-containing media revealed enhanced resistance against chlorotoluron.

Professors can create online meeting room directly from their courses by using the Adobe Connect Meeting module. The web services from the server was utilized for the application integration.

A SMS Moodle plug-in block was developed for the Pai-Chai Moodle virtual learning environment. Professors can send SMS messages directly from their courses by using the SMS block. NuSOAP open source web service library was utilized for XML SOAP based message transfer.
