The purpose of this research was to investigate effects of Samultang water extract (SMT) on cytokine production from immune cells during the late stage of pregnancy in BALB/c mice. SMT(500 mg/kg) was administered p.o. once a day for 7 days, and then thymocytes and peritoneal macrophages were separated. At the late stage of pregnant mice, the proliferation of thymocytes and the production of ${\gamma}-interferon$ in thymocytes were decreased as compared with normal group, but the production of interleukin-2 and interleukin-4 was increased. The production of tumor necrosis $factor-{\alpha}$, nitric oxide and phagocytic activity in peritoneal macrophage was increased as compared with normal group. At the late stage of pregnant mice administered with SMT, the production of interleukin-2 in thymocytes was decreased as compared with a pregnant group, but the proliferation of thymocytes, the production of ${\gamma}-interferon$ and interleukin-4 was increased. The production of tumor necrosis $factor-{\alpha}$ and nitric oxide in peritoneal macrophages were decreased as compared with a pregnant group, but phagocytic activity were increased. These results suggest that SMT has the regulative action on immune function of thymocytes and peritoneal macrophages at the late stage of pregnant mice.

The oral administration of Aurantii nobilis pericarpium (ANP) extract and Aurantii immaturi pericarpium (AIP) extract suppressed the cell viability of both thymocytes and splenocytes in BALB/c mice. The ANP extract (500 mg/kg) enhanced the population of $B220^+$ cells, and the AIP also enhanced the population of B220+ and Thy-1+ cells in splenocytes. The AIP extract enhanced the population of $CD4-CD8^+$ cells in splenic T-lymphocytes. However, the ANP did not affect, whereas the AIP enhanced the phagocytic activity and the nitric oxide production in peritoneal macrophages.

The studies were conducted to investigate the combined effects of several combined preparation of crude drugs and mitomycin C(MMC). The combined effects on the proliferation of Molt-4 cells and activation of human lymphocytes were estimated by MTT colorimetric assays. The drugs itself enhanced the proliferation of Molt-4, but the inhibitory action of MMC was not affected by the combined treatment of the drugs and MMC. Among 9 kinds of the drugs, Sip Jeon Dae Bo Tang(SDT), Saeng Maek San(SMS) and Kwi Bi Tang(KBT) did not inhibit the action of MMC, but activated lymphocytes. When the mice were treated by MMC, the number of leukocytes was decreased significantly at the 1st day, but recovered at the 7th day. In the groups of MMC treated with SDT or KBT, the number of leukocytes was increased significantly than the group of MMC treated only at the 3rd day. The combined treatment of SDT, SMS and MMC retained the body weight of mice at the level of normal mice. The SDT, SMS and KBT did not change the number of plaque forming cells(PFC) and the proliferation of T cells. The combined treatment of SDT and MMC increased the number of PFC significantly than the MMC treated group. The combined treatment of SDT, SMS, KBT and MMC increased the T cell proliferation significantly than the MMC treated group. In conclusion, it is suggested that SDT, SMS and KBT can recover the side effects of MMC, such as weight loss, leukopenia and immunosuppression, without any intercalating the anti-proliferative action of MMC in vivo.

Mortality test has been utilized as a basic method for systemic anaphylactic reaction. Compound 48/80 has been used as a direct and convenient reagent to study the mechanism of anaphylacic reaction. The aqueous extracts of 102 medicinal plants were screened for mortality test using compound 48/80. Sixteen out of the 102 medicinal plants exhibited more than 50% of inhibition on mortality test by their total aqueous extracts with 0.1mg/g as a final concentration.
글리시르히진이 L1210 세포를 이식한 생쥐의 Lymphocytes Subpopulation 변화에 미치는 영향

We have previously observed that glycyrrhizin(GL) inhibits the proliferation of transplanted-L1210 cells via the production of nitric oxide from peritoneal macrophages. In the present study, we examined the effect of GL on lymphocytes subpopulation change of L1210 cells-transplanted mice. GL increased the population of CD4-CD8+ cells of thymocytes in L1210 cells-transplanted mice and the lucigenin chemiluminescence of human polymorphonuclear cells. These results suggest that the proliferation of transplanted-L1210 cells is partly inhibited by an enhancement of cytotoxic T cells population and phagocytic activity in GL-administered mice.

These experiments were investigated effects of the cell death of glycyrrhetinic acid (GA) on transplanted-L1210 cells in BALB/c mice. The GA suppressed the proliferation of L1210 cells in vivo and in vitro system. The administration of GA induced apoptosis of transplanted-L1210 cells via the reduction of mitochondrial transmembrane potential in mice. The GA enhanced the production of nitric oxidation in peritoneal macrophages obtained from L1210 cells-transplanted mice. The apoptosis of L1210 cells were induced by co-culture of the macrophages obtained from GA administered mice and L1210 cells in vitro, and was partly inhibited by the treatment of L-NMMA. These results suggest that GA induces the cytotoxicity and the apoptosis of transplanted-L1210 cells via the production of nitric oxide in peritoneal macrophages.

High soy consumption leading to high exposures of soy isoflavones has been associated with a reduced risk of cancers at many sites. As part of a study focusing on the chemopreventive mechanisms, we have investigated the modulating effects of daidzein and genistein, a prominent and more bioavailable isoflavone in soy foods, on murine immune function. Daidzein (50mg/kg) or genistein (50mg/kg) was administered p. o. once a day for 7 days in BALB/c mice. Daidzein decreased the mitogen-stimulated proliferation of murine splenocyte, but genistein increased it. Daidzein stimulated the secretion of interleukin-4, but inhibited the secretion of gamma-interferon, interleukin-2 and tumor necrosis factor-alpha. Genistein stimulated the secretion of gamma-interferon, interleukin-2 and tumor necrosis factor-alpha, but inhibited the secretion of interleukin-4. Daidzein and genistein inhibited the production of nitric oxide and enhanced the phagocytic activity in peritoneal macrophage. These results suggest that cancer preventive effects of daidzein is partly concerned with the secretion of TH2 cells cytokine and the activation of macrophage phagocytosis, and genistein is partly concerned with the secretion of TH1 cells cytokine, tumor necrosis factor-alpha and the activation of macrophage phagocytosis.

We investigated the effect of aqueous extract of Ammomum xanthioides(AXAE) on immediate hypersensitivity. AXAE inhibited systemic anaphylaxis induced by compound 48/80 in mice. AXAE inhibited serum histamine levels induced by compound 48/80 in mice. Moreover, AXAE dose-dependently inhibited histamine release in peritoneal mast cells activated by compound 48/80. These results indicate that the AXAE may be benificial in the regulation of immediate type allergic reaction.
단삼이 마우스 T-lymphocyte 및 Macrophage에 미치는 영향

The purpose of this research was to investigate the regulative action of water extract of Salviae miltiorrhizae Radix(SE) on thymocyte and macrophage in mice. The proliferation in thymocyte was tested using a colorimetric tetrazolium assay(MTT assay), The apoptosis and subpopulation in thymocyte were tested using a flow cytometry. Nitric oxide(No) production from mouse peritoneal macrophage was tested using a Griess reagents and the phagocytic activity was tested using a lucigenin chemiluminescence. SE did not affect the proliferation, apoptosis and subpopulation(CD4^+/CD8^+) in thymocyte in vivo, but enhanced the proliferation in thymocyte in vitro. The n-BuOH fraction and water fraction of SE suppressed the nitric oxide production and results suggest the SE does not affect the activity of thymocyte, but suppressed the activity of macrophage.
