Characteristics of genomic DNA of Lactobacillus spp. and Bifidobacterium spp. were determined by pulsed field gel electrophoresis and the following results were obtained; Four enzymes, Sma I, Apa I Not I and Sfi I were identified as suitable for generation of relatively few numbers of distinct fragments from the Lactobacillus spp. genomes, whereas five enzymes, Sma I, Apa I, Sfi I and Dra I were suitable for Bifidobacterium spp. genomes. The genome sizes of Lactobacillus spp. ranged from 1.S8Mb to 3.31Mb, the genome sizes of L. brevis ATCC 8287, L. fermentum ATCC 14931, L. casei YIT 9018, L. plantarum KCTC 1048 and L. delbruekii subsp. delbruekii ATCC 9469 were estimated to be 1.93Mb, 1.98Mb, 2.84Mb, 2.81M6 and 1.82Mb, respectively. Those of Bifidobcxterium spp. ranged from 1.85Mb to 2.83Mb. The genome sizes of B. bifrdum ATCC 29521, B. longum ATCC 15707, B. infantis ATCC 15697 and B. bifidum CU 1370 were estimated to be approximately 1.85Mb, 2.83Mb, 1.86Mb and 1.94Mb, respectively.

Four drugs, bithionol, hexachlorophee, hexachloroethane and tetrachloroethylene, were tested on the in vitro effects of vermicidal and laying capacity inhibitory action to the Eurytrema Pancreaticum of Korean cattle, and the results obtained were as follows : 1) Of the four drugs, bithionol was most effective at both high and low concentrations. 2) The effects of hexachlorophene, hexachloroethane and tetrachloroethylene were less effective than that of bithionol, and the laying activity, at low concentration, was observed to increase at the early stage and decrease gradually thereafter. 3) Tetrachloroethylene was least effective of the four drugs.

N/A Studies on the antimutagenicity of Lactobacillus spp. and Bifidobactrium spp. against 2-nitrofluorene have been conducted utilyzing Salmonella typhimurium TA 98 in order to characterize the activity by the starter and non-starter strains. The average antimutagenic activity of Lactobacillus spp. and Bifidobactrium spp. against 2-nitrofluorene was 20.29% and L. plantarum CU 722 revealed the greatest mutation inhibition activity of 50.34%. An intensive antimutagenicity was found in the cell wall and cytoplasm fraction of L. plantarum CU 722 in skim milk culture showing inhibition rate of 34.9% and 24.5% respectively and very low activity remained in cell free broth and in lactic acid The optimum cultivation time for Lactobacillus spp. and Bifidobacterium spp. to inhibit mutation was 24 hours and the optimum preincubation time of the reaction mixture containing the mutagen, lactic culture and indicator strain was 60 minutes, and the optimum incubation time for the test elates was 48 hours.

The antimutagenic activities of Lactobacillus spp. isolated from European kefir and yoghurt and those of non starter Lactobacillus and Bifidobaterium strains against 2-Nitrofluorene were studied by Ames test utilizing Salmonella typhimurium TA 98 to investigate probiotic effects of the strains. The optimal concentration of mutagen 2-nitrofluorene to produce 3,000 revertant colonies per plate turned out to be 10 ㎍/plate. Thirty six strains out of forty Lactobacillus spp. strains isolated from European kefir and yoghurt revealed antimutagenic activity and L. plantarum CU 722 showed the strongest antimutagenic activity of 50.34%. The antimutagenicity of non starter Lactobacillus and Brfidobacterium strains resulted in average 15% and 20% of mutation inhibition rate respectively. The effect of S-9 mix addition on the antimutagenicity rate against 2-Nitrofluorene was species and strain dependent.
Lactobacillus casei YIT 9018 에 의한 Aloe vera 첨가 액상 발효유의 성상

Parameters involved with addition of aloe very on liquid yoghurt fermentation were investigated in order to fortify the probiotic properties of the fermented milk. The following results were obtained; The maximum level of aloe very juice addition turned out to be 20% of skim milk substrate, acid production rate of Lactobacillus casei YIT 9018 increased by the addition of aloe vera juice into skim milk. L. casei viable cells of log 9.63 cfu/㎖ sustained in 20% aloe vera juice supplemented substrate and log 9.23 cfu/㎖ in control. The organic acid content analysis of the fermented milk by HPLC revealed that 1,133 ppm of lactic acid and 126 ppm of acetic acid in L. casei culture. There was no significant difference in organic acid content due to the aloe vera juice supplementation. The adverse effects in pH, titratable acidity, viscosity, precipitation and viable counts during 5 days storage in refrigerator were not observed in the aloe very supplemented liquid fermented milk. Panel test results of aloe vera juice supplemented liquid fermented milk showed that palatability score of 6.25, which was not statistically significant to 6.95 of the control.

Studies were carried out to investigate the nature of cell wall hydrolyzing enzymes of Lactobocillus spp. and Bifidobacterium spp. which were utilized as starter organisms for fermented dairy products, the following results were obtained; Thirteen out of twenty-one strains of Lactobacillus and Bifidobacterium strains showed intensive clear cell wall hydrolyzing zone around the colonies on MRS agar plate containing 0.2% lyophilized Micorcoccus luteus cells. Cell wall hydrolyzing enzymes were isolated by SDS-PAGE and the cell wall hydrolyzing bands were detected on M. luteus cell containing gels. Each strains varied in molecular weight of cell wall hydrolyzing enzyme and in the number of cell wall hydrolyzing enzyme. L. casei YIT 9018 possess a single cell hydrolyzing enzyme of 76kDa. Optimum temperature and pH of the cell wall hydrolyzing enzyme of L. casei YIT 9018 were 37℃ and pH 6.0. Monovalent cations inhibited the enzymatic activity at above 0.1M concentrations and divalent cations inhibited the enzymatic activity extensively.
