황백(黃柏)의 berberine이 DNA의 기능조절에 미치는 영향에 관한 형광이방성 연구

Objectives : We tried to observe the fluorescence anisotropy and intensity of ethidium ion in the intercalating binding interaction between DNA and ethidium ions in the presence of berberine, and then tried to explain the effect of berberine on the intercalating interaction of ethidium ion with DNA. Methods : DNA(calf thymus DNA), berberine and ethidium bromide(EtBr) were purchased from Sigma-Aldrich Co. Proper amount of each compound was dissolved in 20 mM sodium phosphate buffer(pH 7.0) containing 100 mM of NaCl to prepare stock solutions. Collections of the fluorescence anisotropy and intensity data were performed on JASCO FP-8300 spectrofluorometer equipped with a polarizer and a Peltier temperature controller. The excitation of ethidium ion was done at 550 ㎚ and the emission data were collected at 600 nm. For Stern-Volmer plot, the fluorescence data were collected at 18℃ and 30℃. Results : According to the results of this research, the weak competitive binding pattern between ethidium ion and berberine appeared in binding with DNA at low ratio of DNA to ethidium ion. But at high ratio of DNA to ethidium ion, this weak competition disappeared. Instead, berberine might bind to DNA by intercalating way. In other words, berberine could de-intercalate ethidium ion from DNA at low concentration of DNA relative to ethidium ion, but could not at high concentration of DNA relative to ethidium ion. In addition, the mechanism of fluorescence quenching of ethidium ion could also proceed differently, depending on the ratio of the amount of DNA to that of ethidium ion. Conclusions : The effect of berberine on the DNA-ethidium ion intercalating interaction could work differently, depending on the relative ratio of the amount of DNA to that of ethidium ion. This study also showed that fluorescence anisotropy analysis is very useful method to obtain detailed information for investigation of the complex binding interactions. In order to fully understand the mechanism of action of the pharmacological effect by berberine, studies on the effect of berberine on the action of proteins such as various enzymes closely related to berberine-induced medicinal effects should be continued.

본 연구에서는 하나의 제어장치로 서로 직교하는 2방향의 건물응답을 동시에 제어할 수 있는 동조액체질량감쇠기(Tuned Liquid Mass Damper; TLMD)를 제안하고 제어성능을 실험적으로 검증하였다. 본 연구에서 제안된 TLMD는 한 방향으로는 동조액체기둥감쇠기(Tuned Liquid Column Damper; TLCD) 내부에 채워진 액체의 운동에너지를 이용하여 구조물의 응답을 제어하게 된다. 그리고, 다른 한 방향 즉 TLCD의 직각 방향으로는 LM guide(linear motion guide) 위에 놓인 TLCD 수조와 내부의 액체의 질량을 이용하여 동조질량감쇠기(Tuned Mass Damper; TMD)로 거동하게 함으로써 구조물의 응답을 감소시킨다. 이와 같은 TLMD의 양방향 독립거동 특성을 증명하기 위해 실물크기의 구조물에 설치하여 강제진동실험을 수행하였다. 실험결과, 양방향 모두 대상 구조물의 응답을 감소시키는 것을 확인하여 제안된 TLMD의 효용성을 검증하였다. In this paper, a tuned liquid mass damper(TLMD) was proposed and experimentally investigated on its control performance, which can control bi-axial responses of building structures by using only one device. The proposed TLMD controls the structural response in a specific one direction by using a liquid sloshing of TLCD. Also, the TLMD reduces the response of structures in the other orthogonal direction by behaving as a TMD that uses mass of the container itself and liquid within container of TLCD installed on linear motion guides. Force-vibration tests on a real-sized structure installed with the TLMD were performed to verify its independent behavior in two orthogonal directions. Test results showed that the responses of a structure were considerably reduced by using the proposed TLMD and its usefulness for structural control in two orthogonal directions.
실시간 하이브리드 진동대 실험법에 의한 양방향 TLMD의 진동제어 성능평가

An experimental real-time hybrid method, which implements the vibration control of a building structure with only a two-way TLMD, is proposed and verified through a shaking table test. The building structure is divided into the upper experimental TLMD and the lower numerical structural part. The shaking table vibrates the TLMD with the response calculated from the numerical substructure, which is subjected to the excitations of the measured interface control force at its top story and sinusoidal waves input at its base. The results show that the conventional method can be replaced by the proposed methodology with a simple installation and accuracy for evaluating the control performance of a TLMD.
LPG 가열로 및 건조로의 안전장치 설치에 따른 사고빈도에 관한 연구

본 연구에서는 국내 가열로·건조로의 안전장치 유무에 따른 위험변화를 평가하기 위하여 FTA(Fault Tree Analysis)를 통하여 안전장치들의 사고발생 빈도수 도출 및 분석을 실시하였다. 국내 LPG 가열로 및 건조로의 안전장치의 국내 설치기준이 미비하고 가스 누출하여 체류할 수 있는 구조로 폭발의 위험을 가지고 있다. 안전장치는 다른 설비의 연소기 국내 기준과 NFPA를 바탕으로 4가지를 선정하였다. 안전장치 설치 전후의 사고발생 빈도감소효과를 각각 분석하였다. 이를 통해 국내 LPG 가열로?건조로의 가스누출 피해 방지를 위한 최소한의 안전장치를 제시하였다. The purpose of this study is to assess the risk of depending on the presence or absence of safety device of domestic heating and drying furnaces, by derivation and analysis of accident frequency of safety devices through FTA (Fault Tree Analysis). Installation standards are lacking in Korean for the safety device of LPG heating and drying furnace, which have a risk of explosion due to structure to trap the leaked gas. Four different safety devices were selected on the basis of NFPA and national standards for combustors of other equipment. Effects of frequency reduction in accidents were analyzed before and after installing the safety devices respectively. As a result, a minimal leakage safety device was presented for preventing damages from gas leak of domestic LPG heating and drying furnace.

This paper presents a design of a tuned liquid mass damper(TLMD) for controlling bi-directional response of high-rise building structure subjected to windload. The proposed damper behaves as a tuned mass damper(TMD) of which mass is regarded as the mass of a tuned liquid column damper(TLCD) and the case wall of the TLCD itself in one direction and the TLCD in the other direction. Because the proposed device has coupled design parameter along two orthogonal directions, it is very important to select designing components by optimal fine tuning. In the designing TLMD, for easy maintenance, the rubber-bearing with small springs was applied in TMD direction. In this study, the Songdo New City Tower 1A in Korea, which has been designed and constructed two TLCDs in order to control bi-directional response, was chosen as the model building structure. The results of rotation test proved the effectiveness of bi-directional behavior of TLMD.
