ICR 종 생쥐 간에서의 δ - Aminolevulinate Dehydratase 동위효소의 분리 및 정제

Two different molecular forms of hepatic δ-aminolevulinate dehydratase (EC 4. 2. 1. 24) have been isolated and purified by slightly modified method of Doyle and Schimke (1969) on DEAE-cellulose column chromatography by linear gradient elution with 0.4 M NaCl solution from 125 livers of the ICR inbred strain of adult mice. The enzyme fraction I and II were eluted at a NaCI concentration of between 0.05∼0.1M, and between 0.2∼0.3 M, respectively, on the DEAE-cellulose column chromatography, which was equilibrated with 25 mM potassium phospate buffer, pH 6.8 (column size 1 × 50 cm). The specific activity of the enzyme fraction II was 3.5 times as high as that of the enzyme fraction I. Molecular weight for the enzyme fraction I and II was 174,000 and 275,000, respectively, as determined by acrylamide disc gel electrophoresis and by Sephadex G-120-200 gel filtration.
BP부산물을 재활용한 알칼리활성화 지반개량재의 지반개량효과에 관한 연구

The enormous quantity of 'Bayer-Process by-products' (BP by-products) discharged by industries producing alumina from bauxite represents an environmental and economical problem. As it is mainly composed of $Fe_2O_3$, $Al_2O_3$, $SiO_2$, CaO and $Na_2O$, it is thought that using BP by-products as a construction material is an effective way to consume such a large quantity of alkaline waste. In this study, This study evaluates the effect of alkali-activated binder based on recycling BP by-products on soil improvement through the evaluation of slope stability and seepage flow numerical analysis. The results of analysis of ground slope safety at dry season and wet season meet standard (Ministry of Land, Infrastructure and Transport, 2006) Especially, when wet season, the ground used soil improving material meet standard, while the ground used soil-nailing method doesn't. Also, permeability coefficient of improved soil is smaller than that of natural soil and saturation depth of reinforced ground surface with improve soil is lower than that of natural soil. 연구에서는 BP부산물을 혼입한 알칼리활성 무시멘트 지반개량재의 사면안정 및 침투류 수치해석을 통하여 원지반토와 비교하여 개량토의 지반개량 효과를 검토하고자 하였다. 그 결과 사면안정해석을 실시한 결과에서는 건기시, 우기시 모두 건설공사 비탈면 설계기준(국토해양부, 2006)의 기준안전율을 만족하는 것으로 확인되었으며, 우기시 쏘일네일링으로 기준안전율을 만족하지 못하는 사면이 개량토로 보강하여 기준치를 만족한 것으로 나타났다. 또한 침투류 해석을 실시한 결과 개량토의 투수계수가 작아지는 것으로 나타났으며, 원지반토에 비하여 개량토로 표면보강된 경우는 포화심도가 낮아진 것으로 나타났다.

ICR 종 친근교배계 성숙 생쥐 125마리에서 얻은 간 128.8gm에서 $\delta$-ALAD를 Doyle & Schimke(1969)의 변법으로 정제하였던 바, DEAD-cellulose column chromatography 할 때 NaCl 농도 0.05M와 0.2~0.3M에서 유출되는 두 종의 효소분획을 얻었다. 이 두 효소분획을 polyacrylamide disc gel electrophoresis 하여 효소단백질의 이동율로부터 분자량을 산출하였던바, 효소품획 I의 분자량은 174.000이고, 효소분획 II의 분자량은 275,000으로 밝혀졌다. 이상의 성적은 ICR종 친근교배계 쥐의 간 조직내 $\delta$-ALAD에는 2개의 동위효소가 존재하고 있음을 시사하였다. 도플러 초음파를 이용한 개에서 마취에 따른 간문맥 혈류량 변화의 측정

Portal bood flow was measured with pulsed Doppler ultrasound in twenty anesthetized dogs. In anesthetized dogs with xylazine HCl, the average of portal blood flow velocity was reduced($11.68{\pm}1.55cm/sec$ vs. normal $16.67{\pm}1.77cm/sec$). Average portal blood flow was also decreased compared to normal($28.36{\pm}11.61ml/min/kg$ vs. normal $43.12{\pm}14.46 ml/min/kg$). And congestion index was increased($0.0368{\pm}0.0117cm{\cdot}sec$) vs. normal $0.0297{\pm}0.0062cm{\cdot}sec$). In anesthetized dogs with ketamine HCl, portal blood flow velocity was slightly increased ($22.62{\pm}2.53cm/sec$ vs. normal $16.67{\pm}1.77cm/sec$. Also mean portal blood flow was slightly increased($43.12{\pm}14.46ml/min/kg$ vs. normal $55.32{\pm}19.99ml/min/kg$). In anesthetized dogs with tiletamine and zolazepam, portal blood flow velocity and portal blood flow were unchanged.

Radiograph and serum activities were investigated in six dogs with experimental portal vein branch ligation. Operative mesenteric portography showed the incomplete portal vein circulation due to ligation of portal vein branch. Several serum enzyme activities were measured after portal vein branch ligation. Albumin and total protein were decreased following the time. Others were increased. In microscopic findings, hepatocellular necrosis and haemorrhage were identified in midzone and centrilobular zone. And reverse lobulation pattern was found in dogs with portal vein branch ligation.
